The emission spectrum of TaO, excited in a tantalum hollow cathode lamp, has been observed at high resolution using a Fourier transform spectrometer. In addition to previously known transitions, a number of new bands have been identified and assigned as belonging to two new electronic transitions: H2Pi1/2-X2Delta3/2 and K2Phi5/2-X2Delta3/2. A rotational analysis ofthe 0-0 and 0-1 bands of the H2Pi1/2-X2Delta3/2 transition and of the 0-1, 1-2, 0-0, 1-0, and 2-1 bands of theK2Phi5/2-X2Delta3/2 transition has been carried out, providing the following equilibrium constants for the ground X2Delta3/2 state:omegae = 1028.9060(15) cm-1, omegaexe = 3.58928(66) cm-1, Be = 0.40289737(139) cm-1, alphae = 0.00185445(83) cm-1, andre = 1.6873430(29) ?. The principal molecular constants for the H2Pi1/2 state are T00 = 20 634.32758 (40) cm-1,B0 = 0.3766867(31) cm-1, and r0 = 1.7450604(72) ?, while the equilibrium constants for the K2Phi5/2 state areomegae = 905.4549(15) cm-1, omegaexe = 3.67601(64) cm-1, Be = 0.37965102(36) cm-1, alphae = 0.00189370(21) cm-1, andre = 1.7382343(8) ?. Although the H2Pi1/2 and K2Phi5/2 states have been observed previously in matrix isolation experiments, our work on these states is the first in the gas phase. Copyright 1998 Academic Press.  相似文献

Fluorescent imaging of Ca2+ and intracellular recordings were used to assess Ca2+ increases and voltage responses during light presentations in Hermissenda B photoreceptors. Ca2+ levels increased and were sustained during a relatively long exposure to light. Repeated presentations of a brief light induced an elevation of intracellular Ca2+ that persisted throughout short interlight intervals, but which dissipated during long interlight intervals. In all instances, the magnitude of the intracellular Ca2+ signal was inversely related to the amplitude of the light-induced generator potential. Blocking of voltage-dependent Ca2+ channels did not significantly affect the magnitude of the Ca2+ signal, suggesting that the intracellular Ca2+ response arises primarily from release from intracellular stores. These results indicate that Ca2+ plays an important role in the modulation of the voltage responses to light, acting to suppress the response during repetitive or prolonged stimulation.  相似文献

Phosphorylation and dephosphorylation of the retinoblastoma gene product (pRB) are recognized as necessary events in the cell cycle progression. To study the role of pRB in regulation of cell proliferation, the stable cell lines with constitutive expression of the exogenous RB gene can be employed. In order to obtain such cell lines in this work C3H10T1/2 mouse fibroblasts were infected with defective retrovirus encompassing the RB and Neo gene conferring resistance to geniticine (G418). The pRB production and its phosphorylation pattern were analyzed by immunoblotting in cell lysates considering well known data on correlation between pRB phosphorylation pattern and its electrophoretic mobility. Cell lines subjected to G418 selection with the following cloning procedure were identical to the control cells expressing beta-galactosidase, when compared for pRB production and phosphorylation in the cell cycle stages characterized by hyperphosphorylated pRB. However, cells of the experimental cell lines hypophosphorylated pRB much faster and accumulated much more underphosphorylated protein compared to the control cell lines. The doubling time of the cells was not affected either by changes in the pRB phosphorylation pattern or by its overproduction during separate cell cycle stages. These results suggest that maintaining of the physiological level of pRB phosphorylation in cycling cells is strictly controlled and is considered to be a more important condition of the cell cycle progression than pRB dephosphorylation.  相似文献

Models for the surface of cuticle cells in hair fibers consist of a monolayer of fatty acids covalently bound to the underlying protein membrane by thioester linkages. The most prominent of these fatty acids is 18-methyleicosanoic acid (C21a), the synthesis of which requires the oxidative decarboxylation of isoleucine. Maple syrup urine disease (MSUD) is caused by an inherited deficiency in the enzyme branched chain 2-oxo acid dehydrogenase, which leads to the accumulation of branched chain alpha-keto-acids derived from the amino acids, leucine, isoleucine, and valine. Transmission electron microscopy studies of developing hair fibers show a structural defect in the fiber shaft in hair from patients with MSUD. This defect is confined to the cuticle of the fiber, where the cuticle membrane directly apposes the intercellular material. Thus, the defect indicates that C21a is located exclusively on the upper surface of fiber cuticle cells. Lipid analysis of MSUD hairs has demonstrated significant changes in the relative abundance of the covalently bound fatty acids and an almost complete absence of C21a, whereas there was little difference in the amino acid composition compared with normal hair. These results provide further evidence for the existence of the surface lipid monolayer and its crucial role in cellular adhesion.  相似文献

Rett syndrome (RS) is a progressive encephalopathy restricted to the female sex. In the present study we investigated 30 females and one male with RS by cytogenetic and molecular-cytogenetic methods. We failed to identify any chromosomal rearrangements within the female groups and no correlation between fra(X)(p22) and RS in either the female group or the male. The boy with RS has karyotype 46,XY/47,XXY with abnormal cell clone (47,XXY) in 6-12% of his lymphocytes (revealed by fluorescence in situ hybridization analysis (FISH) of interphase cells with chromosome X-specific DNA probe). Our results indicated a possible connection between RS and X-chromosome replication disturbance. A late-replicating X-chromosome with a specific banding pattern (type 'C') has been observed in RS patients only. We propose to analyse the X-chromosome replication pattern as a test for confirmation of RS at preclinical diagnosis.  相似文献

The authors studied hormonal regulation of carbohydrate metabolism by secretion of insulin, C-peptide in 86 patients with chronic glomerulonephritis with different functional condition of the kidneys. There was a decrease in glucose tolerance, basal and reactive hyperinsulinemia, elevated level of C-peptide (relative insulin insufficiency). Mechanism of arising changes in the carbohydrate and insulin metabolism is complex and multicomponent. This includes renal lesion and consequent inhibition of hormone metabolism. Intensification of glomerular filtration is associated with inhibition of filtration of insulin and C-peptide derivants. Accumulation of nitrogen metabolism products results in changed response of pancreatic beta-cells to glucose. General disturbances of metabolism are accompanied by increasing levels of hormonal and nonhormonal contrainsular substances.  相似文献
