5-Methylcytosine and 5-hydroxymethylcytosine are epigenetic modifications involved in gene regulation and cancer. We present a new, simple, and high-throughput platform for multi-color epigenetic analysis. The novelty of our approach is the ability to multiplex methylation and de-methylation signals in the same assay. We utilize an engineered methyltransferase enzyme that recognizes and labels all unmodified CpG sites with a fluorescent cofactor. In combination with the already established labeling of the de-methylation mark 5-hydroxymethylcytosine via enzymatic glycosylation, we obtained a robust platform for simultaneous epigenetic analysis of these marks. We assessed the global epigenetic levels in multiple samples of colorectal cancer and observed a 3.5-fold reduction in 5hmC levels but no change in DNA methylation levels between sick and healthy individuals. We also measured epigenetic modifications in chronic lymphocytic leukemia and observed a decrease in both modification levels (5-hydroxymethylcytosine: whole blood 30 %; peripheral blood mononuclear cells (PBMCs) 40 %. 5-methylcytosine: whole blood 53 %; PBMCs 48 %). Our findings propose using a simple blood test as a viable method for analysis, simplifying sample handling in diagnostics. SiC-fiber–reinforced SiC matrix composite cladding for light water reactor fuel elements must withstand high-temperature steam oxidation in a loss-of-coolant accident scenario (LOCA). Current composite designs include an outer monolithic SiC layer, in part, to increase steam oxidation resistance. However, it is not clear how such a structure would behave under high-temperature steam in the case when the monolithic layer cracks and carbon interphases and SiC fibers are exposed to the environment. To fill this knowledge gap, stress-rupture tests of prototypic SiC composite cladding at 1000°C under steam and inert environments were conducted. The applied stress was ∼120 MPa, which was beyond the initial cracking stress. The failure lifetime under steam was 400–1300 s, while 75% of the composite specimens did not fail after 3 h of total exposure under inert gases. Microstructural observations suggest that steam oxidation activated slow crack growth in the fibers, which led to failure of the composite. The results from this study suggest that stress rupture in steam environments could be a limiting factor of the cladding under reactor LOCA conditions. 相似文献

A carbide boronizing method was first developed to produce dense boron carbide‐ zirconium diboride (“B4C”–ZrB2) composites from zirconium carbide (ZrC) and amorphous boron powders (B) by Spark Plasma Sintering at 1800°C–2000°C. The stoichiometry of “B4C” could be tailored by changing initial boron content, which also has an influence on the processing. The self‐propagating high‐temperature synthesis could be ignited by 1 mol ZrC and 6 mol B at around 1240°C, whereas it was suppressed at a level of 10 mol B. B8C–ZrB2 ceramics sintered at 1800°C with 1 mole ZrC and 10 mole B exhibited super high hardness (40.36 GPa at 2.94 N and 33.4 GPa at 9.8 N). The primary reason for the unusual high hardness of B8C–ZrB2 ceramics was considered to be the formation of nano‐sized ZrB2 grains. 相似文献

许多材料在医学领域应用广泛,例如,整体替换硬组织或软组织的元件(如骨盆、骨头、关节、植牙等)、修补、诊断或矫正仪器(如起搏器、心脏阀等)。这些材料不仅要有好的力学性能,还要保持长期稳定,不能与人体相排斥。由于陶瓷材料在生理环境中具有强度高、生物相容性强和稳定性好的优点,人们研究用陶瓷材料替换骨骼。从20 世纪70 年代起,欧洲人用陶瓷组件置换整个髋关节。这些组件主要由氧化铝和氧化锆单体制成。然而,在有水环境中,氧化锆会发生低温降解。目前人们的研究重点在于提高陶瓷组件的强度和耐磨性,同时缩小其尺寸并延长其使用寿命。研究中使用的材料是氧化锆增韧的氧化铝复合陶瓷和其它氧化铝复合陶瓷,不再是单体陶瓷。另外,还可以使用氧化铝和氧化锆功能梯度复合材料。该梯度材料可以利用电泳沉积法(EPD)制得,其表面为纯氧化铝,中心部分为均匀的氧化铝、氧化锆复合材料,中间过渡部分是呈连续梯度渐变的氧化铝、氧化锆复合材料,烧成后会产生剩余热应力。设计这样的梯度结构是为了使复合材料具有最大表面压应力和最小内部张应力,与纯氧化铝组件相比,提高了强度和耐磨性。 相似文献

Detailed stiffness and internal friction ( Q −1 ) versus temperature curves were obtained for liquid-phase-sintered silicon carbides using advanced resonant beam analysis up to 1400°C. As-sintered materials display a stable Q −1 -peak near 1100°C, superimposed on an increasing background. The change of stiffness associated with the damping peak is quantitatively related to the amount of matter in pockets of the amorphous intergranular phase in which the refractory SiC matrix grains are embedded. The successful removal of the amorphous pockets by annealing at 1900°C is deduced from the disappearance of the damping peak and confirmed with transmission electron microscopy. 相似文献

Nitrogenated diamond-like (DLC:N) carbon thin films have been deposited by microwave surface wave plasma chemical vapor deposition on silicon and quartz substrates, using argon gas, camphor dissolved in ethyl alcohol composition and nitrogen as plasma source. The deposited DLC:N films were characterized for their chemical, optical, structural and electrical properties through X-ray photoelectron spectroscopy, UV/VIS/NIR spectroscopy, Raman spectroscopy, atomic force microscope and current–voltage characteristics. Optical band gap decreased (2.7 to 2.4 eV) with increasing Ar gas flow rate. The photovoltaic measurements of DLC:N / p-Si structure show that the open-circuit voltage (Voc) of 168.8 mV and a short-circuit current density (Jsc) of 8.4 μA/cm2 under light illumination (AM 1.5 100 mW/cm2). The energy conversion efficiency and fill factor were found to be 3.4 × 10− 4% and 0.238 respectively. 相似文献

This study was performed to investigate and scale-up the hot-melt coating process in fluidized beds. A series of well-designed experiments was carried out in a pilot scale unit with 20 kg product capacity to investigate the effects of process variables on the efficiency of the coating of Cefuroxime Axetil with stearic acid. Results showed that the efficiency is at the highest when the fluidization air flow rate is adjusted by considering the changes in the amount of materials present in the unit as well as the changes in the terminal velocities of particles during the process.With the objective to scale-up the hot-melt coating process from pilot to production scale, a dynamic thermodynamic model based on conservation equations of mass and energy was developed. Predictive accuracy of the model was assessed by applying it to the pilot scale unit and comparing its predictions with the online measurements taken on the same unit. Results showed that the predictions of the model agree well with the measurements. Utilizing this model and taking several experiments performed in the pilot scale unit as a basis, scaling up of the hot-melt coating process was carried out. Comparisons of the model predictions with the measurements taken on the production scale unit (200 kg product capacity) revealed that the model is able to reproduce the product attributes and the outlet air temperatures across scales. Therefore, it proves to be a promising tool that can be used in the scale-up of the hot-melt coating processes in fluidized beds. 相似文献

A novel application, utilizing polyacrylamide‐g‐carboxymethyl cellulose (CMC‐g‐PAM) in concentrating dilute solutions of Bovine serum albumin (BSA), was investigated. The grafting reaction parameters were investigated and the hydrogel smartness was verified. FT‐IR proved that the grafting reaction occurred between the hydroxyl group located in anhydroglucose C2 position of CMC and the π‐bond of PAM and SEM confirmed a changed morphology to a fibrillar structure. The pH sensitivity was proved; as the grafted polymer attained its maximum swelling at pH 7.2 while the minimum swelling was observed under acidic conditions (pH 1‐3). The rate of water uptake in the grafted polymer hydrogel was higher than that of the homopolymer hydrogel and the swelling behaviors of both hydrogels obeyed second‐order kinetics. The tested hydrogel showed a high potency towards concentrating BSA solutions with a concentration factor of 1 to 4.5 times and recovery of 60–90%. The concentration factor increased linearly with increasing both the polymer concentration and the process time and decreased with the increase in the protein concentration. The grafted polymer had stable efficiency in the concentration process for 20 cycles. The obtained results have recommended the employment of the prepared CMC‐g‐PAM hydrogel in the down stream protein concentration process in the industrial scale. © 2011 Wiley Periodicals, Inc.
