The most commonly used method for serotyping Salmonella spp. is based on the Kaufmann–White scheme, and is composed of serological reactions using antibodies to LPS agglutinins. The multiplex PCR used in this investigation was established by Kim et al. to serotype the 30 most common clinical Salmonella serotypes, as determined by CDC. The PCR assay consists of two five-plex reactions and a single two-plex PCR reaction, based on six genetic loci from Salmonella enterica serotype Typhimurium and four loci from S. enterica serotype Typhi. In this investigation, we further evaluated the method for serotyping Salmonella spp. using a reference collection, environmental samples collected from a Mid-Atlantic region tomato farm study, four food matrices spiked with different Salmonella serotypes and a proficiency test. The PCR assay was first evaluated using DNA isolated from pure cultures of isolates obtained from various clinical and environmental samples, and then DNA isolated from broth cultures of food matrices of “Red round” and Roma tomatoes, Romaine lettuce, green onions and Serrano peppers spiked with serotypes Newport, Typhimurium, Javiana and Saintpaul, respectively. The results showed that the PCR assay correctly serotyped Salmonella spp. from the clinical, environmental, spiked food matrices, and proficiency test samples. These findings are significant because the PCR assay was successful in the identification of Salmonella in the spiked samples in a broth culture containing other non-salmonella organism. This method may be a useful resource for the food safety community.  相似文献

Felix 01 (F01) is a bacteriophage originally isolated by Felix and Callow which lyses almost all Salmonella strains and has been widely used as a diagnostic test for this genus. Molecular information about this phage is entirely lacking. In the present study, the DNA of the phage was found to be a double-stranded linear molecule of about 80 kb. 11.5 kb has been sequenced and in this region A + T content is 60%. There are relatively few restriction endonuclease cleavage sites in the native genome and clones show this is due to their absence rather than modification. A restriction map of the genome has been constructed. The ends of the molecule cannot be ligated although they contain 5' phosphates. At least 60% of the genome must encode proteins. In the sequenced portion, many open reading frames exist and these are tightly packed together. These have been examined for homology to published proteins but only 1 to 17 shows similarity to known proteins. F01 is therefore the prototype of a new phage family. On the basis of restriction sites, codon usage and the distribution of nonsense codons in the unused reading frames, a strong case can be made for natural selection that reacts to mRNA structure and function.  相似文献

Reliable methods are needed to detect total and pathogenic Vibrio parahaemolyticus. One marker of V. parahaemolyticus virulence is the thermostable-related hemolysin. We developed an alkaline phosphatase-labeled DNA probe method for the specific detection and enumeration of trh-positive V. parahaemolyticus by colony hybridization. The probe was tested against a panel of 200 bacterial strains and determined to be specific for trh-positive V. parahaemolyticus. Additionally, the trh alkaline phosphatase probe colony hybridization was successfully used to detect and enumerate trh-positive V. parahaemolyticus in seafood and water samples collected from the United States and the United Kingdom.  相似文献

The application of quantitative real-time PCR (qPCR) technologies for the rapid identification of fecal bacteria in environmental waters is being considered for use as a national water quality metric in the United States. The transition from research tool to a standardized protocol requires information on the reproducibility and sources of variation associated with qPCR methodology across laboratories. This study examines interlaboratory variability in the measurement of enterococci and Bacteroidales concentrations from standardized, spiked, and environmental sources of DNA using the Entero1a and GenBac3 qPCR methods, respectively. Comparisons are based on data generated from eight different research facilities. Special attention was placed on the influence of the DNA isolation step and effect of simplex and multiplex amplification approaches on interlaboratory variability. Results suggest that a crude lysate is sufficient for DNA isolation unless environmental samples contain substances that can inhibit qPCR amplification. No appreciable difference was observed between simplex and multiplex amplification approaches. Overall, interlaboratory variability levels remained low (<10% coefficient of variation) regardless of qPCR protocol.  相似文献

This study examined whether sociocontextual and sociocognitive variables explained the math/science goals of 409 Mexican American youth using a modified version of R. W. Lent, S. D. Brown, and G. Hackett's (1994) social cognitive career theory. Results from structural equation modeling indicated that the hypothesized model explained a significant amount of variance in math/science goals for both Mexican American girls and boys. Findings suggested that gender did not moderate relations among the variables in the hypothesized model. Results also suggested that most of the social cognitive career theory propositions tested were supported. Specifically, social class predicted math/science past performance accomplishments. Math/science past performance accomplishments and perceived parent support predicted math/science self-efficacy. Furthermore, math/science self-efficacy predicted math/science outcome expectations, and together with math/science interests, these sociocognitive variables predicted math/science interests and goals. Contrary to expectations, generation status, Anglo orientation, and Mexican orientation did not predict math/science past performance accomplishments, and past performance accomplishments did not predict math/science outcome expectations. Furthermore, Anglo orientation and perceived social support from parents, teachers, classmates, and a close friend did not predict math/science goals. This study provides insight into the mechanistic and performance features of the cyclic reduction of NOx by CO in the presence and absence of excess water on a Pt–Rh/Ba/Al2O3 NOx storage and reduction catalyst. At low temperatures (150–200 °C), CO is ineffective in reducing NOx due to self-inhibition while at temperatures exceeding 200 °C, CO effectively reduces NOx to main product N2 (selectivity >70 %) and byproduct N2O. The addition of H2O at these temperatures has a significant promoting effect on NOx conversion while leading to a slight drop in the CO conversion, indicating a more efficient and selective lean reduction process. The appearance of NH3 as a product is attributed either to isocyanate (NCO) hydrolysis and/or reduction of NOx by H2 formed by the water gas shift chemistry. After the switch from the rich to lean phase, second maxima are observed in the N2O and CO2 concentrations versus time, in addition to the maxima observed during the rich phase. These and other product evolution trends provide evidence for the involvement of NCOs as important intermediates, formed during the CO reduction of NO on the precious metal components, followed by their spillover to the storage component. The reversible storage of the NCOs on the Al2O3 and BaO and their reactivity appears to be an important pathway during cyclic operation on Pt–Rh/Ba/Al2O3 catalyst. In the absence of water the NCOs are not completely reacted away during the rich phase, which leads to their reaction with NO and O2 upon switching to the subsequent lean phase, as evidenced by the evolution of N2, N2O and CO2. In contrast, negligible product evolution is observed during the lean phase in the presence of water. This is consistent with a rapid hydrolysis of NCOs to NH3, which results in a deeper regeneration of the catalyst due in part to the reaction of the NH3 with stored NOx. The data reveal more efficient utilization of CO for reducing NOx in the presence of water which further underscores the NCO mechanism. Phenomenological pathways based on the data are proposed that describes the cyclic reduction of NOx by CO under dry and wet conditions.  相似文献

The H2/NO/O2 reaction under lean-burn conditions has been studied by means of in situ DRIFTS, reactor measurements and temperature-programmed desorption with the aim of understanding the very different behavior of Pd/TiO2 and Pd/Al2O3 catalysts. The former deliver very high NO x conversions (70-80%) with good N2 selectivity whereas the latter show very low activity. In addition, PdTiO2 exhibits two distinct NO x reduction pathways, thus greatly extending the useful temperature range. It is shown that the PdTiO2 low-temperature channel involves adsorption and subsequent dissociation of NO on reduced (Pd0) metal sites. The low activity of PdAl2O3 is a consequence of palladium remaining in an oxidized state under reaction conditions. The high-temperature NO reduction channel found with PdTiO2 is associated with the generation and subsequent reaction of NH x species.  相似文献

A significant increase in polymer crystallinity is reported in composites of carbon-nanotubes (CNT) and Poly(3-octylthiophene-2,5-diyl), P3HT and Poly(3-octylthiophene-2,5-diyl), P3OT; Differential scanning calorimetry (DSC) reveal an increase from about 40% crystallinity of the native P3HT to ∼62% in composites containing 25 wt% MWNT. A similar behavior is observed in P3OT with ∼68% crystallinity, a double crystallization peak and higher melting temperature than the native polymers. The effect is unique to CNT and is not induced by fullerenes or graphene layers. High-resolution transmission electron microscopy, (HRTEM) of CNT-polymer dispersions reveal chains stacked upon the CNT in an elongated, stretched conformation. Following a detailed molecular study by Bernardi et al. and the HRTEM observations the DSC results are attributed to a CNT-mediated entropic effect: due to their intrinsic, 1D cylindrical shape the CNT impose an increased conjugation length on chains adsorbed and stacked upon dispersed CNT. Crystallization thus commences from a heterogeneous mixture of native chains and chains with a longer persistence length (higher effective rigidity) and consequentially a lower effective height of the entropic barrier for crystallization. The findings offer a new insight into the origins of CNT-induced polymer nucleation.  相似文献
