Since the chemical synthesis of vitamin B12 requires more than 70 steps, the production of vitamin B12 has been achieved by microorganism fermentation with additional brief chemical modifications. In an effort to increase the productivity of vitamin B12, we tried to express 10 genes belonging to the hem, cob and cbi gene families involved in the synthesis of vitamin B12 in Propionibacterium freudenreichii, which is a known producer of vitamin B12. In a recombinant P. freudenreichii clone that harbored the expression vector containing a cobA, cbiLF, or cbiEGH, we obtained an increase in vitamin B12 production of 1.7-, 1.9-, and 1.5-fold higher, respectively, than that in the microorganism without any cloned genes in the expression vector pPK705. The cobU and cobS genes caused a slight increase in the production of vitamin B12. Furthermore, we achieved multigene expression in P. freudenreichii. In a recombinant P. freudenreichii clone that harbored an exogenous gene, hemA, from Rhodobacter sphaeroides and endogenous hemB and cobA genes, we successfully achieved the production of about 1.7 mg/l vitamin B12, 2.2-fold higher than that produced by P. freudenreichii harboring pPK705.  相似文献

We have constructed a near-real-time combined imager suitable for simultaneous ultrasound and near-infrared diffusive light imaging and coregistration. The imager consists of a combined hand-held probe and the associated electronics for data acquisition. A two-dimensional ultrasound array is deployed at the center of the combined probe, and 12 dual-wavelength laser source fibers (780 and 830 nm) and 8 optical detector fibers are deployed at the periphery. We have experimentally evaluated the effects of missing optical sources in the middle of the combined probe on the accuracy of the reconstructed optical absorption coefficient and assessed the improvements of a reconstructed absorption coefficient with the guidance of the coregistered ultrasound. The results have shown that, when the central ultrasound array area is in the neighborhood of 2 cm x 2 cm, which corresponds to the size of most commercial ultrasound transducers, the optical imaging is not affected. The results have also shown that the iterative inversion algorithm converges quickly with the guidance of a priori three-dimensional target distribution, and only one iteration is needed to reconstruct an accurate optical absorption coefficient.  相似文献

Glass-ceramic materials were developed as a sealant in the solid oxide fuel cell (SOFC) in the temperature range of 800 -850 ℃. The glass materials were based on the glass and glass-ceramic in the BaO-CaO-Al2O3-SiO2-La2O3-B2O3 system. The thermal expansion coefficient (TEC) decreased with lower Ba^2+ content and higher Ca^2+ content, but the glass transition temperature and crystallization temperature increased greatly with an increase in Ca^2+ content and a decrease in Ba^2+ content, when the other components in the sealant were invariable. The TEC of the sealant with Ba^2+ content of 25.4% was 10.8 × 10^-6 K^- 1(temperature range from 25 to 850℃), and its softening temperature was 950 ℃. The TEC of the sealant accorded well with that of La0.9Sr0.1Ga0.5Mg0.2o3- 6(LSGM) with a mismatch of only 3%. The sealant had superior stability and compatibility with the LSGM electrolyte during the process of operation in SOFC. The weight loss of the sealant with Ba^2+ content of 25.4% was approfimately zero after heat-treated at 800℃ for 500 h in H2 and O2 atmosphere, respectively.  相似文献

以双溪口面板堆石坝实测沉降监测资料为基础,通过基于免疫遗传算法的位移反分析法对堆石料E-B模型参数进行反演分析,获取了符合工程实际的模型参数。根据反馈分析得到的计算结果,采用邓肯-张弹性非线性E-B模型,分析研究了大坝竣工期及蓄水期的变形及应力分布状态,计算结果表明,大坝蓄水期位移增量较小,面板应力正常,大坝运行安全。  相似文献

Allicin, the main active component derived from a traditional flavoring agent - garlic, exerts a variety of biological effects. Unfortunately, its high potential in food processing is limited by its sensibility towards heat and alkaline conditions and its pungent smell. In this research, allicin nanoliposomes were prepared using reverse-phase evaporation method in an attempt to circumvent these shortcomings. The process conditions were optimized by response surface methodology, and the optimal parameters obtained were: lecithin-allicin ratio 3.70:1, lecithin-cholesterol ratio 3.77:1, ultrasonic time 3.40 min, organic phase-aqueous phase ratio 3.02:1. The experimental maximum of entrapment efficiency was 75.20 ± 0.62%, which was consistent with the predictive value (75.05%). Various physicochemical characteristics of allicin liposomes were determined and evaluated. The mean size of allicin nanoliposomes with well-defined spherical shape was 145.27 ± 15.19 nm and only a negligible difference was found after 30 days of storage. The allicin nanoliposomes gave a zeta potential of −40.10 ± 0.96 mV. Based upon the in vitro release profiles, allicin nanoliposomes exhibited a sustained-releasing potential in addition to the release behavior followed the first-order equation. The results indicated that the encapsulation of allicin into liposomes proved to be a promising technology for more widespread application.  相似文献

Two sandwich-type electrodes (RTIL/C60/SWNT/GCE and RTIL/C60 nanotube/SWNT/GCE) with fullerene C60 or fullerene C60 nanotubes interlayer covered by room temperature ionic liquid (RTIL) membrane on single-walled carbon nanotubes (SWNTs) modified glassy carbon electrode (GCE) surface were fabricated. Cyclic voltammetry and differential pulse voltammetry were used to characterize their electrochemical behaviors and the difference between fullerene C60 and fullerene C60 nanotubes was investigated and the reason was presumed for the first time. It has been observed that 4 pairs of quasi-reversible redox peaks occurred at the RTIL/C60/SWNT/GCE, while only two peaks at the RTIL/C60 nanotube/SWNT/GCE. The cathodic and anodic peak potentials did not shift with the change of pH, indicating that no protons were involved in the electrode reaction processes. So the redox reaction of the fullerene C60 or fullerene C60 nanotubes could be considered as a multi-step one-electron transfer process and fullerene C60 behaved like an electron accumulator to accept and release electrons. While, the reductive products of fullerene C60 nanotubes could not be oxidized again in the reverse scan and the electrode process was irreversible. The fullerene C60 monomer released the accumulated electrons more easily than the fullerene C60 nanotubes. The electrochemical behaviors showed that the electrochemical processes at both electrodes were adsorption-controlled processes.  相似文献
