Osteocytes are terminally differentiated osteoblasts embedded within the bone matrix and key orchestrators of bone metabolism. However, they are generally not characterized by conventional bone histomorphometry because of their location and the limited resolution of light microscopy. OI is characterized by disturbed bone homeostasis, matrix abnormalities and elevated bone matrix mineralization density. To gain further insights into osteocyte characteristics and bone metabolism in OI, we evaluated 2D osteocyte lacunae sections (OLS) based on quantitative backscattered electron imaging in transiliac bone biopsy samples from children with OI type I (n = 19) and age-matched controls (n = 24). The OLS characteristics were related to previously obtained, re-visited histomorphometric parameters. Moreover, we present pediatric bone mineralization density distribution reference data in OI type I (n = 19) and controls (n = 50) obtained with a field emission scanning electron microscope. Compared to controls, OI has highly increased OLS density in cortical and trabecular bone (+50.66%, +61.73%; both p < 0.001), whereas OLS area is slightly decreased in trabecular bone (−10.28%; p = 0.015). Correlation analyses show a low to moderate, positive association of OLS density with surface-based bone formation parameters and negative association with indices of osteoblast function. In conclusion, hyperosteocytosis of the hypermineralized OI bone matrix associates with abnormal bone cell metabolism and might further impact the mechanical competence of the bone tissue.  相似文献

Aluminium films implanted with 30 keV lithium ions of doses from 2.2 × 1016 to 1 × 1017 ions cm?2 were studied by transmission electron microscopy. Samples implanted with 6 × 1016 and 1 × 1017 ions cm?2 showed the presence of the Al-Li phase, while no change was observed in specimens implanted with 2.2 × 1016 ions cm?. Microdiffraction patterns obtained from different areas of an aluminium film implanted with 2.0 × 1017 Li+ ions cm?2 at 35 keV, revealed the presence of a stable Al-Li phase along with a metastable phase. Furthermore, on annealing the implanted samples at 200°C for 20 min an increase in ‘d’ values was noticed when compared with non-annealed specimens. However the annealed samples showed a very small lattice mismatch between the metastable phase and the Al matrix, thus indicating the presence of a metastable Al3Li phase, responsible for hardening in binary Al-Li alloy.  相似文献

In the context of optical communications, we study experimentally the behavior of a pump-modulated fiber laser at frequencies close to the relaxation oscillation frequency. We show that correlations can appear between successive “1” bits in various situations. This “memory effect” can be interpreted using a simple deterministic model, and we show that this effect is related to the behavior of the population inversion, which does not follow adiabatically the photon number. Our results also suggest the existence of an optimum pump bias value for optical communications, slightly below threshold  相似文献

End-stage renal disease is associated with a defect in immunologic functions. Previous studies have demonstrated that uremic ultrafiltrate (UUF) contains factors that suppress calcitriol synthesis and its biological actions. In the present study, the effect of UUF on basal and calcitriol-induced membrane bound CD14 expression of monocytes activated by phorbol 12-myristate 13-acetate was evaluated. CD14 acts as a receptor for the complexes of lipopolysaccharide and lipopolysaccharide-binding protein. Monocytes isolated from normal donors were used for the assay of monocyte CD14 expression. A calcitriol induced rise in monocyte CD14 expression (1966+/-423 to 2421+/-436 fluorescence intensity) was found. However, UUF not only suppressed basal CD14 expression of monocytes (from 1966+/-423 to 1240+/-203, P < 0.05) but also significantly blunted calcitriol-induced CD14 expression (from 2421+/-436 to 1744+/-229, P < 0.05). HPLC fractionated UUF collected from 8 to 16 min (fraction 1, F1) and from 25 to 40 min (fraction 3, F3) also significantly suppressed the expression of CD14. Because purine derivatives coeluted within F1, their effect on monocyte CD14 expression was also tested. Uric acid, xanthine, and hypoxanthine was found to suppress basal as well as calcitriol-induced CD14 expression of monocytes in a dose-dependent manner. In conclusion, UUF contains factors that impair calcitriol activated function of monocytes.  相似文献

Lipid binding properties of cytochrome c (cyt c) were investigated by using semisynthetic mutant protein having amino acid substitution on the evolutionary invariant residue Arg91. We demonstrate here that the membrane binding properties of cyt c are dramatically altered by substituting norleucine for the invariant Arg91. More specifically, while the binding of this mutant to liposomes per se is indistinguishable from the wild type protein, it completely lacks the ability of the native cyt c to rupture liposome bilayers.  相似文献

A digital computer program has been developed for analysis of the nystagmus which results from sinusoidal head rotation. For any sinusoidal vestibular stimulation between 0.05 Hz and 1 Hz, the program, accepts sampled records of eye position and head velocity and yields cumulative eye position as well the phase and the gain of the vestibuloocular reflex. Described in this paper are new algorithms by which fast phases are detected and by which slow phase cumulative eye position is reconstructed using and iterative procedure of best least squares fit. These algorithms are useful in neurophysiological studies on normal animals, but not in clinical analysis of nystagmus.  相似文献

The aim of this study was to evaluate the bacterial spreading by using ultrasonic scaler and air-polisher. During ultrasonic scaling, bacterial spray is important near patient's mouth (30 cm), principally without good aspiration devices. In this case, clear spreading is observed at 2 m from the patient. Air-polishing compared to ultrasonic scaling produces larger microorganisms pollution.  相似文献

Both tumor necrosis factor-α (TNF-α) and EFA deficiency (EFAD) have been established as causes of marked perturbations in lipid and lipoprotein metabolism. Excessive levels of circulating TNF-α can coexist with EFAD in various clinical disorders such as cystic fibrosis and type I diabetes. The present study therefore aimed to investigate their combined effects on lipid profile and lipoprotein composition by administering TNF-α to EFAD rats. Lipoprotein lipase (LPL), the ratelimiting enzyme in TG catabolism, was also measured in epididymal adipose tissue. EFAD, after a 4-wk period, induced significant increases in plasma TG (80%, P<0.001), total cholesterol (TC, 27%, P<0.025), and HDL-cholesterol (HDL-C, 62%). Two hours after the administration of TNF-α, a further rise in TG (43%, P<0.05) was noted in controls, but not EFAD animals. TC and HDL-C were unaffected by TNF-α treatment. In addition, TNF-α modified lipoprotein-lipid composition. VLDL and HDL2 derived from EFAD rats were depleted in apolipoprotein (apo) E and apo A-II, and enriched in apo A-12 h after TNF-α administration. Finally, TNF-α decreased adipose tissue LPL activity in both control and EFAD animals. The TNF-α-induced inhibition was more marked in EFAD rats. The present results demonstrated that TNF-α can amplify or antagonize the effects of EFAD on lipid profile, lipoprotein composition, and LPL activity. These data also suggest that the host's nutritional status is a determining factor for the modulating effect of TNF-α on lipid metabolism.  相似文献

The preparation of YBa2Cu3O7 - δ-based ceramics was made from a mixture of oxides taken in the molar proportions α:1:2 BaCuO2:Y2BaCuO5:CuO(0.95 α 3). The densification of the ceramics is strongly dependent on the initial amount of BaCuO2. The highest density is obtained when α= 1. All the ceramics present a superconducting transition. A sintering mechanism is proposed in which the densification is mainly governed by the appearance of a metastable BaCuO2-based liquid at around 900°C.  相似文献

To investigate the capability of the lead isotope signature technique to support a source apportionment study at a Continental scale, atmospheric particulate matter was collected at Cap Gris-Nez (Eastern Channel, northern France), over one year (1995-1996). Four days retrospective trajectories of air masses were available during each sampling experiment. Twenty-eight samples, for which the origin of aerosols was unambiguously determined, were selected for isotopic measurements. Considering the Enrichment Factors, EF(Crust) of lead and its size distribution, we show that lead is mostly from anthropogenic origin and mainly associated with [0.4 < diameter < 0.9 microm] particles. The extent to which various Continental sources influence the lead abundance in aerosols is exhibited by considering both the lead concentration and the origin of air masses. Lead concentration is higher by a factor of approximately seven, when air masses are derived from Continental Europe, by comparison with marine air masses. Taking into account these concentrations and the vertical movements of air masses, we compare the different isotopic compositions using a statistical non-parametric test (Kolmogorov-Smirnov). We produce evidence that, for most of the cases, air masses originating from Continental Europe exhibit a more radiogenic composition (1.134 < 206Pb/207Pb < 1.172) than air masses coming from the United Kingdom (1.106 < 206Pb/207Pb < 1.124). Generally, lead isotopic compositions in aerosols are clearly distinct from the gasoline signatures in European countries, strongly suggesting that automotive lead is no longer the major component of this metal in the air. Gasoline and industrial isotopic signatures could explain the origin of lead in our aerosol samples. A source apportionment based upon 206Pb/207Pb ratios, suggests that the difference between British (206Pb/207Pb = 1.122 +/- 0.038) and Continental (206Pb/207Pb = 1.155 +/- 0.022) signatures may be largely explained by differences in the petrol lead content of aerosols (23-62% in Great Britain vs. 10-36% in Continental Europe).  相似文献
