The inositol 1,4,5-triphosphate receptor type 1 (ITPR1) gene encodes an InsP3-gated calcium channel that modulates intracellular Ca2+ release and is particularly expressed in cerebellar Purkinje cells. Pathogenic variants in the ITPR1 gene are associated with different types of autosomal dominant spinocerebellar ataxia: SCA15 (adult onset), SCA29 (early-onset), and Gillespie syndrome. Cerebellar atrophy/hypoplasia is invariably detected, but a recognizable neuroradiological pattern has not been identified yet. With the aim of describing ITPR1-related neuroimaging findings, the brain MRI of 14 patients with ITPR1 variants (11 SCA29, 1 SCA15, and 2 Gillespie) were reviewed by expert neuroradiologists. To further evaluate the role of superior vermian and hemispheric cerebellar atrophy as a clue for the diagnosis of ITPR1-related conditions, the ITPR1 gene was sequenced in 5 patients with similar MRI pattern, detecting pathogenic variants in 4 of them. Considering the whole cohort, a distinctive neuroradiological pattern consisting in superior vermian and hemispheric cerebellar atrophy was identified in 83% patients with causative ITPR1 variants, suggesting this MRI finding could represent a hallmark for ITPR1-related disorders.  相似文献

The transient receptor potential ankyrin 1 (TRPA1), a member of the TRP superfamily of channels, is primarily localized in a subpopulation of primary sensory neurons of the trigeminal, vagal, and dorsal root ganglia, where its activation mediates neurogenic inflammatory responses. TRPA1 expression in resident tissue cells, inflammatory, and immune cells, through the indirect modulation of a large series of intracellular pathways, orchestrates a range of cellular processes, such as cytokine production, cell differentiation, and cytotoxicity. Therefore, the TRPA1 pathway has been proposed as a protective mechanism to detect and respond to harmful agents in various pathological conditions, including several inflammatory diseases. Specific attention has been paid to TRPA1 contribution to the transition of inflammation and immune responses from an early defensive response to a chronic pathological condition. In this view, TRPA1 antagonists may be regarded as beneficial tools for the treatment of inflammatory conditions.  相似文献

The fatty acid (FA) profile of oysters generally reflects the dietary FA composition. Moreover, incorporation of FA into tissues is modulated by various metabolic factors, and final composition will depend upon the dietary sources, cumulative intake, and oysters' development stage. Thus, the aim of this study was to assess the impact of dietary incorporation of seaweed (SW) Ulva rigida, in replacement of traditional microalgae diet, on the FA composition of Pacific oysters Crassostrea gigas, during broodstock conditioning. The dietary conditioning consisted of direct replacement of microalgae (33% Tisochrysis lutea, 50.25% Skeletonema costatum, and 16.75% Chaetoceros calcitrans) by SW at four different substitution levels (0%, 25%, 50%, and 100% diet). The dietary docosahexaenoic acid (DHA) (22:6n-3) and eicosapentaenoic acid (EPA) (20:5n-3) contents showed a positive correlation with the dietary microalgae level. During the trial, oysters fed with higher percentages of microalgae revealed a depletion of DHA and accumulation of EPA. The 100% SW caused a significant reduction in oxygen consumption and, consequently, in the standard metabolic rate. Based on these results, a partial substitution of up to 25% of dietary microalgae seems to be a suitable alternative, because it elicited similar results to the commercial 100% microalgae diet.  相似文献

The influence of regular air cold storage (7 °C and 85 ± 5% RH) followed by ripening at shelf-life conditions (19–21 °C and 65 ± 5% RH), on bioactive compounds of Hass avocados was investigated. Results showed that the content of mannoheptulose and perseitol decreased significantly already during cold storage and ripening period. The fatty acid profile and contents of tocopherols (α- and β-tocopherol) and phytosterols (β-sitosterol, stigmasterol, campesterol) remained unchanged from day 0 to edible ripeness. Total phenolics, hydrophilic and lipophilic antioxidant capacity remained unchanged during cold storage and increased during the ripening period. At edible ripeness, significant amounts of phenolic acids, p-coumaric and caffeic and their derivatives were synthesised. Our results demonstrated that regular air cold storage for up to 37 days followed by ripening at shelf-life conditions enhances the phenolic compounds and mainly the hydrophilic antioxidant capacity without affecting the remaining bioactive compounds in Hass avocado.  相似文献

The transient receptor potential ankyrin 1 (TRPA1), a member of the TRP superfamily of channels, acts as ‘polymodal cellular sensor’ on primary sensory neurons where it mediates the peripheral and central processing of pain, itch, and thermal sensation. However, the TRPA1 expression extends far beyond the sensory nerves. In recent years, much attention has been paid to its expression and function in non-neuronal cell types including skin cells, such as keratinocytes, melanocytes, mast cells, dendritic cells, and endothelial cells. TRPA1 seems critically involved in a series of physiological skin functions, including formation and maintenance of physico-chemical skin barriers, skin cells, and tissue growth and differentiation. TRPA1 appears to be implicated in mechanistic processes in various immunological inflammatory diseases and cancers of the skin, such as atopic and allergic contact dermatitis, psoriasis, bullous pemphigoid, cutaneous T-cell lymphoma, and melanoma. Here, we report recent findings on the implication of TRPA1 in skin physiology and pathophysiology. The potential use of TRPA1 antagonists in the treatment of inflammatory and immunological skin disorders will be also addressed.  相似文献

With the growing amount of multimedial content over the internet and broadcast systems, mechanisms for efficient information organization, manipulation and transmission are becoming indispensable. Optimization of the multimedia search and retrieval processes is nowadays an important area of development due to the difficulty to browse, filter and manage that big amount of data. The adoption of the MPEG-7 standard has a significant importance to simplify the image retrieval process. However, performance issues are still relevant when the retrieval must be accomplished in real time. This work presents an innovative and efficient approach of a Content-Based Retrieval Process using metric spaces implemented in heterogeneous resources according to the demand of computational power. Several implementations were made and comparative results are shown evidencing the benefits of the proposed approach.  相似文献

Response surface methodology (RSM) was used to optimise alkaline protein extraction from cañihua grain meal as to maximise protein extraction yield. Different factors (temperature, extraction time, solvent/meal ratio, pH and NaCl molar concentration) were screened, and their significant influence on the extracted protein yield was evaluated. The first four factors were selected and studied by RSM using a central composite design (CCD). The obtained model produced a satisfactory fitting of the results (R2 = 0.801). Optimal cañihua protein extraction conditions corresponded to a temperature of 21 °C, time extraction of 5 min, solvent/meal ratio of 37/1 (v/w) at pH 10 resulting in a protein yield of 80.4 ± 1.3%, which closely agree with the predicted value of 81.4%. Moreover, protein degradation was studied via differential scanning calorimetry (DSC) obtaining a denaturation temperature of 93.4 °C and an enthalpy value of 1.22 ± 0.05 J g?1. These results are interesting from a technological point to help in designing an optimal protein extraction process and cañihua food processing strategies.  相似文献

Quercetin is the main flavonol in the human diet, and the most commonly used in studies of biological activity. The major circulating forms of quercetin found in the human plasma after consumption of food containing distinct quercetin glycosides are glucuronides and sulfates. In this work quercetin glucuronides have been obtained from green beans (quercetin 3-glucuronide) and by enzymic synthesis (quercetin 4′-glucuronide) using a modification of the method described by Plumb et al. (Methods in polyphenol analysis, The Royal Society of Chemistry, Cambridge, pp 187, 2003) so as to improve the original low yields of that methodology. The method finally optimised got yields of 19% in the preparation of quercetin 4′-glucuronide, which allows its further isolation for their use in biological assays. In addition, quercetin 3′-glucuronide, 3-glucuronide and a diglucuronide were synthesised with lower yields. The compounds prepared have been employed to perform assays in order to obtain data for their identification by HPLC coupled to photodiode array detection and tandem mass spectrometry. It was observed that the analysis by HPLC–ESI/MS/MS could allow the identification of different quercetin glucuronides based on the presence of some minor key MS2 fragments.  相似文献

The identification and quantification of Glucosinolates (Gls) via UPLC® MS-MS/PDA/qToF in ten mashua cultivars was carried out. Gls total contents were within the 4.9–54.2 μmol g−1 dry matter range, of which 96–99% corresponded to glucoaubrietin. Other less abundant Gls were glucotropaeolin and tentatively two isomers of hydroxybenzyl Gls. Postharvest refrigeration and shade storage conditions increased the content of Gls up to day 6 in 39.7% and 51.5% respectively. Sun exposure increased the Gls content in 40% up to day 3 but from day 6 considerable losses were attained (92% at day 15). Low correlation levels (R2) between the Gls and myrosinase (MYR) activity of 0.57, 0.28 and 0.39 for the refrigeration, shade and sun exposure treatments were obtained. The cooking regimes tested, boiling, microwaving and baking totally inactivated MYR without affecting the Gls content.  相似文献
