Highly sensitive label-free techniques of DNA determination are particularly interesting in relation to the present development of the DNA sensors. We show that subnanomolar concentrations (related to monomer content) of unlabeled DNA can be determined using copper solid amalgam electrodes or hanging mercury drop electrodes in the presence of copper. DNA is first treated with acid (e.g., 0.5 M perchloric acid), and the acid-released purine bases are directly determined by the cathodic stripping voltammetry. Volumes of 5-3 microL of acid-treated DNA can easily be analyzed, thus making possible the determination of picogram and subpicogram amounts of DNA corresponding to attomole and subattomole quantities of 1000-base pair DNA. Application of this determination in DNA hybridization detection is demonstrated using surface H for the hybridization (superparamagnetic beads with covalently attached DNA probe) and the mercury electrodes only for the determination of DNA selectively captured at surface H. 相似文献

A 2‐in. active‐matrix display was demonstrated, containing 4096 solution‐processed polymer‐based transistors. By using the polymer‐dispersed liquid‐crystal display (LCD) effect, this results in a reflective, low‐power display with paper‐like contrast. The influence of the transistor parameters on the display performance is analyzed by use of a model for charging and discharging of the pixel capacitors. Good agreement was obtained between the model and the experimental data. Scaling behavior allows estimation of the performance required for transistors in a quarter‐VGA display. We studied the late-stage spinodal decomposition of the polystyrene–cyclohexanol system in relation to membrane formation. Phase separation was effected by the removal of thermal energy from the homogeneous polymer solution. The ultimate morphology of the phase-separated systems has been studied using electron microscopy and has been found to be strongly affected by the quenching time. A shift from a highly interconnected open-cell structure to a closed-cell structure has been observed, indicating the transition of the spinodal decomposition from early to late stages. The cell growth in the late stage has been quantitatively analyzed and a power-law relationship between the cell size and quenching time has been found. The resulting exponent of 0.61 is consistent with literature values from light-scattering measurements, as well as from theoretical derivations. © 1995 John Wiley & Sons, Inc. 相似文献

Methyl linoleate was oxidized in model systems consisting of either cellulose or casein with which the lipid was dispersed
with water containing cobalt salts. The dispersion was extruded into Warburg flasks, frozen and freeze-dried at 100 μ Hg and
with platen temperatures of 80 F. The samples were then humidified over saturated salt solutions to give moisture contents
from less than 1 g H2O/100 g solids up to 30 g H2O/100 g solids. The higher moisture contents were obtained by addition of glycerol to the model system during preparation
and humidification at 60–75% RH. Chelating agents including EDTA and citric acid in concentrations of 1 to 10 moles per mole
of cobalt ion were used in some experiments. Oxidation was followed manometrically and by peroxide analysis. At low water
contents, water acts as an antioxidant through hydration of metallic catalysts. As the moisture content increases, water promotes
oxidation through its solvent activity. In the region of capillary condensation, antioxidant effect of metal hydration is
overshadowed by the prooxidant effect of metal solubilization. The water soluble chelating agents such as EDTA act on metals
in aqueous solution and their activity is promoted by increased moisture content.

In this study, the acute copper sensitivity of field-collected cladoceran species was determined using their natural surface waters and a standard reconstituted test water as test medium. A total of 43 species were collected on two occasions from six different sites, representing different water types and chemistries in Europe. The collected species belonged to four different families (Daphniidae, Bosminidae, Macrothricidae, Chydoridae) and 11 different genera (Daphnia, Ctenodaphnia, Ceriodaphnia, Simocephalus, Scapholeberis, Alona, Acroperus, Chydorus, Eurycercus, Disparalona, Pleuroxus). In acute experiments with immobilization as end point, the 48-h median effective concentrations (48-h EC50) for the cladoceran species ranged from 5.30 to 70.6 microg of Cu L(-1) in standard test water and from 9.60 to 853 microg of Cu L(-1) in natural waters. The mean site sensitivity (the geometric mean of 48-h EC50 values of species within a community) ranged from 10.1 to 27.4 microg of Cu L(-1) in standard water and from 16.4 to 281 microg of Cu L(-1) in natural water. This indicates that bioavailability is more importantthan inter-community (species composition) differences in determining the variability of copper toxicity across different aquatic systems. For the four surface waters that had a pH within the range for which the acute Daphnia magna biotic ligand model (BLM) has previously been successfully validated, the BLM predicted 48-h EC50 values for 27 of the 28 tested cladoceran species within factor of 2 of the observed values. For the same sites, all community sensitivities were predicted within a factor of 2.3. The BLM was clearly over-protective for the two acidic surface waters tested. Hence, the BLM can be considered a valuable tool for estimating the potentially harmful effects of copperto natural cladoceran communities, but more research will be needed for acidic surface waters. 相似文献

The main aim of data analysis in biochemical metrology is the extraction of relevant information from biochemical data measurements. A system of extended exploratory data analysis (EDA) based on the concept of graphical tools for sample data summarization and exploration is proposed and the original EDA algorithm in S-Plus is available on the Internet at http://www.trilobyte.cz/EDA. To check basic assumptions about biochemical and medical data is to examine the independence of sample elements, sample normality and homogeneity. The exact assessment of the mean-value and the variance of steroid levels in controls is necessary for the correct assessment of the samples from patients. Data examination procedures are illustrated by a determination of the mean-value of 17-hydroxypregnenolone in the umbilical blood of newborns. For an asymmetric, strongly skewed sample distribution corrupted with outliers the best estimate of location seems to be the median. The Box–Cox transformation improves a sample symmetry. The proposed procedure gives reliable estimates of a mean-value for an asymmetric distribution of 17-hydroxypregnenolone when the arithmetic mean can not be used. 相似文献

The proteins streptavidin and avidin were electrochemically detected in solution by adsorptive transfer stripping square wave voltammetry (AdTS SWV) at a carbon paste electrode (CPE). AdTS SWV was used to quantify biotinylated oligonucleotides, DNA hybridizations, and avidin in extracts of transgenic avidin maize. The detection limits of denatured and native streptavidin were 6 pM and 120 nM, respectively. The results demonstrated that streptavidin/avidin AdTS SWV is a sensitive and specific method for quantifying DNA and proteins in biological samples such as foods and tissue extracts, including genetically modified crops (avidin maize) and other plants in neighboring fields. 相似文献
