The charge transport through ferrocene 1,1′‐diamine (FDA) molecules between gold electrodes is investigated using the mechanically controllable break junction technique combined with a theoretical framework of density functional theory simulations to understand the physics of these molecular junctions. The characteristic conductances of the molecule are measured at low bias as well as current–voltage (IV) characteristics. By fitting the IV characteristics to the single‐level model, the values for the position of the molecular level, mainly responsible for the transport, and its coupling to the leads, are obtained. The influence of the binding sites, molecular conformation, and electrode distance are systematically studied from a theoretical perspective. While a strong dependence of conductance on the adsorption geometry is found, the decrease of conductance as a function of electrode distance arises mainly from a decrease of coupling strength of the molecular electronic orbitals through a reduced overlap and, to a lesser extent, from a shift of their alignment with respect to the Fermi energy. 相似文献

Photoquadrats and underwater video surveys are standard non-destructive monitoring methods in marine ecosystems and are becoming more common in freshwater systems. The high water clarity found in most of the Laurentian Great Lakes make them ideally suited for photoquadrat sampling. We compared the effectiveness of baited photoquadrats to monitor benthic fish communities of shallow, littoral habitats of northern Lake Michigan. We compared our results with baited minnow traps, a technique commonly used to monitor benthic fish communities in freshwater ecosystems. Photoquadrats baited with lake trout (Salvelinus namaycush) eggs, the most effective attractant, proved to be an efficient tool for sampling round goby (Neogobius melanostomus) relative abundance and resulted in higher round goby catches than in photoquadrats baited with a commercially available fish attractant or unbaited photoqudrats. This method allowed sites to be surveyed rapidly (requiring <1?h per site), and replicate samples produced data with low variability. In contrast, baited minnow traps produced highly variable catch per unit effort (CPUE) irrespective of soak time. Photoquadrat methods appear to be an improvement over traditional sampling with minnow traps for round goby and may also be better for other nearshore benthic fishes. 相似文献

In recent decades Saccharomyces cerevisiae has proven to be one of the most valuable model organisms of aging research. Pathways such as autophagy or the effect of substances like resveratrol and spermidine that prolong the replicative as well as chronological lifespan of cells were described for the first time in S. cerevisiae. In this study we describe the establishment of an aging reporter that allows a reliable and relative quick screening of substances and genes that have an impact on the replicative lifespan. A cDNA library of the flatworm Dugesia tigrina that can be immortalized by beheading was screened using this aging reporter. Of all the flatworm genes, only one could be identified that significantly increased the replicative lifespan of S.cerevisiae. This gene is the cysteine protease cathepsin L that was sequenced for the first time in this study. We were able to show that this protease has the capability to degrade such proteins as the yeast Sup35 protein or the human α‐synuclein protein in yeast cells that are both capable of forming cytosolic toxic aggregates. The degradation of these proteins by cathepsin L prevents the formation of these unfolded protein aggregates and this seems to be responsible for the increase in replicative lifespan. 相似文献

Verifying who someone is on the Internet is a prerequisite for online services that process sensitive or valuable information. While this has been solved with national or sectorial electronic identification (eID) schemes, general, cross-border solutions are rare. Cross-border eID difficulties have several origins: (i) incompatible national eID models; (ii) different legislations with incompatible objectives; (iii) lack of common language and semantics; (iv) different common procedures, specially in what concerns mandates and delegation; (v) different implementations of the same eID models. These have been addressed by STORK, a project that developed a federated cross-border eID system that was piloted in about twenty European Union Member States in service sectors as sensitive as eBanking and eHealth. STORK designed and implemented a large-scale interoperability framework, allowing different systems of different models to coexist, using a common language with a common semantics and satisfying national privacy legislations. The experience gained from this large-scale pilot fed into EU policy-making, in particular, the recently enacted eIDAS Regulation requiring mutual recognition of eID by 2018 has been directly influenced by STORK and its lessons learned. 相似文献

Start-up of two acidogenic reactors under mesophilic (37 degrees C) and thermophilic (55 degrees C) conditions was carried out with methanogenic granular sludge as an inoculum and dairy wastewater as feed. During these 71 days of the start-up period, microbial community dynamics in these two acidogenic reactors, as monitored by denaturing gradient gel electrophoresis (DGGE) and dot-blot hybridization with group-specific oligonucleotide probes, was correlated to reactor performance. Due to pH drop to 5.5, DGGE community fingerprints for domains Bacteria and Archaea populations showed significant shifts after 13 days of operation, and this change was accompanied with an increase in volatile fatty acid production, a decrease in methane formation, and rapid sludge disintegration. Dot-blot hybridization results further indicated that the decrease in methane production was related to the decrease in Archaea population in particular with methanogens from 34.1% of total 16S-rRNA in the seed sludge to 8% within the first 13 days, and to 2-5% at day 71. Among the methanogens monitored, the class Methanomicrobiales was the most abundant followed up by Methanobacteriales and Methanococcales. Due to an elevated temperature, the microbial community change was more significant and rapid in the thermophilic reactor than in the mesophilic reactor. Significant microbial population changes took place at the first 13 days for both reactors, but a longer period up to 71 days was required to establish a microbial community with a stable metabolic activity. 相似文献
