Bryophytes are the dominant ground cover vegetation layer in many boreal forests and in some of these forests the net primary production of bryophytes exceeds the overstory. Therefore it is necessary to quantify their spatial coverage and species composition in boreal forests to improve boreal forest carbon budget estimates. We present results from a small exploratory test using airborne lidar and multispectral remote sensing data to estimate the percentage of ground cover for mosses in a boreal black spruce forest in Manitoba, Canada. Multiple linear regression was used to fit models that combined spectral reflectance data from CASI and indices computed from the SLICER canopy height profile. Three models explained 63-79% of the measured variation of feathermoss cover while three models explained 69-92% of the measured variation of sphagnum cover. Root mean square errors ranged from 3-15% when predicting feathermoss, sphagnum, and total moss ground cover. The results from this case study warrant further testing for a wider range of boreal forest types and geographic regions.  相似文献

Previously, we have purified and characterized DNA helicase III from the yeast Saccharomyces cerevisiae [Shimizu, K. and Sugino, A. (1993) J. Biol. Chem. 268, 9578-9584]. Here, we have further characterized DNA helicase III activity. It was found that the combined action of the helicase III, yeast DNA topoisomerase I (yTop I), and yeast RPA protein on a covalently closed, circular DNA generates a highly underwound DNA species that has been called form I* or form U. Furthermore, these underwound structures can be accessed by yeast DNA polymerase I (alpha)-primase to initiate DNA synthesis. These reactions mimic in vivo initiation of chromosomal DNA replication. In order to clone the gene encoding DNA helicase III, a partial amino acid sequence of the purified DNA helicase III polypeptide was determined. Using a mix oligonucleotides synthesized based on the amino acid sequence of the helicase, we cloned the gene encoding the helicase III and found it to be identical to YER176W (HEL1) on chromosome V. The amino acid sequence predicted from the nucleotide sequence of the gene has conserved DNA helicase domains that are highly homologous to those of DNA helicases required for DNA replication. However, complete deletion of the gene from the chromosome did not result in any growth defect, suggesting that the gene product is not required for DNA synthesis or that it is functionally substituted by other helicase(s). Furthermore, the deletion strain does not exhibit sensitivity to any DNA-damaging reagents, although it is hypersensitive to calcofluor white, hygromycin, and papulacandin.  相似文献

Immunoaffinity-purified paired helical filaments (PHFs) from Alzheimer's disease (AD) brain homogenates contain an associated protein kinase activity that is able to induce the phosphorylation of PHF proteins on addition of exogenous MgCl2 and ATP. PHF kinase activity is shown to be present in immunoaffinity-purified PHFs from both sporadic and familial AD, Down's syndrome, and Pick's disease but not from normal brain homogenates. Although initial studies failed to show that the kinase was able to induce the phosphorylation of tau, additional studies presented in this article show that only cyclic AMP-dependent protein kinase-pretreated recombinant tau is a substrate for the PHF kinase activity. Deletional mutagenesis, phosphopeptide mapping, and site-directed mutagenesis have identified the PHF kinase phosphorylation sites as amino acids Thr361 and Ser412 in htau40. In addition, the cyclic AMP-dependent protein kinase phosphorylation sites that direct the PHF kinase have been mapped to amino acids Ser356 and Ser409 in htau40. Additional data demonstrate that these hierarchical phosphorylations in the extreme C terminus of tau allow for the incorporation of recombinant tau into exogenously added AD-derived PHFs, providing evidence that certain unique phosphorylations of tau may play a role in the pathogenesis of neurofibrillary pathology in AD.  相似文献

The stable pools formed by spur dikes can be a significant factor for improving aquatic habitat in unstable degraded streams. Prediction of the volume and geometry of the scour hole associated with a spur dike yields information that is valuable for assessing the potential benefit to the aquatic habitat. Volumes of the scour hole in the vicinity of model spur dikes were measured in a laboratory flume under clear-water overtopping flows. Spur dike length, flow depth, and shear velocity ratio were varied in the experiments and found to significantly influence the volume of the scour hole. For overtopping flows, the ratio of the flow depth to the spur dike height was found to be an important control on the geometry of the resulting scour hole. At higher flow depth to spur dike height ratios, the location of the maximum depth of scour changed from the upstream point of the dike toward the channel bank and caused a secondary scour zone to form downstream of the spur dike. A preliminary technique is proposed to predict the volume of scour for spur dikes perpendicular to the bank.  相似文献

Acute administration of single high doses of cocaine (50 or 60 mg/kg) produces liver injury in mice that have been pretreated with inducers of mixed function oxidases. Multiple low doses of cocaine (10-30 mg/kg) will produce hepatotoxicity without prior induction. To establish whether cocaine can induce its own activation, mice were given three daily injections of cocaine. Total cytochrome P450 content of the liver did not change. After 3 days the amount of cytochrome P450 2B10, as measured by pentoxy resorufin-O-dealkylase activity and immunoblotting, increased 3-fold. Cytochrome P450 2A5-catalyzed coumarin 7-hydroxylase activity and immunoreactive protein increased by about 50%. Enzyme activities and Western blotting of isoforms 1A, 2E, and 3A showed no change during this time. Chronic cocaine increased N-hydroxylation of norcocaine. Immunoinhibition studies showed that cytochrome P450 2A5 was the major isoform responsible for norcocaine N-hydroxylation. These results demonstrate that chronic cocaine can induce its own metabolism. Similar increases were also observed in mice not susceptible to liver injury from chronic cocaine.  相似文献

Stoichiometric mullite, 3Al2O3·2SiO2, has been prepared from a variety of colloidal and solution precursors. In order to change the level of mixing, the size of the silica particles has been varied while keeping the alumina (boehmite) particle size constant. The effect of varying particle size on the crystallization and densification behavior has been studied. Densification behavior was characterized by measuring the bulk density of pellets as a function of heat treatment temperature and by dilatometry. Phase development was examined by thermal analysis and X-ray diffraction. Results showed that the formation of crystalline phases including mullite inhibited densification. The formation of mullite was controlled by the initial segregation of alumina and silica in the gel. A heat treatment time-temperature profile designed to optimize the densification of the colloidal powders was investigated in order to compare the ability of the different gels to densify.  相似文献
