Alkaloids of Thalictrum. XV. Isolation and identification of the hypotensive alkaloids of the root of Thalictrum lucidum

Sixteen alkaloids, namely homoaromoline, obaberine, O-methyl-thalicberine, oxyberberine, thalidasine, thaliglucinone, thalrugosine, obamegine, oxyacanthine, berberine, columbamine, jatrorrhizine, magnoflorine, palmatine, thalifendine and base A chloride, plus the artifact, 8-trichloromethyldihydroberberine, were isolated from the alkaloid fraction of the roots of Thalictrum lucidum L. Of these, obamegine, thalrugosine, O-methylthalicberine, thaliglucinone, obaberine and homoaromoline were found to possess hypotensive activity in normotensive dogs. Thalidasine, homoaromoline, thalrugosine, thaliglucinone, obamegine, jatrorrhizine, and columbamine were found to possess antimicrobial activity against Mycobacterium smegmatis at a concentration of 100 mug/ml or less.     

Analytical expressions for tunnel currents in metal-insulator-metal (MIM) and metal-insulator-semiconductor (MIS) structures in a two-band model

A new closed-form approximation for the tunnel current in MIM and MIS structures is derived assuming a general E-k relationship and barrier shape. An upper limit for the error involved in this approximation is estimated. The existing closed-form expressions for the tunnel currents are discussed and their limitations are exposed.     

The effects of norgestrel on carbohydrate and lipid metabolism over one year

Carbohydrate and lipid metabolism were studied with the use of a three-hour oral glucose tolerance test in 71 women before and after one year of daily oral norgestrel, 0.075 mg., treatment. There was no significant change in weight or fasting plasma triglyceride and cholesterol levels. There was a significant elevation of both the blood glucose and plasma insulin levels after one year of treatment. This was true for both the fasting and the glucose-stimulated values. Whereas all of the individual glucose tolerance curves at the pretreatment control test were normal by selection, 15.5 per cent of the curves were borderline abnormal to abnormal at the one-year test. The significance of these metabolic alterations is discussed.     

Ultrastructural studies of prepubertal porcine uterine tube epithelium

Ultrastructural details of prepubertal porcine uterine tube (oviduct) were studied in normal, growing gilts and compared with observations reported in other species. Tissues from the ampulla region of uterine tube were taken from 6 prepubertal gilts (106 to 139 days old) to determine cytodifferentiation of ciliated and secretory cells. The epithelium consisted of 2 distinctive cells, the ciliated and the secretory cells. Cilia were observed in the uterine tube of prepubertal gilts; however, degeneration of cilia was not observed in the present study. Most prominent observations were the occurrence of fibrous granules in the apical cytoplasm of ciliated cells. These fibrous granules contained electron-dense material and were present near basal bodies. The most unusual feature was the occurrence of procentrioles around a condensation form. These data indicate that ciliated cells are sensitive to estrogen. Intimate morphologic association between fibrous granules and basal bodies indicate that fibrous granules might provide precursor material for the development of cilia and rootlets. The cytoplasm of the secretory cells contained rough endoplasmic reticulum of tubular form and numerous ribosomes. Evidence for synthesis, storage, and release of secretory granules was not apparent. It is suggested that the secretory cells are not sensitive to the low, circulating concentration of plasma estrogen. The ultrastructure of the stromal cells and lymphatic capillary was described for the 1st time. The uterine tube stromal cells were characterized by prominent nucleus and a few cytoplasmic organelles. The lymphatic capillaries were distinguished by the blood capillaries, their much wider lumen, endothelium with an attenuated cytoplasm, absence of basal lamina, and overlapping and interdigitating intercellular junctions. The fine structure of the porcine uterine tube lymphatic capillary generally resembled that of other mammalian species.     

Distribution of Water Masses in and Near the Straits of Mackinac

Three cruises in the Straits of Mackinac area were conducted from 30 August to 8 October 1973. Samples were taken from up to 11 depths during each cruise and analyzed for silica, nitrate, total and soluble phosphorus, chlorophyll a, pH, water temperature, and specific conductance. Cluster analyses were used to visualize the relationship between stations for each depth on all cruises. The clearest relationships observed were from 0, 5, and 10 m depths, while clusters of data from other depths were not as useful in describing water masses. Analyses of 0 - 10 m depths showed a consistent homogeneous water mass extending through the northern edge of the Straits proper, past the western edge of Mackinac Island, to south of Bois Blanc Island. This cluster of stations was found during most wind conditions at the three depths considered. The last cruise, 6-8 October, occurred during a period of light winds from the southeast which partially disrupted this cluster. Relationships between the variables were defined by factor analysis which showed silica and nitrate increased with decreasing water temperature and pH, and partly as a function of depth. Secchi disc values tended to vary independently of most other variables, implying these readings had little value in explaining water quality in the Straits region.     

Ruptures of the tendo achillis. An objective assessment of surgical and non-surgical treatment

A comparative study is presented of seventy-nine patients with rupture of the tendo achillis, forty-eight of whom were treated surgically and thirty-one, non-surgically. With one exception, all were healthy, athletic individuals. Twenty-two per cent of the patients had an initial misdiagnosis. The patients treated surgically were more satisfied with the results of their treatment. Strength, power, and endurance as measured on the Cybex II dynamometer revealed that the patients treated non-surgically attained only 72 per cent of normal strength and 70 per cent of normal power and endurance as compared with the surgically treated patients. Wound complications occurred in only two patients and no reruptures occurred in the surgically treated group, whereas there were nine rereptures in the group not treated surgically.     

Differential expression of plasma membrane calcium pump mRNA isoforms in rat osteoblast-like cells

Cryptococcal meningitis is a common infection in patients with AIDS. Using a murine cryptococcosis model, we compared treatment with a new triazole, D0870, and fluconazole. Groups of ICR (Institute for Cancer Research) mice were infected intracerebrally with eight different isolates of Cryptococcus neoformans variety neoformans with different in vitro susceptibilities to fluconazole. For survival studies mice were challenged with two to four times LD(50) or six to nine times LD(50). Treatment was given for 10 days. Mice were observed through to day 30. To assess the effect of treatment on fungal tissue burden, mice received a three to five times LD(50) inoculum and treatment for 10 days. They were sacrificed on day 12 and serial dilutions of brain homogenates were cultured. Fluconazole prolonged survival primarily in isolates which were susceptible in vitro. D0870 prolonged survival in all isolates except one, which was also resistant in vitro to D0870 and fluconazole. Both drugs reduced colony counts of all isolates. D0870 warrants further development for use in cryptococcosis, and appears effective for isolates relatively resistant to fluconazole. There is a relative correlation of in vivo and in vitro susceptibility to D0870 as well as fluconazole.
