A real-time PCR assay was designed to detect a 162-bp fragment of the ssrA gene in Listeria monocytogenes. The specificity of the assay for L. monocytogenes was confirmed against a panel of 6 Listeria species and 26 other bacterial species. A detection limit of 1-10 genome equivalents was determined for the assay. Application of the assay in natural and artificially contaminated culture enriched foods, including soft cheese, meat, milk, vegetables and fish, enabled detection of 1-5 CFU L. monocytogenes per 25g/ml of food sample in 30h. The performance of the assay was compared with the Roche Diagnostics 'LightCycler foodproof Listeria monocytogenes Detection Kit'. Both methods detected L. monocytogenes in all artificially contaminated retail samples (n=27) and L. monocytogenes was not detected by either system in 27 natural retail food samples. The method developed in this study has the potential to enable the specific detection of L. monocytogenes in a variety of food types in a time-frame considerably faster than current standard methods. The potential of the ssrA gene as a nucleic acid diagnostic (NAD) target has been demonstrated in L. monocytogenes. We are currently developing NAD tests based on the ssrA gene for a range of common foodborne and clinically relevant bacterial pathogens. 相似文献
Ten wells (EW-4, EW-5, EW-6, EW-7, EW-8, EW-9, EW-10, EW-12, EW-13 and EW-15) were interpreted using the composite well logs, data of core analysis, gamma-ray logs, formation micro-imager logs (FMI), and 3D seismic data in SEGY format to understand the stratigraphy of the onshore, Nile Delta, Egypt. The amplitude analysis of 3-D seismic horizon slice of Lower Abu Madi rock unit together with the lithostratigraphic correlation through the study area depending on the gamma-ray log “HSGR” (left to right increasing), and the identification of type of bed geometry, nature of bed contacts, type of the sedimentary structures and the dominant formative paleocurrents by using some available borehole micro-resistivity images (FMI) and core photos. All of these techniques are used together to define the different depositional facies and depositional environment of the Messinian clastics (Lower Abu Madi rock unit), which is considered to be the main reservoir in the El-Wastani gas field, onshore Nile Delta, Egypt. The present study of depositional pattern of the Upper Miocene clastics reservoir (Lower Abu Madi rock unit) revealed that it is represented by high sinuous meandering channels or paleo-valley and three types of fluvial facies were defined; channel fill, channel margin, and floodplain basin. 相似文献

We present an approach for the visualization and interactive analysis of dynamic graphs that contain a large number of time steps. A specific focus is put on the support of analyzing temporal aspects in the data. Central to our approach is a static, volumetric representation of the dynamic graph based on the concept of space-time cubes that we create by stacking the adjacency matrices of all time steps. The use of GPU-accelerated volume rendering techniques allows us to render this representation interactively. We identified four classes of analytics methods as being important for the analysis of large and complex graph data, which we discuss in detail: data views, aggregation and filtering, comparison, and evolution provenance. Implementations of the respective methods are presented in an integrated application, enabling interactive exploration and analysis of large graphs. We demonstrate the applicability, usefulness, and scalability of our approach by presenting two examples for analyzing dynamic graphs. Furthermore, we let visualization experts evaluate our analytics approach.
