Decaffeination of food and beverage products is in high demand. In this study, a caffeine-degrading bacterium Burkholderia spp. was isolated from coffee plantation area of Chiang Mai province of Thailand. The bacterial isolates were first identified by morphological, physiological, and biochemical tests followed by 16S rDNA analysis. The bacterial isolate of Burkholderia spp. showed 45.5% of caffeine degradation in caffeine containing media (2.5 g/L) after 110 h of incubation period. Burkholderia spp. showed only 2.6% caffeine degradation when exposed to high concentrations of caffeine containing medium (20 g/L). The growth rate of Burkholderia spp. declined with the increase in the caffeine concentration, which indicated the inhibiting effect of caffeine at very high concentrations. The maximum growth rate of 0.053 h?1 was observed at 2.5 g/L of caffeine. Overall due to high caffeine tolerance and biodegradation of caffeine, Burkholderia spp. can be effectively used to degrade caffeine from agro-industrial wastes targeted for value added food applications and environmental remediation.  相似文献

Infection by Shiga toxin (Stx)-producing Escherichia coli of non-O157 and O157 serotypes are rare in China, but infection by O157 serotype was found in Shandong Province and three other provinces in China. To understand the reason for these rare infections and to determine the safety of retail meats in Shandong Province, we examined the distribution of Shiga toxin gene (stx)-bearing E. coli in retail meats and characterized the isolated stx-bearing strains. We used hybridization with DNA probes and isolated stx1- and/or stx2-positive E. coli from 31 (58%) of 53 retail meat samples, with beef showing the highest frequency (68%). Of 42 stx-positive isolates, none belonged to O157. Using the O157-specific immunomagnetic bead technique, we isolated E. coli O157 carrying the eae and stx2 genes from eight beef samples (26%). These strains produced little or no Stx2 and carried a unique q gene. Replication of the stx2 phages was detected in these strains, whereas stx2 phage replication was not detected in our previous study in which we examined similar stx2-bearing E. coli O157 strains from other Asian countries. Analysis of E. coli C600 lysogenized with the stx2 phages found in this study suggests that the lack of Stx2 production is due to changes in non-q gene region(s) of the phage genome or chromosomal mutation(s) in the host. Our data and reports by other workers suggest it is necessary to determine if various stx2-bearing E. coli O157 strains producing Stx2 to varying degrees are distributed in meats in various locations in China.  相似文献

Liposomes are used as effective nanodelivery devices to improve the physicochemical stability and biological efficacy of the encapsulated peptides and proteins. In this study, nanoliposome composite of lipoid S75-entrapped angiotensin I-converting enzyme (ACE)-inhibitory biopeptides was prepared by conventional (BLS75-CM) and direct heating (BLS75-DHM) methods. The nanoliposomes (BLS75-CM and BLS75-DHM) were stored at 4°C for 8 weeks and evaluated for physicochemical stability in terms of particle size, polydispersity index (pdi), zeta potential, and encapsulation efficiency (EE). These were also studied for residual ACE-inhibitory efficacy following their digestion under simulated gastrointestinal tract condition. The BLS75-CM was found to maintain higher physicochemical stability in terms of particle size, pdi, and zeta potential compared to BLS75-DHM. However, the BLS75-DHM indicated higher EE and efficacy with greater residual ACE-inhibitory activity of 47.37% compared to 44.18% and 36.84% that were obtained for the digested BLS75-CM and digested biopeptides without encapsulation, respectively. In vitro release study showed a cumulative biopeptides release of 66.41% and 69.00% from BLS75-CM and BLS75-DHM, respectively. The results of transmission electron microscopy showed spherical appearance of the nanoliposome capsules while Fourier Transform Infrared spectroscopy indicated the presence of ionic complexation and hydrogen bonds between the biopeptides and their phospholipid matrix.  相似文献

The market for functional foods has seen a sharp rise in demand in the recent years. This has driven researchers to multiply their efforts in producing functional meat products also. Feed manipulation and post-mortem modification of meat coupled with enrichment of bioactive compounds are gaining importance. This review discusses the candidate ingredients and strategies, utilized in crafting such functional meat products, and the notable developments and commercial successes in functional meat industry. Dry fermented sausages meet the conditions required to carry viable probiotic microbes. This article enlists various microorganisms that are being commercially used in functional food products and potential bacteria for probiotic sausage production.  相似文献

A thermophilic microorganism, SBS-4S, was isolated from a hot spring located in Gilgit, Northern Areas of Pakistan. It was found to be an aerobic, gram-positive, rod-shaped, thermophilic bacterium that grew on various sugars, carboxylic acids and hydrocarbons at temperatures between 45°C and 75°C. Complete 16S rRNA gene sequence of the microorganism exhibited homology to various species of genus Geobacillus. A highest homology of 99.8% was found with Geobacillus kaustophilus. A partial (0.7 kbp) chaperonin gene sequence also showed a highest homology of 99.4% to that of G. kaustophilus whereas biochemical characteristics of the microorganism were similar to Geobacillus uzenensis. Based on biochemical characterization, 16S rRNA and chaperonin gene sequences, we identified SBS-4S as a strain of genus Geobacillus. Strain SBS-4S produced several extracellular enzymes including amylase, protease and lipase. The lipase encoding gene was cloned, expressed in Escherichia coli and the gene product was characterized. The recombinant lipase was optimally active at 60°C with stability at wide pH range (6-12). The enzyme activity was enhanced remarkably in the presence of Ca(+2). The K(m) and the V(max) for the hydrolysis of p-nitrophenyl acetate were 3.8mM and 2273 μmol min(-1)mg(-1), respectively. The ability of the recombinant enzyme to be stable at a wide pH range makes it a potential candidate for use in industry.  相似文献

Lupin is a nonstarchy grain legume with high protein, dietary fibre and low fat contents. The industrial shift of lupin seed utilisation from feed to food has recently increased the scientific interest to explore its phytochemical composition and biological activities. Lupin seeds contain significant amounts of polyphenols, carotenoids, phytosterols, tocopherols, alkaloids and peptides with antioxidant, antimicrobial, anticarcinogenic and anti‐inflammatory activities. Among polyphenols, genistein and their derivatives (isoflavones) are of great importance because of their phytoestrogenic potential.
