The objective of this study was to control the survival or biofilm formation of Cronobacter spp. on stainless steel surfaces using Paenibacillus polymyxa. The antibacterial activity of a cell‐free culture supernatant (CFCS) of P. polymyxa against Cronobacter spp. was found to vary with P. polymyxa incubation time. Maximum activity occurred when P. polymyxa was incubated at 25 or 30 °C for 96 h. When the CFCS was introduced to Cronobacter spp. adhered to stainless steel strips at 25 °C for up to 72 h, the CFCS successfully inhibited Cronobacter biofilm formation. Additionally, stainless steel surfaces with a preformed P. polymyxa biofilm were exposed to Cronobacter spp. suspensions in PBS or 0.1% peptone water at 3, 5, or 7 log CFU/mL to facilitate its attachment. The Cronobacter population significantly decreased on this surface, regardless of inoculum level or carrier, when the P. polymyxa biofilm was present. However, the microbial population decreased within 6 h and remained unchanged thereafter when the surface was immersed in an inoculum suspended in 0.1% peptone water at 5 or 7 log CFU/mL. Directed energy deposition (DED), equipped with 5-axis tool mechanism and no additional machining process, turned out to be able to deposit overhang/undercut features with no support directly on a part in multiple directions. When performing the deposition, two additional axes of rotating and tilting newly added to the working table where the part is typically fixed need to be precisely controlled using an advanced process management technique. The previous approach for slicing algorithm corresponding with multi-axis tool paths along the part and providing a simple stepwise two dimensional layering process of repetitive ±90 degree tilting of the base table can cause unavoidable process delay whenever the base table needs to be tilted for conforming the geometry of overhang/undercut features. Another clear and present barrier to the approach, in addition, is the physical interferences between the tool and the workpiece moving separately during the build. This study proposes a Spherically curved layer (SCL) model providing interference-free 5-axis tool-part movement, a continuous and synchronized interaction between the tool and the part each separately but precisely controlled while kept contacted together, for the build of overhangs. A methodological approach generating SCL information directly from an STL model, followed by the implementation onto various overhang problems as case studies using the proposed methodology, will be discussed in this paper. 相似文献

The chronic effects of dietary selenium (Se) exposure in juvenile Sacramento splittail (Pogonichthys macrolepidotus) were investigated in the laboratory. A total of 960 (40 fish per tank, 3 tanks per diet) 7-month-old juvenile splittail were fed one of eight Purified-Casein diets supplemented with selenized yeast for 9 months in a flow-through system. These diets contained the following: 0.4 (control), 0.7, 1.4, 2.7, 6.6, 12.6, 26.0, and 57.6 mg of Se kg(-1) dry weight. Survival, Se tissue concentration, growth, gross morphology, and liver histopathology were assessed at 5- and 9-month of exposure. Mortalities occurred only in the two highest Se treatments and were accounted for 8.3 and 18.3% at 5-month and 10.0 and 34.3% at 9-month, respectively. Liver and muscle Se concentration were significantly correlated with dietary Se concentration. Fish exposed to 0.4-12.6 mg of Se kg(-1) diets had reached equilibrium in liver Se concentration by 5 month. Splittail fed diets at concentrations > or =26.0 mg of Se kg(-1) had not reached equilibrium in liver, and muscle Se concentrations and grew significantly slower (p < 0.05) at 5- and 9-month exposure. Se-induced deformities were observed in fish fed > or =2.7 mg of Se kg(-1) diets at 5-month and in fish fed > or =0.7 mg of Se kg(-1) diets at 9-month. Fish fed 26.0 and 57.6 mg of Se kg(-1) diets had higher liver lesion scores at 5-month while fish fed 6.6 and 57.6 mg of Se kg(-1) diet had higher liver lesion scores at 9-month. Results indicate that survivals, growth, changes of tissue Se concentrations, and histopathology of juvenile splittail were dose-dependent, but their response thresholds to dietary Se concentrations differed and depended on treatment concentrations and duration of exposure. Chronic exposure to 6.6 mg of Se kg(-1) diet induced deleterious health effects that can potentially impact survival of juvenile splittail. 相似文献

The paper presents a numerical analysis to predict the behavior of the flexible media by considering the effect of dynamic contact interaction between media and guide, and the aerodynamic forces. The result of numerical model is compared with that of experiment, showing a good agreement under the flexible media condition of low exit velocity. In a state of high exit velocity for the flexible media, both results of the simulation and experiments show a difference which may be needed a more accurate analysis of fluid dynamics. 相似文献

Adrenal insufficiency is a complication of chronic corticosteroid therapy. Unexplained hypotension may be a manifestation of an adrenal insufficient state in patients with a history of corticosteroid therapy on hemodialysis. We present a series of five cases of patients on nocturnal home hemodialysis with hypotension as the main manifestation of adrenal insufficiency. Unexplained hypotension in patients with a history of corticosteroid therapy should prompt the managing clinician to consider adrenal insufficiency as a possible cause. 相似文献

A two-step calcination synthesis, considering the potential for mass production, of lithium titanium oxide powder was carried
out to fabricate a single Li4Ti5O12 phase, which is useful for anode electrode material of Li-based rechargeable battery as well as an electrode for supercapacitor.
The final composition is controlled by adding more TiO2 powder into powder gained at one calcination process during the two calcination process. We investigated the influence of
excess TiO2 on the structural characteristics of lithium titanium oxide synthesized by the two-step calcination method. X-ray diffraction
(XRD) and Fourier transform infrared spectroscopy (FT-IR) measurements showed that the as-synthesized powder had a spinel
crystal structure as well as A composition of 4: 5: 12. In addition, a high resolution transmission electron microscopy (HRTEM)
analysis revealed that the fabricated powder exhibited a single crystalline phase formation. These results indicated that
the powder synthesized in the one-step calcination process showed coexistence crystalline phases, which are the Li4Ti5O12 and Li2.39Ti3.4O8 phase. However, in the two-step calcination process, the powder synthesized showed the single crystalline Li4Ti5O12 phase. A very uniform grain size of the as-synthesized powder was shown in a field emission scanning electron microscopy
(FESEM). These results suggested that the two-step calcination process can be used for synthesis of single crystalline Li4Ti5O12 powder with uniform grain shape and provide motivation to pursue mass production of lithium titanium based oxide powder for

Tyrosinase catalyzes two distinct sequential reactions in melanin biosynthesis: The hydroxylation of tyrosine to dihydroxyphenylalanine (DOPA) and the oxidation of DOPA to dopaquinone. Developing functional modulators of tyrosinase is important for therapeutic and cosmetic purposes. Given the abundance of thiourea moiety in known tyrosinase inhibitors, we studied other thiourea-containing drugs as potential tyrosinase inhibitors. The thiourea-containing drugs in clinical use were retrieved and tested for their ability to inhibit tyrosinase. We observed that methimazole, thiouracil, methylthiouracil, propylthiouracil, ambazone, and thioacetazone inhibited mushroom tyrosinase. Except for methimazole, there was limited information regarding the activity of other drugs against tyrosinase. Both thioacetazone and ambazone significantly inhibited tyrosinase, with IC50 of 14 and 15 μM, respectively. Ambazone decreased melanin content without causing cellular toxicity at 20 μM in B16F10 cells. The activity of ambazone was stronger than that of kojic acid both in enzyme and melanin content assays. Kinetics of enzyme inhibition assigned the thiourea-containg drugs as non-competitive inhibitors. The complex models by docking simulation suggested that the intermolecular hydrogen bond via the nitrogen of thiourea and the contacts via thione were equally important for interacting with tyrosinase.
