Previously, we showed that the N-terminal recognition domain (T1) of Kv1.3 was not required for assembly of functional channels [Tu et al. (1996) J. Biol. Chem. 271, 18904-18911]. Moreover, specific Kv1.3 peptide fragments including regions of the central core are able to inhibit expression of current produced from a channel lacking the T1 domain, Kv1.3(T1-). To elucidate the mechanism whereby Kv1.3 peptide fragments suppress Kv1.3(T1-) current, we have studied the ability of peptide fragments containing the transmembrane segments S1, S1-S2, or S1-S2-S3 to physically associate with the Kv1.3(T1-) polypeptide subunit in vitro in microsomal membranes. Using c-myc (9E10) epitope-labeled peptide fragments and anti-myc antibody as well as antisera to the Kv1.3 C-terminus, we now demonstrate specific association of these peptide fragments with Kv1.3(T1-). Association of peptide fragments with Kv1.3(T1-) was correlated with integration of both proteins into the membrane. Furthermore, the relative strength and kinetics of this association directly correlated with the ability of fragments to suppress Kv1.3(T1-) current. The rate-limiting step in the sequential synthesis, integration, and formation of a complex was the association of integrated polypeptides within the plane of the lipid bilayer. These results strongly suggest that the physical association of transmembrane segments provides the basis for suppression of K+ channel function by K+ channel peptide fragments in vivo. Moreover, the S1-S2-S3 peptide fragment potently suppressed full-length Kv1.3, thus implicating a role for the S1-S2-S3 region of Kv1.3 in the assembly of the Kv1.3 channel. Microfibre reinforced cement composites reinforced with high volume fractions of carbon, steel and polypropylene fibres were tested in uniaxial tension. Composites investigated included those with only one type of fibre (mono-fibre composites) and those with two or more types of fibres (hybrid-fibre composites) in the same mix. Considerable strengthening, toughening and stiffening of the host matrix due to microfibre reinforcement were observed. In the hybrid-fibre composites, different fibres appear to act as additive phases; i.e., they maintain their individual reinforcing capabilities. The composites were also impact tested in uniaxial tension using a newly designed instrumented impact machine. When compared with static test results, considerable sensitivity to stress rate was noted; composites were found to be stronger and tougher under impact and the improvements were more pronounced at higher fibre volume fractions. The potential of these composites for use in thin sheet products and other similar applications is recognized, and the need for continued research is stressed.  相似文献

准噶尔盆地腹部普遍发育深层(大于4000m)超压系统,很多钻井都钻遇超压。通过对超压段钻井、地质、测井、地震特征系统研究发现,超压段泥浆密度大幅增加,具有高声波时差、低电阻率以及地震低速的超压响应特征。利用Dix公式计算地震层速度和利用Fillippone公式计算地层压力,得到超压数据体。准噶尔盆地腹部超压在白垩系及其以下沉积地层中均有分布,超压层的深度范围不同。分1井西凹陷边缘以及昌吉凹陷西南有空层现象。超压顶面分布的层位由北往南、从东到西逐渐变浅,从三叠系顶部到白垩系底部。深部超压对油气分布和富集有着重要的控制作用,准噶尔盆地腹部超压层分布的研究将对本区油气勘探起积极的推动作用。  相似文献

多数ITO透明导电玻璃生产线在实现SiO2膜与ITO膜在线联镀时,应用SiO2靶射频溅射沉积SiO2膜工艺和ITO靶直流溅射沉积ITO膜工艺,如果SiO2膜应用硅靶反应磁控溅射工艺,存在这种工艺是否可以与ITO靶直流溅射沉积ITO膜工艺在线联用以及如何实现联用的问题。作者对现有的生产线进行了改造设计、加工,做了大量实验、质谱分析和多项测试研究,成功地实现反应溅射SiO2膜与ITO膜在线联镀,做到SiO2镀膜室的工作状态的变化基本上不影响ITO镀膜室的工艺条件。  相似文献

为了开发新型的生体材料,本文对自蔓延高温合成TiNi多孔体合金——人造骨材料进行了研究。以单质钛粉和镍粉作原料,在氩气保护条件下,采用预热点燃模式和热爆模式制得了不同形态的多孔TiNi形状记忆合金。采用SEM和XRD分析了样品的孔洞特征和相组成,测试了其力学性能,并研究了合成条件参数与样品表面形貌和孔隙状态之间的关系。结果表明,用SHS法制备TiNi多孔体合金是可行的。  相似文献

 The paper investigates the performance of various time stepping schemes for coupled displacement and pore pressure analysis. A number of alternative forms of the automatic time stepping method proposed by Sloan and Abbo (1999a) are also presented. These alternative schemes use different updates for the displacements and pore pressures and also adopt different starting conditions for the iterations. The automatic schemes are compared with an implicit θ-method, as well as an explicit method, through analysis of a variety of problems involving undrained loading, drained loading, and consolidation for Mohr-Coulomb and critical state models. As expected, the numerical results confirm that the explicit scheme is neither accurate nor robust. Although the implicit θ-method is accurate and fast, it fails to give a solution in a number of cases where the time step is large. The automatic schemes are shown to be accurate, fast and generally robust. Two of the automatic schemes proposed never fail to furnish a solution for the cases considered. In addition, all the automatic schemes are able to constrain the time-stepping (temporal integration) error in the displacements and pore pressures to lie near a prescribed tolerance, provided the iteration error tolerance is properly chosen. For complex soil models, it is important that the latter is set sufficiently small in order for the schemes to be able to constrain the time-stepping error to lie within a prescribed tolerance.
