Cell division cycle 25A (Cdc25A) is a dual-specificity phosphatase that is overexpressed in several cancer cells and promotes tumorigenesis. In normal cells, Cdc25A expression is regulated tightly, but the changes in expression patterns in cancer cells that lead to tumorigenesis are unknown. In this study, we showed that ubiquitin-specific protease 29 (USP29) stabilized Cdc25A protein expression in cancer cell lines by protecting it from ubiquitin-mediated proteasomal degradation. The presence of USP29 effectively blocked polyubiquitination of Cdc25A and extended its half-life. CRISPR-Cas9-mediated knockdown of USP29 in HeLa cells resulted in cell cycle arrest at the G0/G1 phase. We also showed that USP29 knockdown hampered Cdc25A-mediated cell proliferation, migration, and invasion of cancer cells in vitro. Moreover, NSG nude mice transplanted with USP29-depleted cells significantly reduced the size of the tumors, whereas the reconstitution of Cdc25A in USP29-depleted cells significantly increased the tumor size. Altogether, our results implied that USP29 promoted cell cycle progression and oncogenic transformation by regulating protein turnover of Cdc25A.  相似文献

Nanosized gold particles (27 +/− 3 nm) have been proven to be effective in ameliorating the symptoms of mycobacterial-, collagen- and pristane-induced arthritis in rat models. This contrasts with the drug sodium aurothiomalate that was only effective against mycobacterial-induced arthritis but not to the same extent as Au0. Gold in the traditional Indian Ayurvedic medicine,Swarna bhasma (gold ash), has been characterized as globular particles of gold with an average size of 56–57 nm.  相似文献

A 25 nm thick α-alumina layer was deposited on a turbine-grade silicon nitride by sol-gel dip coating and subsequent heat treatment in air at 1200°C. This layer had a nanometer grain structure. Silicon nitride protected by this thin layer showed a significant improvement in oxidation resistance over its uncoated counterpart after 200 cyclic exposures in air at 1250°C. The oxide layer grown on the coated silicon nitride also exhibited superior surface morphology, compared with the uncoated silicon nitride.  相似文献

Polymethylphenylsilsesquioxane (PMPSQ–OH) and trimethylsilyl end‐blocked PMPSQ (PMPSQ–EC) were prepared. The thermal decomposition behavior of these polymers was studied by thermogravimetric analysis (TGA) and FT‐Raman spectroscopy. Hydroxyl‐functionalized polystyrene (PS–OH) was also prepared by anionic living polymerization. Thin hybrid films of PMPSQ/PS–OH with various blend ratios were obtained by spin‐coating on freshly cleaned glass. The surface morphology of the hybrid films was investigated by atomic force microscopy (AFM). In 80/20 PMPSQ/PS–OH hybrid film, the PS–OH component produced a very uniformly dispersed phase. This hybrid film contained small domains of PS–OH whose size ranged from 60 to 80 nm. As the content of PS–OH was increased, the domain morphology coarsened and phase inversion took place around 50 wt %. In the phase‐inversed system, the PMPSQ‐rich phase was uniformly distributed in the PS–OH‐rich continuous phase. In addition, temperature‐dependent dielectric properties of PMPSQ/PS–OH hybrids were investigated. Relaxation of the hybrids was observed with an increasing content of the PS–OH component due to the amorphous glass transition behavior of PS–OH. © 2003 Wiley Periodicals, Inc. Process planning is a very important function in the modern manufacturing system. It impacts the efficiency of the manufacturing system greatly. The process planning problem has been proved to be a NP-hard problem. The traditional algorithms cannot solve this problem very well. Therefore, due to the intractability and importance of process planning problem, it is very necessary to develop efficiency algorithms which can obtain a good process plan with minimal global machining cost in reasonable time. In this paper, a new method based on honey bees mating optimization (HBMO) algorithm is proposed to optimize the process planning problem. With respect to the characteristics of process planning problem, the solution encoding, crossover operator, local search strategies have been developed. To evaluate the performance of the proposed algorithm, three experiments have been carried out, and the comparisons among HBMO and some other existing algorithms are also presented. The results demonstrate that the HBMO algorithm has achieved satisfactory improvement.  相似文献

Summary The surface modification of low-density polyethylene(PE) by liquid phase photograft polymerization with acrylic acid(AA), acrylamide(AM) and glycidyl methacrylate(GMA) was described. The grafting of AA and AM was proved and characterized by electron spectroscopy for chemical analysis(ESCA). It was found that fully hydrophilic surface can be obtained in very short irradiation time. With ESCA and attenuated total reflection infrared spectroscopy(ATR-IR), it can be confirmed that bifunctional monomer GMA was grafted onto the PE film surface. Through further reaction with GMA grafted film, heparin and protamine were immobilized onto the grafted film surface.  相似文献

Polyelectrolyte complexes (PECs) composed of chitosan and hyaluronic acid (HA) were prepared in various pH regions and at different weight ratios. At low pHs, there was a strong ionic interaction between NH groups in chitosan and both COO? and COOH groups in HA due to the deprotonation of HA, whereas weak linkages were formed at high pHs because only the carboxyl groups of HA could interact with NH groups in chitosan. The formation of PECs resulted in a decrease in the crystallinity and thermal stability caused by the interactions between polyions. With variations in the degree of ionization of polyions at various pH conditions, novel PEC sponges were prepared by the freeze drying of PEC solutions. Furthermore, for the evaluation of the wound‐healing effect of PEC sponges with or without an antimicrobial agent (silver sulfadiazine), they were applied to a full‐skin defect of a Wistar rat in vivo. The histology and computerized morphometric analysis of the epidermal healing confirmed the proliferation of fibroblasts in the wound bed and a distinct reduction in infectious agents. © 2003 Wiley Periodicals, Inc. An interpenetrating polymer network (IPN) hydrogel composed of chitosan and polyallylamine exhibited electric‐sensitive behavior. The chitosan/polyallylamine IPN hydrogel was synthesized by radical polymerization using 2,2‐dimethoxy‐2‐phenylacetophenone (DMPAP) and methylene bisacrylicamide (MBAAm) as initiator and crosslinker, respectively. The swelling behavior of the IPN was studied by immersion of the gel samples in aqueous NaCl solutions at various concentrations and pHs. The swelling ratio decreased with increasing concentration and pH of electrolyte solution. The stimuli response of the IPN hydrogel in electric fields was also investigated. When a swollen the IPN was placed between a pair of electrodes, the IPN exhibited bending behavior in response to the applied electric field. The IPN also showed stepwise bending behavior depending on the electric stimulus. In addition, thermal properties of the IPN were investigated by differential scanning calorimetry (DSC) and dielectric analysis (DEA). © 2002 Wiley Periodicals, Inc.
