Characteristics of the growth kinetics of Escherichia coli cells in pouched mashed potatoes under various conditions were studied with a mathematical model. Bacterial cells were inoculated in sterile mashed potatoes and then sealed in vinyl pouches, in which a very small amount of air was included. The growth curves of cells in the pouched mashed potatoes at constant temperature (12-34 degrees C) were sigmoidal with time on a semi-logarithmic plot and were successfully described with a new logistic model recently developed by us. The rate constant of growth showed a highly linear relationship to the temperature with the square-root model, and the lag period was longer at lower temperatures. The growth curve in glass tubes containing a large volume of air was similar to that in pouches, showing that the rate of growth was not affected by the volume of the surrounding air. The growth curves in pouched mashed potatoes were very similar to those in nutrient broth or on the surface of nutrient agar, which we previously reported. These results suggested that the growth kinetics of the bacterial cells under various conditions of rich nutrition might be almost identical, and can be described with a simple growth model like ours.  相似文献

Experimental results on vertical manipulation on an insulator surface using non-contact atomic force microscopy are presented. Cleaved ionic KCl(100) single crystal is used as an insulator surface. With the nanoindentation method used, the vertical manipulation of a single atom in an ionic crystal surface is more difficult than in a semiconductor surface. Therefore, in many cases, more than one surface atom is manipulated while, in rare cases, single-atom manipulation is successfully performed. Lateral manipulation of a vacancy has occasionally succeeded on the KCl(100) surface. We have presumed that the lateral manipulation was induced by pulling.  相似文献

Previous studies showed that water-free, premixed calcium phosphate cements (Pre-CPCs) exhibited longer hardening times and lower strengths than conventional CPCs, but were stable in the package. The materials hardened only after being delivered to a wet environment and formed hydroxyapatite as the only product. Pre-CPCs also demonstrated good washout resistance and excellent biocompatibility when implanted in subcutaneous tissues in rats. The present study evaluated characteristics of Pre-CPCs when implanted in subcutaneous tissues (Study I) and used for repairing surgically created two-wall periodontal defects (Study II). Pre-CPC pastes were prepared by combining CPC powders that consisted of CPC-1: Ca(4)(PO(4))(2)O and CaHPO(4), CPC-2: α-Ca(3)(PO(4))(2) and CaCO(3) or CPC-3: DCPA and Ca(OH)(2) with a glycerol at powder-to-liquid mass ratios of 3.5, 2.5, and 2.5, respectively. In each cement mixture, the Ca to P molar ratio was 1.67. The glycerol contained Na(2)HPO(4) (30 mass %) and hydroxypropyl methylcellulose (0.55 %) to accelerate cement hardening and improve washout resistance, respectively. In Study I, the test materials were implanted subcutaneously in rats. Four weeks after the operation, the animals were sacrificed and histopathological observations were performed. The results showed that all of the implanted materials exhibited very slight or negligible inflammatory reactions in tissues contacted with the implants. In Study II, the mandibular premolar teeth of mature beagle dogs were extracted. One month later, two-wall periodontal bone defects were surgically created adjacent to the teeth of the mandibular bone. The defects were filled with the Pre-CPC pastes and the flaps replaced in the preoperative position. The dogs were sacrificed at 1, 3 and 6 months after surgery and sections of filled defects resected. Results showed that one month after surgery, the implanted Pre-CPC-1 paste was partially replaced by bone and was converted to bone at 6 months. The pockets filled with Pre-CPC-2 were completely covered by newly formed bone in 1 month. The Pre-CPC-2 was partially replaced by trabecular bone in 1 month and was completely replaced by bone in 6 months. Examination of 1 month and 3 month samples indicated that Pre-CPC-2 resorbed and was replaced by bone more rapidly than Pre-CPC 1. Both Pre-CPC pastes were highly osteoconductive. When implanted in periodontal defects, Pre-CPC-2 was replaced by bone more rapidly than Pre-CPC-1.  相似文献

The effects of nitrogen source on arachidonic acid (AA) production and morphological changes during the culture of Mortierella alpina were investigated using an image analysis system. When yeast extract, gluten meal, or corn steep liquor was used, a circular pellet morphology was obtained. However, when Pharmamedia, fish meal, or soybean meal was used, M. alpina formed radial filamentous mycelia. The radial filamentous area in the case of soybean meal was 75% of the total mycelial area. In a jar fermentor culture, M. alpina morphology varied with the cultivation period: (i) at 0-6 h culture, the inoculated pellet-like mycelia were adapted to the new environment, (ii) at 6 h-1 d culture, filamentous mycelia grew exponentially which yielded a feather-like morphology, (iii) at 1-2 d culture, the filamentous mycelia became disentangled as a result of the mechanical agitation; consequently, the proportion of filamentous mycelia was increased, (iv) at 2-4 d culture, mycelia showed stationary growth, but the AA concentration increased rapidly, and (v) at 4-6 d culture, hyphae grew thick radially with the AA concentration continuing to increase gradually. In the case of the cultures with feather-like morphology obtained using soybean meal, the AA yield was 0.14 g/g dry cell weight, which was two times higher than that in cultures grown using yeast extract. These results suggest that the feather-like morphology of culture of M. alpina is suitable for AA production.  相似文献

Micro-mesoporous bimodal carbon nanospheres with high surface areas were synthesized by a combined use of surfactant templating technique and BaO2 chemical activation one. Starting spherical nanopolymer/surfactant composites were prepared by the NaOH-catalyzed reaction of resorcinol (R) and formaldehyde (F) in the presence of cetyltrimethylammonium bromide (CTAB) as a core template and 1,3,5-trimethylbenzene (TMB) and tert-butanol (t-BuOH) as cosurfactants. After pretreatment with hydrochloric acid, the composite materials were calcined at 1000 °C in N2 coexistent with varying weight ratios of BaO2 to RF polymer ranging from 0 to 11. It produced micro-mesoporous bimodal carbon nanospheres of 124–143 nm diameter, with specific surface areas as high as 1884 m2 g−1 or up to 3301 m2 g−1, in contrast to microporous ones with smaller surface areas obtained at low BaO2-loadings. The electrochemical double layer capacitance of the resulting nanocarbons in 0.5 M H2SO4 showed a marked increase with specific surface areas, up to as high as 219 F g−1 for the highest surface area carbon material.  相似文献

Nanoporous gold (NPG), made by dealloying low carat gold alloys, is a relatively new nanomaterial finding application in catalysis, sensing, and as a support for biomolecules. NPG has attracted considerable interest due to its open bicontinuous structure, high surface-to-volume ratio, tunable porosity, chemical stability and biocompatibility. NPG also has the attractive feature of being able to be modified by self-assembled monolayers. Here we use scanning electron microscopy (SEM) and atomic force microscopy (AFM) to characterize a highly efficient approach for protein immobilization on NPG using N-hydroxysuccinimide (NHS) ester functionalized self-assembled monolayers on NPG with pore sizes in the range of tens of nanometres. Comparison of coupling under static versus flow conditions suggests that BSA (Bovine Serum Albumin) and IgG (Immunoglobulin G) can only be immobilized onto the interior surfaces of free standing NPG monoliths with good coverage under flow conditions. AFM is used to examine protein coverage on both the exterior and interior of protein modified NPG. Access to the interior surface of NPG for AFM imaging is achieved using a special procedure for cleaving NPG. AFM is also used to examine BSA immobilized on rough gold surfaces as a comparative study. In principle, the general approach described should be applicable to many enzymes, proteins and protein complexes since both pore sizes and functional groups present on the NPG surfaces are controllable.  相似文献

X-ray diffraction analysis was used to study the structure of Fe97?x Zr3N x thin films produced under different conditions of rf magnetron sputtering (on rotary and stationary substrates) and subsequent annealing. Either a single-phase structure, namely, a supersaturated bcc α-(FeN) solid solution, or a mixed structure that consists of two crystalline phases, such as a supersaturated bcc α-Fe(N) solid solution and an fcc Fe4N nitride, and a Fe-based amorphous phase were shown to be formed in films deposited on either stationary or rotating substrate, respectively. As the annealing temperature increases, the content of nitrogen in the bcc phase decreases; the grain size of the bcc phase in the films deposited on rotating and stationary substrates increases from 8 to 14 nm and from 10 to 30 nm, respectively; the amount of the Fe4N phase increases. The results of the study of the magnetic properties of the films are reported. The maximum saturation induction B s reached is 1.8–1.9 T; the minimum coercive force H c is 1 Oe. The optimum combination of the magnetic properties (B s = 1.8–1.9 T and H c = 1.5–1.8 Oe) is observed for the films deposited on the rotating substrate and subsequently annealed at 400°C.  相似文献

A snake, which has a simple shape without limbs, can move over rough terrain, climb a wall or tree, swim, and glide through the air. It is difficult to understand the gliding flight of a snake in aerodynamics assuming a static model, because a snake undulates its body trunk and turns in air. This paper presents an experimental system for investigation of the relationship between the shape of the gliding snake model and gliding characteristics. Results of a gliding experiment confirm a sufficiently high reproducibility of the experiment.  相似文献

Presented in this paper are the results of a series of column experiments and a pilot scale test conducted to develop the treatment system of freshwater and wastewater by their percolation through soil and the other materials at the rate of a few metres per day. Materials that had high ability to sorb fulvic acid (FA hereafter) and phosphate ion were selected based on the results of the batch sorption tests and were processed into beads before subjecting them to the column test. The results of the column experiment suggested that the reagents added to increase the strength of the beads reduced the sorption of FA, thus causing the early breakthrough of the FA injected into the column. Less effect from the bead processing was observed for the removal of phosphate ion. Pilot scale test showed that the removal of particulate fraction of phosphorus differed among the materials used. The bead processing technique that can ensure the high water permeability while maintaining the sorption capacity of the material was required for further development of the treatment system.  相似文献

A fuzzy self-tuning parallel genetic algorithm for optimization   总被引:1,自引:0,他引:1  
The genetic algorithm (GA) is now a very popular tool for solving optimization problems. Each operator has its special approach route to a solution. For example, a GA using crossover as its major operator arrives at solutions depending on its initial conditions. In other words, a GA with multiple operators should be more robust in global search. However, a multiple operator GA needs a large population size thus taking a huge time for evaluation. We therefore apply fuzzy reasoning to give effective operators more opportunity to search while keeping the overall population size constant. We propose a fuzzy self-tuning parallel genetic algorithm (FPGA) for optimization problems. In our test case FPGA there are four operators—crossover, mutation, sub-exchange, and sub-copy. These operators are modified using the eugenic concept under the assumption that the individuals with higher fitness values have a higher probability of breeding new better individuals. All operators are executed in each generation through parallel processing, but the populations of these operators are decided by fuzzy reasoning. The fuzzy reasoning senses the contributions of these operators, and then decides their population sizes. The contribution of each operator is defined as an accumulative increment of fitness value due to each operator's success in searching. We make the assumption that the operators that give higher contribution are more suitable for the typical optimization problem. The fuzzy reasoning is built under this concept and adjusts the population sizes in each generation. As a test case, a FPGA is applied to the optimization of the fuzzy rule set for a model reference adaptive control system. The simulation results show that the FPGA is better at finding optimal solutions than a traditional GA.  相似文献
