Pulmonary Langerhans cell granulomatosis (LCG), also called histiocytosis X, is a disorder of unknown etiology characterized by the presence of destructive granulomas containing numerous Langerhans cells (LCs). The process may be localized or multifocal, and it remains unclear whether the same pathogenic mechanism is involved in all forms of the disease. It is often assumed that the massive accumulation of LCs at the sites of the lesions results from the abnormal proliferation of these cells, although it has been suggested that LCG in adults, at least in the lung, could be a reactive disorder initiated by activated LCs. Little is known, however, concerning the mechanisms responsible for the accumulation of large numbers of LCs in the course of the disease, and the relative contribution of recruitment and local proliferation of these cells remains to be established. To investigate this question, the proportion of replicating LCs was evaluated in biopsied granulomas from patients with localized or diffuse form of LCG by means of several histopathological techniques currently used in assessment of cell proliferation. The findings demonstrate that, except for proliferating cell nuclear antigen (PCNA), all parameters measured are low in all forms of the disease. They are similar to those of renewing epithelial cells and clearly less than those of neoplastic cells. These data strongly suggest that LCs in LCG granulomas are not a rapidly dividing cell population and that local LC replication makes only a minimal contribution to granuloma maintenance. Optical pick‐up lenses are used in optical storage devices such as CD, DVD, and Blu‐Ray Disc. The production of such lenses is based on UV‐induced polymerization of a mixture of a dimethacrylate monomer and an initiator on a spherical glass substrate. The shape of the polymer layer is defined with an aspherical transparent mold. This means that the coating is completely surrounded by glassy materials during processing. Raman spectroscopy is applied in situ to monitor the polymerization reaction under conditions that closely resemble the actual production process. As a result improvements can be made to the reaction conditions if necessary. Data are compared to results obtained with IR spectroscopy in an off‐line approach. The value of the in situ characterization using Raman spectroscopy is illustrated by the observation that contrary to expectation, the local rate of polymerization is not influenced by shrinkage effects caused by local variations in volume relaxation in the wedge‐shaped sample volume. Instead, even quartz glass mold plates with a thickness of 30 times that of liquid monomer were deformed to accommodate for thermodynamically required volume shrinkage. BACKGROUND: Citrate, used for the anticoagulation of the extracorporeal dialysis circuit, reduces ionized calcium by chelation and has been claimed to attenuate dialyser membrane bioincompatibility. Dialysis with complement-activating cuprophane membranes is associated with leukopenia which has been related to an increase in adhesion molecule expression on the surface of circulating leukocytes. METHODS: The effect of citrate anticoagulation on the expression of CD11b, CD11c and CD45 on the surface of granulocytes and CD14 on monocytes during haemodialysis with cuprophane membranes, was evaluated by flow cytometric analysis. A comparison of standard heparin vs citrate was performed in 14 chronic haemodialysis patients. During citrate anticoagulation a calcium-free dialysate was used and citrate was infused to obtain a concentration of 4.3 mmol/l blood. The unchallenged 'baseline state' expression of the surface molecules and the increase after ex vivo stimulation with phorbol 12-myristate 13-acetate (delta-PMA) or formyl-methionyl-leucyl-phenylalanine (delta-fMLP) was studied. RESULTS: With heparin, as well as with citrate, a sharp fall in granulocyte and monocyte count was observed after 15 min of dialysis, followed by a recovery at the end of the session. The expression of CD11b, CD11c and CD45 on granulocytes increased markedly during cuprophane dialysis with a peak at 15 min; there were no differences in response between heparin and citrate anticoagulation. Delta-PMA and delta-fMLP for CD45, CD11c and CD14 showed a decrease during cuprophane dialysis vs t0; again there were no differences between heparin and citrate. CONCLUSION: We conclude that the use of citrate was not associated with reduced leukocyte activation as measured by the expression of surface molecules during cuprophane dialysis and that no effect on dialysis leukocytopenia could be registered. 相似文献

Water vapor isotherms were examined and 1 H-magic-angle spinning nuclear magnetic resonance (1 H-MAS NMR) analysis was conducted to determine the amount of water vapor adsorbed and desorbed on hydrothermally solidified materials derived from clay minerals. The amount of water vapor adsorbed and desorbed decreased with increasing mesopore volume of the materials, which disagreed with the estimation obtained using the Kelvin equation. Water vapor was physisorbed on alumina-like sites in the materials. An increase in the binding energy between water vapor and the materials led to an improvement in the water vapor adsorption–desorption properties of materials. 相似文献

Although related to inflammatory markers in adults, little is known about the association between cigarette smoking and C-reactive protein (CRP) in adolescent smokers. We examined the association between high-sensitivity CRP (hs-CRP) concentrations and smoking in youth. We used data from a cross-sectional, province-wide survey of a representative sample of youth conducted in Quebec, Canada, in 1999. Data were collected in self-report questionnaires completed by participants and their parents. Participants provided a fasting blood sample, and anthropometric measures were undertaken by trained technicians. The present analysis pertains to 1,501 adolescents aged 13 and 16 years who completed questionnaires and for whom blood samples were available. The independent association between a six-category indicator of smoking status and elevated hs-CRP, defined as a value at least in the 90th percentile of the age- and sex-specific CRP distribution, was assessed in multiple logistic regression analyses controlling for potential confounders. Relative to never-smokers, the odds ratios (95% confidence intervals) for puffers (i.e., never smoked a whole cigarette), those who smoked but not in the past month, light past-month smokers, moderate past-month smokers, and heavy past-month smokers were 1.04 (0.55-1.98), 1.76 (1.06-2.94), 1.39 (0.70-2.76), 2.07 (0.96-4.42), and 2.40 (1.18-4.88), respectively. Our data suggest a positive association between smoking status and elevated CRP in adolescents, and in particular among heavier past-month smokers. Damage related to cigarette smoking may begin soon after tobacco use initiation, reinforcing the preventive message that no level of smoking is safe in youth. 相似文献

Experiments were performed to evaluate disinfection by-products in model humic acid solutions which were ozonated at three different ozone to carbon levels and then chlorinated. These experiments were conducted in order to help understand whether the ozone/post-chlorination process alters the amount and type of mutagenic by-products formed, from those produced by chlorination of humic acid alone. Disinfection by-products were identified by gas chromatography/mass spectrometry (GC/MS). Samples of clarified and sand-filtered Mississippi River water at a pilotscale drinking water treatment plant in Jefferson Parish, Louisiana, that were ozonated and post-disinfected with chlorine, also were analyzed by GC/MS. A comparison of the by-products in the pilot plant study versus those in our laboratory study showed that similar compounds were produced. The effect of bromide ion in the pilot plant water on by-product formation also is discussed. 相似文献
