Spin-lattice (T1) relaxation times were measured by an inversion-recovery magnetic resonance imaging method with a slice-selective inversion pulse (SIP), a non-selective rectangular inversion pulse (RIP), or a B1-insensitive adiabatic inversion pulse (AIP). Data analysis either assumed perfect inversion (two-parameter fit) or allowed for imperfect inversion (three-parameter fit). Imperfect inversion pulses caused low T1 values in phantoms with a two-parameter fit, while three-parameter T1 estimates were accurate over the range 430-2670 ms. A difference of approximately 10% between two-parameter and three-parameter T1 values in normal human brain tissue was attributed to B1 inhomogeneity with the slice-selective inversion pulse and rectangular inversion pulse, to the slice profile with the slice-selective inversion pulse, and to T2 effects for the adiabatic inversion pulse. Any T1 method that relies on accurate flip angles may have a significant systematic error in vivo. Phantom accuracy does not ensure accuracy in vivo, because phantoms may have a more homogeneous B1 field and a longer T2 than do biological samples.  相似文献

The aim of this study was to evaluate clinically, histologically and histometrically the use of anorganic bovine bone matrix (i.e. Bio-oss) as a grafting material for maxillary sinus augmentation procedures. In 4 adult male rhesus monkeys (i.e. Macaca mulatta) the 1st, 2nd and 3rd maxillary molars on one side of the jaws were extracted. The remaining bone between the alveolar crest and the bottom of the sinus was then reduced to 3-4 mm. After 3 months, maxillary sinus augmentation procedures were performed on one side of the jaws in each monkey and the sinuses were grafted with the bovine bone matrix. At that time, 2 IMZ pure titanium plasma coated implants were immediately placed into the augmented sinuses (i.e. simultaneous implants-loaded group). After 4 months, 2 additional similar implants were placed into these previously augmented sinuses (i.e. delayed implants-loaded group). Four months later, the abutment connection was performed and all 4 implants were loaded with a gold-alloy bridge for 6 months (i.e. until sacrifice of the animals). The contralateral side of each monkey received the same treatment with the exception that the extractions were performed 7 months after those in the opposite side and that the implants in this side were not loaded. Thus, 2 additional study groups (i.e. simultaneous implants-unloaded group and delayed implants-unloaded group) were obtained. Clinically, all loaded implants were stable at the day of sacrifice. Histologically, the grafted sinuses exhibited significant bone formation with integration of the bovine bone matrix particles to the new bone. Direct mineralized bone-to-implant contact was greater for the delayed implant placement groups than for the implants installed simultaneously with the sinus augmentation. Furthermore, the percentage of direct mineralized bone-to-implant contact was greater in the residual bone than in the augmented area. It was concluded that the anorganic bovine bone matrix facilitated bone formation and implant osseointegration in the augmented sinuses and that the delayed implant placement in combination with the sinus augmentation procedure seemed to be preferable.  相似文献

The aim of this work was to investigate the effect of tissue culture on the intracellular amino acid pool in both freshly isolated and surgically prepared saphenous vein segments taken from patients undergoing coronary artery bypass surgery (number of patients, n = 8). Viability of freshly isolated vein rings, indicated by ATP concentration, was maintained in culture (321 +/- 41 vs. 277 +/- 31 nmol (g wet wt)-1, 0 vs. 14 days). The initial decrease in ATP concentration in surgically prepared rings was significantly reversed following 14 days in culture from 135 +/- 26 to 201 +/- 18 nmol (g wet wt)-1 (P < 0.05). Freshly isolated vein rings maintained their intracellular free amino acid pool during the 14 days in culture (from 166 +/- 25 to 166 +/- 23 mumol (g protein)-1). Surgical preparation of vein rings induced a decrease in the intracellular free amino acid pool (from 166 +/- 25 to 87 +/- 15 mumol (g protein)-1, P < 0.05). This decrease was partially reversed after 14 days in culture (140 +/- 19 mumol (g protein)-1). Although the total amino acid pool in both types of vein rings after 14 days in culture was similar, there were variations in individual amino acid concentrations. Freshly isolated rings showed an increase in glutamine concentration and a decrease in alanine and aspartate concentrations after 14 days in culture. Surgically prepared vein rings showed a decrease in aspartate concentration and an increase in concentrations of glutamine, asparagine, glutamate and glycine. The changes in individual intracellular free amino acid concentrations, which were largely determined by the corresponding concentrations in the medium, indicates that culture media should be supplemented with taurine, aspartate and alanine.  相似文献

The impact of cardiac hypertrophy on myocardial biochemical and physiological responses to ischaemia-reperfusion (I-R) was investigated in vivo. Hypertrophy was produced by aortic constriction (PH) or swimming training (TH). Open-chest rat hearts in PH, TH and a sedentary control group (SC) were subjected: (1) to ischaemia, by surgical occlusion of the main descending branch of the left coronary artery for 30 min; (2) to I-R, by releasing the occluded blood vessel for 15 min; or (3) to a sham operation. Ischaemia per se had little effect on heart oxidative and antioxidant status, or lipid peroxidation. However, I-R significantly decreased glutathione (GSH) content, increased glutathione disulfide (GSSG) content, and reduced GSH/GSSG ratio in the SC hearts. These alterations were associated with decreased activities of GSH peroxidase and GSSG reductase, and an increase in lipid peroxidation. Myocardial ATP, total adenine nucleotide content and energy charge in SC were significantly decreased after ischaemia, whereas levels of purine nucleotide derivatives, particularly adenosine, were elevated. No significant alteration of GSH status of adenine nucleotide metabolism occurred after ischaemia or I-R in hypertrophied hearts. In both PH and TH, glutathione content was significantly higher than in SC, whereas activities of GSH peroxidase and GSSG reductases were lower. TH rats maintained a higher heart rate (HR), peak systolic pressure, and energy charge during I-R. These data indicate that hypertrophied but well-functioned hearts may be more resistant to I-R induced disturbances of myocardial oxidative and antioxidant functions.  相似文献

A survey on use of benzodiazepines was conducted among participants in the Florida Geriatric Research Program. In 1978-79, benzodiazepine use was reported by 14.4% of 1448 women and 9.7% of 855 men; in 1984-85, by 12.0% of 1429 women and 5.9% of 784 men; and in 1991-92, by 13.4% of 1124 women and 6.6% of 497 men. The changes were not significant. During this period the mean ages of the women increased from 74.6 to 78.1 years and the men from 75.0 to 80.2 years. Chlordiazepoxide, diazepam, and flurazepam accounted for 98.1% of all benzodiazepines used in 1978-79 and for 35.5% in 1991-92, when alprazolam, lorazepam, and temazepam accounted for 47.1% of benzodiazepines used.  相似文献

This paper describes a novel application of singular value decomposition (SVD) of subsets of the phase-space trajectory for calculation of the attractor dimension of a small data set. A certain number of local centres (M) are chosen randomly on the attractor and an adequate number of nearest neighbours (q=50) are ordered around each centre. The local intrinsic dimension of a local centre is determined by the number of significant singular values and the attractor dimension (D 2) by the average of the local intrinsic dimensions of the local centres. The SVD method has been evaluated for model data and EEG. The results indicate that the SVD method is a reliable approach for estimation of attractor dimension at moderate signal to noise ratios. The paper emphasises the importance of SVD approach to EEG analysis.  相似文献

In myelodysplastic syndromes (MDS), pancytopenia leads to a high risk of infectious and hemorrhagic complications. The progression to acute myeloid leukemia adds to morbidity and mortality. While transfusions of red blood cells and platelets are still a cornerstone of the therapy, the clinical use of recombinant hematopoietic growth factors has enlarged the range of therapeutic applications in patients with MDS. It is possible to reverse neutropenia by administration of G-CSF (granulocyte colony stimulating factor) or GM-CSF (granulocyte-monocyte colony stimulating factor). In the case of a severe infection, therapeutic administration of G-CSF together with antibiotics might be justified in otherwise neutropenic MDS patients. Since especially patients with only slight impairment of erythropoiesis and no transfusion dependency have the highest response rates but need erythropoietin (EPO) the least, pharmacoeconomic analyses are urgently needed. Controlled randomized trials will have to ascertain wether combinations of EPO with G-CSF or GM-CSF are of benefit. Clinical studies with thrombopoietin (megakaryocyte growth and differentiation factor) have to be initiated to find out whether thrombocytopenia in MDS can be reversed.  相似文献

Large dynamic nuclear polarization signal enhancements (up to a factor of 100) were obtained in the solid-state magic-angle spinning nuclear magnetic resonance (NMR) spectra of arginine and the protein T4 lysozyme in frozen glycerol-water solutions with the use of dynamic nuclear polarization. Polarization was transferred from the unpaired electrons of nitroxide free radicals to nuclear spins through microwave irradiation near the electron paramagnetic resonance frequency. This approach may be a generally applicable signal enhancement scheme for the high-resolution solid-state NMR spectroscopy of biomolecules.  相似文献
