Klebsiella pneumoniae 40bXX, a mutant strain that constitutively produces D-arabinose isomerase (D-AI), was isolated through a series of repeated subcultures from the parent strain on a mineral salt medium supplemented with L-Xylose as the sole carbon source. D-AI could be efficiently immobilized on chitopearl beads. The optimum temperature for the activity of the immobilized enzyme was 40 degrees C and the enzyme was stable up to 50 degrees C. The D-Al was active at pH 10.0 and was stable in the range of pH 6.0-11.0. The enzyme required manganese ions for maximum activity. Three immobilized enzymes, D-xylose isomerase (D-XI), D-tagatose 3-epimerase (D-TE and D-AI were used for the preparation of D-arabinose from D-xylose in a coupling reaction. After completion of the reaction, degradation of D-xylulose was carried out by Saccharomyces cerevisiae. The reaction mixture containing D-Xylose, D-ribulose and the product was then separated by ion exchange column chromatography. After crystallization, the product was checked by HPLC, IR spectroscopy, NMR spectroscopy and optical rotation measurements. Finally, 2.0 g of D-arabinose could be obtained from 5 g of the substrate.  相似文献

Utilization of renewable and alternative energy feedstocks such as nonedible seeds oil to deal with the increasing energy crises and related ecological concerns have gained the attention of researchers. Biodiesel is an efficient and renewable substitute for diesel engine. This work investigates the potential of inexpensive nonedible seed oil of Linum usitatissimum to synthesize biodiesel using iron sulfate green nanocatalyst through the process of transesterification. Flax seed contains about 37.5% oil content estimated through Soxhlet apparatus. Light microscopy revealed that seed size varies from 3.0 to 6.0 cm in length, 2.0 to 3.3 cm in width, and 0.7 to 1.0 mm in diameter. Color of seed varied from yellow to brown. Characterization of biodiesel is performed through GC–MS and FTIR. Scanning electron microscopy was carried out to study the morphological features of seed coat. Catalyst was characterized by scanning electron microscopy, energy diffraction X-ray, and X-ray diffraction. The diffraction peaks of Fe3O4 green nanoparticles were found to be in 2θ values, 30.24°, 35.62°, 38.26°, 49.56°, 57.12°, and 62.78°. Fuel properties of biodiesel are also determined and compared with ASTM standards. Linum usitatissimum biodiesel has density 0.8722 (15°C kg/L), kinetic viscosity 5.45 (40°C cSt), flash point (90°C), pour point (−13°C), cloud point (−9°C), sulfur (0.0432% wt), and total acid number (0.245 mg KOH/g). It is concluded that L. usitatissimum seed oil is a highly potential source for biodiesel production to cope with the challenge of present energy demand.  相似文献

Environmentally friendly waterborne polyurethane (WPU) coatings are used extensively due to their low VOCs emission than solvent based PU coatings. Additionally, WPU coatings have low temperature flexibility, pH stability, water resistance, superior solvent resistance, outstanding weathering resistance and desirable chemical and mechanical properties. This review provides an overview on the recent developments of WPU coatings and their value added applications in the coatings and paint industry. UV-cured WPU coatings provide an important class of green and ecofriendly coatings with outstanding mechanical properties and rapid curing system. Hyper-branched polyurethanes (PUs) show interesting properties, such as high solubility, reactivity and good rheological behavior owing to multiple end groups, compact molecular structure and diminishing chain entanglement. Inherently, WPU coatings have reduced stiffness and mechanical strength that can be increased by the addition of nanoparticles, like Ag, Cu, TiO2, SiO2 and many more. Fire retardants, commonly phosphorous, are incorporated in the WPU structure to increase the flame retardancy of WPU coatings.  相似文献

A total of 125 (ready to eat) processed food samples (70 intended for infant and 55 for adult intake) belonging to 20 different food categories were analyzed for aflatoxins contamination using Reverse Phase High Performance Liquid Chromatography (RP-HPLC) with fluorescent detection. A solvent mixture of acetonitrile-water was used for the extraction followed by immunoaffinity clean-up to enhance sensitivity of the method. The limit of detection (LOD) (0.01-0.02 ng·g(-1)) and limit of quantification (LOQ) (0.02 ng·g(-1)) was established for aflatoxins based on signal to noise ratio of 3:1 and 10:1, respectively. Of the processed food samples tested, 38% were contaminated with four types of aflatoxins, i.e., AFB1 (0.02-1.24 μg·kg(-1)), AFB2 (0.02-0.37 μg·kg(-1)), AFG1 (0.25-2.7 μg·kg(-1)) and AFG2 (0.21-1.3 μg·kg(-1)). In addition, the results showed that 21% of the processed foods intended for infants contained AFB1 levels higher than the European Union permissible limits (0.1 μg·kg(-1)), while all of those intended for adult consumption had aflatoxin contamination levels within the permitted limits.  相似文献

The amount of phenolics, flavonoids, and anthocyanins in ethanol extracts and antioxidant activity of both ethanol and lipophilic extracts of common fruity vegetables in Bangladesh were studied. Among the ethanol extracts of 15 fruity vegetables, M. oleifera had the highest total polyphenol content (85.05 mg gallic acid equivalent/g extract) followed by L. acutangula (61.74 mg gallic acid equivalent/g extract) and A. esculentus (48.92 mg gallic acid equivalent/g extract). L. acutangula had high a content of flavonoids (14.46 mg (+)-catechin equivalent/g extract), which was almost similar to L. siceraria (13.67 mg catechin equivalent/g extract) followed by A. esculentus (11.95 mg catechin equivalent/g extract) and S. melongena (11.42 mg catechin equivalent/g extract). Highest anthocyanins content was in F. hispida (2.22 μmol/g extract) followed by S. melongena (1.04 μmol/g extract). Ethanol extracts of A. esculentus, F. hispida, L. acutangula, L. siceraria, and S. melongena exhibited high DPPH free radical scavenging activity with IC50 of 70.4, 64.9, 70.4, 64.9, and 94.3 μg/mL respectively, whereas for the same lipophilic extracts of F. hispida and S. melongena showed lowest (37 μg/mL) IC50 followed by M. oleifera (47.6 μg/mL), L. siceraria (57.5 μg/mL), and A. esculentus (63.3 μg/mL). These vegetables also showed high reducing powers, NO scavenging and total antioxidant capacity. Therefore, the top five potential fruity vegetables consist of both hydrophilic and lipophilic antioxidant(s), the order being F. hispida > M. oleifera > A. esculentus, L. acutangula > L. siceraria > and S. melongena.  相似文献

Bioactive polyphenols, cartenoids, and anthocyanins present in fruits and vegetables are receiving much attention because of their potential antioxidant activity. This study was conducted to determine antioxidant activity of leaves, peels, stem bark, and kernel of mango varieties langra and chonsa. Total phenolic (TPC) and total flavonoid contents (TFCs) in segments of langra ranged from 63.89 to 116.80 mg GAE/g DW and 45.56 to 90.89 mg CE/g DW, respectively, and that of chonsa were 69.24 to 122.60 mg GAE/g DW and 48.43 to 92.55 mg CE/g DW, respectively. The 2, 2-diphenyl-1-picrylhydrazyl (DPPH) scavenging activity and linoleic inhibition capacity in segments of langra ranged from 53.30% to 61.10% and 40.0% to 47.20%, respectively, whereas for chonsa; 56.40% to 66.0% and 48.1% to 49.0%, respectively. The reducing potentials of different segments of langra and chonsa at concentration of 10 mg/mL were 0.512 to 0.850 and 0.595 to 0.665 mV, respectively. Comparison between both varieties showed chonsa exhibited better antioxidant activity. The apparent average molar masses (Mw,app), apparent average radii of gyration (Rg,app), of native sago starch and fractions were determined using asymmetrical flow field‐flow fractionation coupled with multi‐angle light scattering and refractive index detectors (AF4/MALS/RI). Amylose‐type (Fraction A) and amylopectin‐type (Fraction B) were chemically separated from native sago starch. Native sago starch and Fractions (A–B) were dissolved in 1M KSCN using a high pressure microwave vessel. The effect of varying cross flow rates at a fixed channel flow rate upon the Mw,app and Rg,app distributions of native sago starch and Fractions (A–B) were investigated. The average Mw,app values of native sago starch, Fraction (A) and Fraction (B) were 60 × 106, 1.5 × 106 and 60 × 106 g/mol, respectively, with average Rg,app values of 142, 75 and 127 nm, respectively. The sphere‐equivalent hydrodynamic radii (Rh) values for native sago starch and fractions were determined from AF4 experimental parameters.  相似文献

The present work describes the trend of linear polyurethane elastomers for its thermal and mechanical behavior. The linear polyurethane elastomers have been synthesized in two different series by changing the composition of the polymers. Both series have common macrodiol of hydroxyl-terminated polybutadiene and butanediol while they vary in the presence of diisocyanate. One of the series, contain an aromatic diisocyanate, toluene diisocyanate (TDI), and the other consist of 1,6-hexamethylene diisocyanate (HDI) as an aliphatic diisocyanate. For linear structure, ratio of OH and NCO group has been maintained 1.0. The hydrogen bonding index is determined from Fourier transform infrared to calculate the extent of interchain hydrogen bonding which manifest the microphase separation. Young's modulus (5.79 MPa) and tensile strength (1.14 MPa) are recorded for HDI-derived elastomers, while Young's modulus varies from 1.14 to 1.63 MPa for TDI-based elastomers which also resulted in higher thermal stability. Thermogravimetric curves exhibit two-step degradation with the stability of 300 °C. HDI-based polyurethane elastomers show better tensile properties and HDI but poor thermal stability as compared to TDI-based elastomers. © 2018 Wiley Periodicals, Inc. J. Appl. Polym. In this study, plants belonging to family Solanaceae growing in Western Himalaya region have been observed palynologically under Light Microscope and Scanning electron microscope. Present investigation comprises of 10 genera and 23 species, namely, Atropa acuminata, Capsicum decoraticus, Capsicum frutescens, Cestrum aurantiacum, Cestrum diurnum, Cestrum nocturnum, Datura alba, Datura innoxia, Datura stramonium, Hyoscymus niger, Lycopersicon esculentum, Nicotiana rustica, Nicotiana tabacum, Petunia alba, Petunia hybrida, Solanum erianthum, Solanum melongena, Solanum miniatum, Solanum pseudocapsicum, Solanum surratense, Solanum tuberosum, Withania coagulans, Withania somnifera. Solanaceae is a eurypalynous family. Grains are usually Tricolporate and Tetracolporate, radially symmetrical, isopolar, prolate‐spheroidal to oblate‐spheroidal to oblate‐spheroidal to subprolate to per prolate or suboblate to oblate, size range: 8.55–72 μm, amb circular, semi‐angular or subangular, aperture drop‐type, labrum common‐type, exine usually 2 μm thick, nexine 1–1.5 μm thick. Tectum usually psilate, sexine reticulate, granulate or striato‐reticulate, with obscure pattern, sexine 1–2 μm thick, nexine 1–1.5 μm thick, and intine 0.5–1 μm thick. Most striking variation has been found in the shape class, aperture‐type, and tectal surface. Based on these characters, taxonomic keys have been made for correct identification of members in Solanaceae. However, the grains of this family are usually tricolporate and have direct relationship with certain members of the family Scrophulariaceae. Palyno‐morphological characters of family Solanaceae have been studied for the first time in Western Himalayan region of Pakistan. These palyno‐morphological characters are significant for identification of the members of family Solanaceae.  相似文献
