Sugarcane’s (Saccharum spp.) response to Diatraea saccharalis (F.) (Lepidoptera: (Crambidae) herbivory was investigated using a macroarray spotted with 248 sugarcane Expressed Sequence Tags (ESTs) encoding serine peptidase inhibitors, serine peptidases. and Clp protease system subunits. Our results showed that after nine hours of herbivory, 13 sugarcane genes were upregulated and nine were downregulated. Among the upregulated genes, nine were similar to serine peptidase inhibitors and four were similar to Bowman-Birk Inhibitors (BBIs). Phylogenetic analysis revealed that these sequences belong to a phylogenetic group of sugarcane BBIs that are potentially involved in plant defense against insect predation. The remaining four upregulated genes included serine peptidases and one homolog to the Arabidopsis AAA+ chaperone subunit ClpD, which is a member of the Clp protease system. Among the downregulated genes, five were homologous to serine peptidases and four were homologous to Arabidopsis Clp subunits (three homologous to Clp AAA+ chaperones and one to a ClpP-related ClpR subunit). Although the roles of serine peptidase inhibitors in plant defenses against herbivory have been extensively investigated, the roles of plant serine peptidases and the Clp protease system represent a new and underexplored field of study. The up- and downregulated D. saccharalis genes presented in this study may be candidate genes for the further investigation of the sugarcane response to herbivory.  相似文献

The lipase/acyltransferase from Candida parapsilosis is an unusual enzyme that preferably catalyses alcoholysis over hydrolysis in biphasic aqueous/organic media. The aim of this study was to evaluate the operational stability of an immobilised form of this enzyme during the interesterification of fat blends containing n‐3 polyunsaturated fatty acids, in solvent‐free media, at 60 °C, carried out continuously and batchwise. When the interesterification was performed in a continuous fluidised‐bed reactor, an operational half‐life of 9 h was estimated. The biocatalyst was also reused in consecutive 23‐h batches, in a total of four batches, either using fresh medium with no water addition or adding water to rehydrate the biocatalyst. When no water and extra water was added to the reaction medium, the obtained half‐lives were 10 and 18 h, respectively. Thus, the loss of activity may be explained by a progressive dehydration occurring along the reaction rather than by product or substrate inhibition effects. The interesterification activity was accompanied by changes in the acylglycerol profile. An increase in compounds of low equivalent carbon number (ECN) and in triacylglycerols (TAG) of ECN 42 and 44 was observed. This increase was accompanied by the consumption of TAG of ECN 46, 48 and 50.  相似文献

Hybrid electrospun nanofibers of polycaprolactone (PCL)/gelatin are considered as drug-delivery systems for increasing the treatment efficacy in superficial (skin) wounds. Continuous delivery of therapeutic agents, skin extracellular matrix similarity, management of wound exudate, and antimicrobial barrier effect are the major advantages of electrospun nanofibers in skin applications. Additionally, combining the favorable properties of PCL and gelatin, regarding their biocompatibility, biodegradability and mechanical performance have been revealed promising parameters to be considered for blend in hybrid structures. However, the usual optimization protocol of nanofibers’ production in electrospinning is based on the observation of one-variable-at-time being this methodology expensive and time-consuming. Therefore, in this research work, a statistical model based on four input variables namely, the flow rate, the needle-working distance, the applied voltage, and the ratio of PCL in the solution, is developed to predict the behavior of nanofibers. The performance of nanofibers is monitored by measurements of fiber's diameter, mesh's thickness, and mesh's permeability. Overall, the model showed to be statistically significant (p-value < 0.05) and an independent analysis validated the predicted response for optimal condition. Finally, a delivery study is performed to evaluate the electrospun mesh performance as a drug carrier.  相似文献

Monodisperse FePt nanoparticles with size of 4.5 and 6.0 nm were prepared by simultaneous reduction of platinum acetylacetonate and thermal decomposition of iron pentacarbonyl in benzylether. The crystallography structure, size, and composition of the FePt nanoparticles were examined by X-ray diffraction and transmission electron microscopy. Energy dispersive X-ray spectrometry measurements of individual particles indicate a broad compositional distribution in both the 4.5 and 6 nm FePt nanoparticles. The effects of compositional distribution on the phase-transition and magnetic properties of the FePt nanoparticles were investigated.  相似文献

The present work deals with the interdependences between the strain rate and the strain hardening on stainless austenitic steels. Uniaxial tension tests were conducted on a 304 stainless steel at different strain rates in order to analyse the influence of this parameter on the strain hardening and on the material formability. For the strain rates levels analysed (10‐4 to 10‐1 s‐1) it was also observed that increasing the strain rate from 10‐4 up to 10‐1 s‐1 leads to a 25 % difference in uniform tension elongation revealing the curve‐crossing phenomenon. Namely, strain rates equal or higher than 10‐2 lead to a stagnation of strain hardening after a tensile strain of about 0.2. In order to investigate the results obtained, microstructural and thermal analyses were conducted and numerical simulations were performed. It was observed that the decreasing of formability of the material is essentially due to thermal aspects. In the discussion, the experimental and numerical results are analysed in terms of thermal softening, phase transformation and strain rate sensibility.  相似文献

Design data are assigned in geometric and non-geometric form in order to meet design requirements. These data and information must be encapsulated in a data structure that has significance for design applications in each design process phase. The main goal of this research is to find design data groups that represent each mechanical design phase, which will be called phase's design signature. In addition, current data should be an evolution of the geometric and non-geometric information of the previous design phase. In this paper, the purpose is to identify and model a set of design features that encapsulate the design data and their transformations which occurred during the mechanical design phases. This database must capture the designer's intents that can be modeled and implemented using feature-based model in the conventional CAD systems, object-oriented modeling, and Java classes.  相似文献

The Ras protein and its homolog, Rap1A, have an identical "effector region" (residues 32-40) preceded by Asp30-Glu31 and Glu30-Lys31, respectively. In the complex of the "Ras-like" E30D/K31E mutant Rap1A with the Ras-binding domain (RBD), residues 51-131 of Raf-1, Glu31 in Rap1A forms a tight salt bridge with Lys84 in Raf-1. However, we have recently found that Raf-1 RBD binding of Ras is indeed reduced by the E31K mutation, but is not affected by the E31A mutation. Here, the "Rap1A-like" D30E/E31K mutant of Ras was prepared and shown to bind the Raf-1 RBD less strongly than wild-type Ras, but slightly more tightly than the E31K mutant. The backbone 1H, 13C, and 15N magnetic resonances of the Raf-1 RBD were assigned in complexes with the wild-type and D30E/E31K mutant Ras proteins in the guanosine 5'-O-(beta,gamma-imidotriphosphate)-bound form. The Lys84 residue in the Raf-1 RBD exhibited a large change in chemical shift upon binding wild-type Ras, suggesting that Lys84 interacts with wild-type Ras. The D30E/E31K mutant of Ras caused nearly the same perturbations in Raf-1 chemical shifts, including that of Lys84. We hypothesized that Glu31 in Ras may not be the major salt bridge partner of Lys84 in Raf-1. A molecular dynamics simulation of a model structure of the Raf-1 RBD.Ras.GTP complex suggested that Lys84 in Raf-1 might instead form a tight salt bridge with Asp33 in Ras. Consistent with this, the D33A mutation in Ras greatly reduced its Raf-I RBD binding activity. We conclude that the major salt bridge partner of Lys84 in Raf-1 may be Asp33 in Ras.  相似文献

Dissected tissue pieces of the pituitary pars intermedia from the amphibian Xenopus laevis was directly subjected to matrix-assisted laser desorption/ionization (MALDI) mass analysis. The obtained MALDI peptide profile revealed both previously known and unexpected processing products of the proopiomelanocortin gene. Mass spectrometric peptide sequencing of a few of these neuropeptides was performed by employing MALDI combined with postsource decay (PSD) fragment ion mass analysis. The potential of MALDI-PSD for sequence analysis of peptides directly from unfractionated tissue samples was examined for the first time for the known desacetyl-alpha-MSH-NH2 and the presumed vasotocin neuropeptide. In addition, the sequence of an unknown peptide which was present in the pars intermedia tissue sample at mass 1392.7 u was determined. The MALDI-PSD mass spectrum of precursor ion 1392.7 u contained sufficient structural information to uniquely identify the sequence by searching protein sequence databases. The determined amino acid sequence corresponds to the vasotocin peptide with a C-terminal extension of Gly-Lys-Arg ("vasotocinyl-GKR"), indicating incomplete processing of the vasotocin precursor protein in the pituitary pars intermediate of X. laevis. Both vasotocin and vasotocinyl-GKR are nonlinear peptides containing a disulfide (S-S) bridge between two cysteine residues. Interpretation of the spectra of these two peptides reveals three different forms of characteristic fragment ions of the cysteine side chain: peptide-CH2-SH (regular mass of Cys-containing fragment ions), peptide-CH2-S-SH (regular mass + 32 u) and peptide = CH2 (regular mass -34 u) due to cleavage on either side of the sulfur atoms.  相似文献

STUDY DESIGN: Iliac crest corticocancellous allografts for anterior interbody fusion were harvested from six cadavers. The grafts were cut sequentially from left and right crests and randomly assigned to tricortical or bicortical preparations. Their compression strengths then were determined and compared by matched pair analysis. OBJECTIVES: To quantify the failure strength of the grafts from different iliac locations and determine the optimal type of preparation of the grafts for anterior interbody fusion. SUMMARY OF BACKGROUND DATA: Iliac crest corticocancellous autografts and allografts commonly are used for interbody cervical fusions. However, graft strengths for specific sites have not been determined fully. METHODS: Six paired, fresh frozen, iliac crests were sectioned using a customized miter box into multiple 1-cm-thick grafts 1.5 cm in depth to simulate cervical interbody grafts. The left and right sides of each pair were randomly assigned to tricortical and bicortical preparations. The samples were tested by applying a compressive load to failure using a specialized fixture to simulate vertebral body loading. RESULTS: The grafts closer to the anterosuperior iliac spine had significantly higher failure loads and failure strengths than those closer to the posterosuperior iliac spine. The strengths of the bicortical grafts were 72 +/- 14% of the strengths of the tricortical grafts (P < 0.001). CONCLUSIONS: Anterior iliac crest grafts were stronger in compression, even after removal of one cortical surface, than posterior iliac crest grafts.  相似文献
