In 2000, 3M Company, the primary global manufacturer, announced a phase-out of perfluorooctanesulfonyl fluoride (POSF, C8F17SO2F)-based materials after perfluorooctanesulfonate (PFOS, C8F17SO3-) was reported in human populations and wildlife. The purpose of this study was to determine whether PFOS and other polyfluoroalkyl concentrations in plasma samples, collected in 2006 from six American Red Cross adult blood donor centers, have declined compared to nonpaired serum samples from the same locations in 2000-2001. For each location, 100 samples were obtained evenly distributed by age (20-69 years) and sex. Analytes measured, using tandem mass spectrometry, were PFOS, perfluorooctanoate (PFOA), perfluorohexanesulfonate (PFHxS), perfluorobutanesulfonate (PFBS), N-methyl perfluorooctanesulfonamidoacetate (Me-PFOSA-AcOH), and N-ethyl perfluorooctanesulfonamidoacetate (Et-PFOSA-AcOH). The geometric mean plasma concentrations were for PFOS 14.5 ng/mL (95% CI 13.9-15.2), PFOA 3.4 ng/ mL (95% CI 3.3-3.6), and PFHxS 1.5 ng/mL (95% CI 1.4-1.6). The majority of PFBS, Me-PFOSA-AcOH, and Et-PFOSA-AcOH concentrations were less than the lower limit of quantitation. Age- and sex-adjusted geometric means were lower in 2006 (approximately 60% for PFOS, 25% for PFOA, and 30% for PFHxS) than those in 2000-2001. The declines for PFOS and PFHxS are consistent with their serum elimination half-lives and the time since the phase-out of POSF-based materials. The shorter serum elimination half-life for PFOA and its smaller percentage decline than PFOS suggests PFOA concentrations measured in the general population are unlikely to be solely attributed to POSF-based materials. Direct and indirect exposure sources of PFOA could include historic and ongoing electrochemical cell fluorination (ECF) of PFOA, telomer production of PFOA, fluorotelomer-based precursors, and other fluoropoly-mer production.  相似文献

The yeast Kluyveromyces marxianus produces a range of volatile molecules with applications as fragrances or flavours. The purpose of this study was to establish how nutritional conditions influence the production of these metabolites. Four strains were grown on synthetic media, using a variety of carbon and nitrogen sources and volatile metabolites analysed using gas chromatography–mass spectrometry (GC–MS). The nitrogen source had pronounced effects on metabolite production: levels of the fusel alcohols 2‐phenylethanol and isoamyl alcohol were highest when yeast extract was the nitrogen source, and ammonium had a strong repressing effect on production of 2‐phenylethyl acetate. In contrast, the nitrogen source did not affect production of isoamyl acetate or ethyl acetate, indicating that more than one alcohol acetyl transferase activity is present in K. marxianus. Production of all acetate esters was low when cells were growing on lactose (as opposed to glucose or fructose), with a lower intracellular pool of acetyl CoA being one explanation for this observation. Bioinformatic and phylogenetic analysis of the known yeast alcohol acetyl transferases ATF1 and ATF2 suggests that the ancestral protein Atf2p may not be involved in synthesis of volatile acetate esters in K. marxianus, and raises interesting questions as to what other genes encode this activity in non‐Saccharomyces yeasts. Identification of all the genes involved in ester synthesis will be important for development of the K. marxianus platform for flavour and fragrance production. Hypotheses on the role of gluten in bread staling range from gluten having an anti-firming effect, or no effect on firming, to gluten-starch interactions being essential for bread firming. To test these hypotheses, the firming rate of starch bread made from protein-free synthetic flour was compared with that of starch-gluten breads made from synthetic flours containing 1–15% gluten (Fig. 1). Only loaves of similar specific loaf volume and crumb moisture content were compared to eliminate these parameters as variables that might influence firming rate. The starch breads clearly increased in firmness up to six days, indicating that gluten was not essential to the firming process, starch alone causing bread to firm with time. The starch-10% gluten breads and starch-15% gluten breads had very similar specific loaf volumes, moisture contents and firming rates to that of the starch breads. This indicates that protein possibly has some role in firming, because if only starch has a role in firming then adding gluten would effectively dilute the starch and reduce the rate of firming. We propose that increasing bread firmness results from glucan chains of partially leached amylose and amylo-pectin attached to swollen starch granules forming hydrogen bonds with other starch granules and, to a smaller extent, with gluten fibrils.  相似文献

Prior to puberty, elevated nutrient intake has been shown to negatively affect prepubertal mammary development in the heifer. The objective of this study was to evaluate the effects of increased nutrient intake on mammary development in Holstein heifers at multiple body weights from birth through puberty. Specifically, this study evaluated the effects of nutrient intake and body weight at harvest on 1) total weight and DNA content of the parenchyma (PAR) and mammary fat pad (MFP) and 2) PAR and MFP composition. Starting at 45 kg of body weight, heifers (n = 78) were assigned to either a restricted (R) or elevated (E) level of nutrient intake supporting 650 (R) or 950 (E) g/d of body weight gain. Heifers were harvested at 50-kg increments from 100 to 350 kg of body weight. Mammary fat pad weight and DNA content were greater in E- than in R-heifers. Additionally, E-heifers had a greater fraction of lipids and a smaller fraction of protein in their MFP than did R-heifers. Parenchyma weight and DNA were lower in E- than in R-heifers; however, when analyzed with age as a covariate term, treatment was no longer a significant term in the model. Level of nutrient intake had no effect on the lipid, protein, or hydroxyproline composition of the PAR. Collectively, these data demonstrate that PAR is refractory to the level of nutrient intake whereas MFP is not. Furthermore, the covariate analysis demonstrated that age at harvest, not the level of nutrient intake, was the single greatest determinant of total PAR DNA content.  相似文献

IPTV, an emerging Internet application, would revolutionize the entertainment and media industries; however, IPTV also has the potential to overwhelm the Internet backbone and access networks with traffic. To date, IPTV over P2P streaming networks has advanced significantly using two different approaches: tree-push versus mesh-pull. In particular, the mesh-pull streaming approach has achieved a number of successful commercial deployments. In this article, we examine the current progress in the research and development of mesh-pull P2P streaming systems. We provide an overview of the general mesh-pull streaming architecture and review various challenges, design issues, and interesting research problems in this approach. We discuss the construction costs for providing an IPTV service with service guarantees. We outline a measurement technique for monitoring the video playback quality of mesh-pull streaming systems. We emphasize that the future P2P IPTV systems should be designed to meet the expectations of users for quality-of-experience. We also identify a few other important issues for IPTV over P2P streaming networks, including the traffic pressure on ISPs, various security concerns, and the necessity to re-examine the most appropriate P2P architecture. Insights obtained in this study will be valuable for the development and deployment of future P2P IPTV systems.  相似文献

The influence of different promoters (CeO2, MnOx, K2CO3) on various properties of a standard coprecipitated Cu/ZnO/Al2O3 catalyst has been examined. The catalysts prepared were characterized by Cu surface area, PTIR and TPR measurements. It was found that addition of K2CO3 reduced the Cu surface area by about 30%, whereas the Cu surface area did not decrease with addition of the other promoters. The reduction behaviour was affected by the addition of K2CO3 as well as by MnOx, but not by CeO2. The cause of these effects is possibly an electronic interaction between the promoter and Cu ions.The effect of the different promoters on the activity and selectivity has also been studied. The K2CO3 promoted catalyst has an optimum selectivity to higher alcohols at 280 °C; addition of Mn made the catalyst more selective towards methanol. At 300 °C, the Ce promoted catalyst had a high selectivity to methanol and iso-butanol. The promoting effect of the additives may be caused by stabilization of the surface intermediates leading to alcohols. Infrared measurements of adsorbed CO or adsorbed methanol on materials with and without K did not, however, provide any evidence for a difference in reaction mechanism.  相似文献

The efficacy of bacteriophages e11/2 and e4/1c as potential biocontrol agents for Escherichia coli O157:H7 in food applications was assessed under conditions relevant to the food chain environment. The stability of each phage was determined following exposure to varying environmental conditions (pH, temperature, water activity, and sodium chloride) and the ability of each phage to infect and reduce E. coli O157:H7 numbers under selected conditions was also examined. Both e11/2 and e4/1c significantly (p < 0.05) reduced numbers of E. coli O157:H7 when exposed to pH values ranging from pH > 4 to pH 9, temperatures from 4 °C to 37 °C, water activity values of 0.87 or 0.91 to 1.00 and NaCl concentrations of 1% to 2.5%. Subsequently, a cocktail of both phages was used (e11/2 and e4/1c) to assess reduction of E. coli O157:H7 on cattle hide pieces. This involved inoculating pieces of hide (20 × 20 cm) with E. coli O157:H7 (approximately 106 cfu/cm2) which were subsequently treated with either a suspension of a phage cocktail, consisting of e11/2 and e4/1c (multiplicity of infection of 1000 and 10,000, respectively) or water or not treated. Two different investigations were carried out; immediately or 1 h after treatment application was performed in different experiments. Swab samples taken immediately after phage treatment showed no significant (p > 0.05) reduction of E. coli O157:H7 numbers compared to the water treated or untreated samples. However, an extended exposure time of 1 h following phage application revealed a significant reduction (p < 0.05) (1.5 log10 cfu/cm2 reduction) in E. coli O157:H7 numbers compared to the numbers recovered on samples treated with water only. These findings demonstrate the potential use of e11/2 and e4/1c phages as a biocontrol agent for E. coli O157:H7 within various stages of the food chain, including on cattle hide.  相似文献
