Lipid droplets (LDs) are intracellular organelles that are ubiquitous in many types of cells. The LD core consists of triacylglycerols (TGs) surrounded by a phospholipid monolayer and surface proteins such as perilipin 2 (PLIN2). Although TGs accumulate in the phospholipid bilayer of the endoplasmic reticulum (ER) and subsequently nascent LDs buds from ER, the mechanism by which LD proteins are transported to LD particles is not fully understood. Sar1 is a GTPase known as a regulator of coat protein complex Ⅱ (COPⅡ) vesicle budding, and its role in LD formation was investigated in this study. HuH7 human hepatoma cells were infected with adenoviral particles containing genes coding GFP fused with wild-type Sar1 (Sar1 WT) or a GTPase mutant form (Sar1 H79G). When HuH7 cells were treated with oleic acid, Sar1 WT formed a ring-like structure around the LDs. The transient expression of Sar1 did not significantly alter the levels of TG and PLIN2 in the cells. However, the localization of PLIN2 to the LDs decreased in the cells expressing Sar1 H79G. Furthermore, the effects of Sar1 on PLIN2 localization to the LDs were verified by the suppression of endogenous Sar1 using the short hairpin RNA technique. In conclusion, it was found that Sar1 has some roles in the intracellular distribution of PLIN2 to LDs in liver cells.  相似文献

It is well known that the properties of hematopoietic stem/progenitor cells (HSCs), such as their self-renewal ability and multipotency, are maintained through interactions with mesenchymal stem/stromal cells (MSCs). MSCs are rare cells that are present in the bone marrow and are useful for clinical applications due to their functional ability. To obtain the necessary number of cells, MSCs must be cultured to expand, but this causes a remarkable decrease in stem cell properties, such as multipotency and proliferation ability. In this study, we show that the c-Mpl signal, which is related to the maintenance of hematopoietic stem cells, has an important effect on the proliferation and differentiation ability of MSCs. Utilizing a co-culture system comprising MSCs and HSCs, it is suggested that signaling from hematopoietic cells to MSCs supports cell proliferation. Interestingly, the enhanced proliferation ability of the HSCs was decreased in c-Mpl knock-out HSCs (c-Mpl-KO). In addition, the MSCs co-cultured with c-Mpl-KO HSCs had reduced MSC marker expression (PDGFRa and Sca-1) compared to the MSCs co-cultured with c-Mpl-wild-type HSCs. These results suggest that a hematopoietic–mesenchymal signal exists, and that the state of the HSCs is important for the stability of MSC properties.  相似文献

Sample disturbance caused by difference in sampling tube geometry was evaluated by two nondestructive methods: the measurement of the residual effective stress (pr') by ceramic disc; and the use of the bender element to ascertain the shear wave velocity (Vs), and thus the maximum shear modulus (GBE). Samples were measured under atmosphere, i.e., not under confined stress conditions. The soil samples were obtained from two sources: reconstituted Kasaoka clay prepared in the laboratory, and at the test site at Takuhoku, Hokkaido, Japan. Samplers with different geometrical designs, referring to the Japanese standard stationary piston sampler, were used for the model ground and field sampling. The geometrical effects of the sampling tube, for example, the thickness of the tube wall, the edge angle, and the existence of a piston were carefully examined. The quality of the samples taken with different samplers was evaluated by pr' and GBE, values which were normalized by the in situ vertical effective stress (σ'vo) and Gf measured by the seismic cone test in the field. It was found from these studies that pr'/σ'vo and GBE/Gf vary considerably due to the geometry of the sampler, with the edge angle of sampling tubes being the most important feature in obtaining high quality samples. The wall thickness, and thus, the area ratio of the sampler is not critical to the sample quality if the edge angle is sharp enough. The existence of the piston does not significantly influence the sample quality in field samples. Furthermore, the correlation between GBE and pr' was also investigated, and it was found that the two parameters are strongly dependent.  相似文献

In evaluating the uncertainty of the standard measuring system for lightning‐impulse high voltages, which is composed of a standard voltage divider, a digital recorder, and calibrators, step‐response tests of the standard voltage divider may be useful. In this paper, a convolution algorithm is employed to calculate the output impulse voltage waveforms from measured step‐response waveforms. The uncertainties of peak‐value measurement due to the influence of the nominal epoch, uncertainty of the peak‐value measurement due to dispersion of the AC scale factor, and uncertainty of the virtual front‐time measurement due to long‐term stability are evaluated. Furthermore, the error of the virtual front time of the output waveforms is discussed. The front part of the step‐response waveform, tT30%, does not influence the error of the virtual front time. Therefore, for the standard voltage divider, the step‐response parameters, that is, the experimental response time, partial response time, settling time, and overshoot, have almost nothing to do with the error of the virtual front time. © 2012 Wiley Periodicals, Inc. In order to realize a noninvasive blood glucose monitor, we monitored the gingival crevicular fluid (GCF). In this paper, the clinical evaluations were performed on not only normal subjects but also diabetic subjects using a GCF-glucose monitor to determine blood glucose levels. Meal load tests were carried out and the time-course changes in blood glucose level and GCF glucose level were measured continuously. A positive correlation of more than 0.9 was found between blood glucose level and GCF glucose level, necessitating the calibration of individual correlations for every subject. Finally, the performance of the GCF-glucose monitor was evaluated using Error Grid Analysis. As the results, significant information for the glucose level decision was obtained not only for normal subjects, but also for diabetic subjects.  相似文献

This paper presents an information-theoretic analysis of neural spike trains in an auditory nerve fiber (ANF) model stimulated extracellularly with Gaussian or sinusoidal waveforms in the presence of a pseudospontaneous activity of spike firings. In the computer simulation, stimulus current waveforms were applied repeatedly to a stimulating electrode located 1 mm above the 26th node of Ranvier, in an ANF axon model having 50 nodes of Ranvier, each consisting of stochastic sodium and potassium channels. From spike firing times recorded at the 36th node of Ranvier, a post-stimulus time histogram (PSTH) was generated, and raster plots were depicted for 30 stimulus presentations, in order to investigate the temporal precision and reliability of the spike firing times. Also, inter spike intervals were generated and then "total" and "noise" entropies were estimated to obtain the mutual information and the information rate of the spike trains. It was shown in the case of Gaussian electric stimuli that the temporal precision of spike firing times and the reliability of spike firings were found to increase as the standard deviation (SD) of the Gaussian electric stimuli increased. It was also shown in the case of sinusoidal electric stimuli where there was a specific amplitude of sinusoidal waveforms, the information rate being maximized. It was implied that setting the parameters of electric stimuli to the specific values which maximize the information rate might contribute to efficiently encoding information into the spike trains in the presence of a pseudospontaneous activity of spike firings.  相似文献

Energy-filtering transmission electron microscopy (EFTEM) was applied for investigating interfaces between a polymer and an adhesive. The sample employed in this work is polybutylene terephtharate (PBT) sheets laminated with an epoxy adhesive. It was found that heat aging of the PBT at 180 degrees C in air for > 9 h prior to adhesion decreases the adhesion strength drastically. To investigate this unfavourable aging effect on the adhesion strength, we performed elemental mapping and image EELS using EFTEM. A weak boundary layer with a thickness of < 50 nm was visualized at the PBT-adhesive interface by elemental mapping in the sample subjected to the heat aging and image EELS revealed the origin of this layer. Thus, we clearly correlated the nanoscale interfacial structure with the adhesion strength by EFTEM.  相似文献

Results from double directional ultrawideband (UWB) channel sounding in a wooden house are described. The double directional channel sounder estimates directional information at both ends of the link, so that we can separate antenna directivity from the channel sounding results. We investigated the dominant propagation mechanisms by introducing cluster analyses. The detected propagation paths from the channel sounding were first classified into clusters in the angular-delay domain, and then properties of the clusters such as standard deviation of path positions, dynamic range of path power, and power distribution of clusters were derived. From the results, we discussed the similarities and differences between the measurement environment and the physical propagation phenomena. Finally, different types of scattering losses of the propagation paths were derived and modeled. The results from sounding and analysis contribute in the development of UWB propagation models and can be used in UWB propagation simulations  相似文献

Twisted nematic liquid crystal displays (TN-LCDs), doped with the nanoparticles of metal, such as Pd, Ag, or Ag-Pd, which are protected with ligand molecules, such as nematic liquid crystal, exhibit a frequency modulation (FM) electro-optical (EO) response with short response time of milliseconds (ms) or sub-ms order together with the ordinary rms voltage response. These devices are called FM/AM-TN-LCDs; they are distinct from the ordinary LCDs featured by the amplitude modulation (AM) response. The phenomena of the FM/AM LCDs may be attributed to the dielectric dispersion of a heterogeneous dielectric medium known as the Maxwell-Wagner effect. It is experimentally shown that the frequency range spreads from several tens hertz to several tens kilohertz and the spectrum is more or less centered about the dielectric relaxation frequency. We formulated a theory based on an equivalent circuit model to evaluate the dielectric relaxation frequency and the dielectric strengths; and we succeeded in explaining the dependence of the dielectric relaxation frequency on the concentration of nanoparticles and the their dielectric and electrical properties, whereas conventional theories based on electromagnetic theory are unable to explain this concentration dependence. This paper reports on the experimental results of the EO effects and the dielectric spectroscopy including the dielectric relaxation times and the dielectric strengths of nematic liquid crystal, 5CB (4-pentyl-4'-cyanobiphenyl), doped with the metal nanoparticles of I'd alone and Ag-Pd composite; and discusses how the observed dielectric relaxation frequency or dielectric relaxation time depend on the concentration of the doped nanoparticles and also their electrical and dielectric properties.  相似文献

780-nm-band high-power and 650-nm-band laser diodes (LDs) with real refractive index guided self-aligned (RISA) structures are monolithically integrated for the first time. High-power and fundamental transverse mode operation at an output power of 100-mW continuous wave (CW) up to 80°C is attained for the 780-nm-band LD.
