In the development of novel biomarkers, the proteomic approach is advantageous because using it the cancer-associated proteins can be directly identified. We previously developed a 2-nitrobenzenesulfenyl (NBS) method to improve quantitative proteome analysis. Here, we applied this method to proteomic profiling of colorectal carcinoma (CRC) to identify novel proteins with altered expression in CRC. Each pair of tumor and normal tissue specimens from 12 CRC patients was analyzed, and approximately 5000 NBS-labeled paired peaks were quantified. Peaks with altered signal intensities (>1.5-fold) and occurring frequently in the samples (>70%) were selected, and 128 proteins were identified by MS/MS analyses as differentially expressed proteins in CRC tissues. Many proteins were newly revealed to be CRC related; 30 were reported in earlier studies of CRC. Six proteins that were up-regulated in CRC (ZYX, RAN, RCN1, AHCY, LGALS1, and VIM) were further characterized and validated by Western blot and immunohistochemistry. All six were found to be CRC-localized, either in cancer cells or in stroma cells near the cancer cells. These results indicate that the proteins identified in this study are novel candidates for CRC markers, and that the NBS method is useful in proteome mining to discover novel biomarkers.  相似文献

The extrusion/equal channel angular pressing (EX-ECAP) processing procedure, in which magnesium-based alloys are subjected to extrusion followed by ECAP, was applied to a Mg-7.5 pct Al-0.2 pct Zr alloy prepared by casting. Microstructural inspection showed the EX-ECAP process was effective in reducing the grain size from ∼21 μm after extrusion to an as-pressed grain size of ∼0.8 μm. It is shown through static annealing that these ultrafine grains are reasonably stable up to 473 K, but grain growth occurs at higher temperatures. Tensile specimens were cut from the billets prepared by EX-ECAP and testing showed these specimens exhibited superplasticity at relatively low temperatures with maximum elongations up to >700 pct. By processing through EX-ECAP to a higher imposed strain and thereby increasing the area fraction of high-angle boundaries, it is demonstrated that there is a potential for achieving high-strain-rate superplasticity. Experiments were conducted to evaluate the utility of a new processing procedure developed for Mg-based alloys in which samples are subjected to a two-step processing route of extrusion followed by equal-channel angular pressing (designated as EX-ECAP). The experiments were conducted using a Mg-0.6 wt pct Zr alloy and, for comparison purposes, samples of pure Mg. It is shown that the potential for successfully using ECAP increases in both materials when adopting the EX-ECAP procedure. For the Mg-Zr alloy, the use of EX-ECAP produces a grain size of ∼1.4 μm when the pressing is undertaken at 573 K. By contrast, using EX-ECAP with pure Mg at 573 K produces a grain size of ∼26 μm. Tensile testing of the Mg-Zr alloy at 523 and 573 K after processing by EX-ECAP revealed the occurrence of significantly enhanced ductilities with maximum elongations of ∼300 to 400 pct.  相似文献

In this paper, it is proposed to use a new type of solid particle impact test (slurry jet) to swiftly evaluate wear properties of thin, single layered or multilayered coatings. By the slurry jet, 1.2 μm alumina particles were impacted at high velocity perpendicular to thin PVD coatings of TiN deposited on high speed steel substrate materials under various substrate temperatures. Since the coatings have a much higher wear resistance than the substrate material, the wear rate increases significantly to the higher level of the HSS material when the coatings are penetrated. This is utilized in the quantification of the assessment of coating wear. A ranking of wear resistance and correlations to the coating surface hardness measured by nano-indentation tests, and coating morphology and structures are given and discussed. The TiN deposited under the highest substrate temperature proved to have the highest wear resistance although it had a relatively low hardness. The wear rate of the TiN coatings varies with the orientation of grains, that is, the {1 1 1} orientation that dominates for the high temperature deposition shows a higher wear resistance than the {1 0 0} orientation, which corresponds with the cleavage fracture behavior. Thus, it can be recommended as a screening test when evaluating coatings and coated materials.  相似文献

Thermal analysis has been utilized in polymer research for a long time and its usefulness is widely recognized. High‐performance commercial instruments have driven significant progress in the thermal studies of polymers. On the other hand, new experimental instruments have been developed in laboratories as well. In this paper attention is paid to the latter category. The laboratory‐made instrument focused on the target material and/or phenomena provides data that are not available from other techniques. Experiments using the temperature modulation technique, which has been successfully utilized in laboratory‐made instruments, are introduced in this paper. The temperature modulation technique reveals a new aspect of temperature useful for studies of material properties. © 2016 Society of Chemical Industry  相似文献

Solid electrolyte interface (SEI) films formed on Li1−xCoO2 electrodes were observed with hard X-ray photoelectron spectroscopy (HX-PES). This paper particularly focuses on film thickness estimation using HX-PES with theoretical calculation. The validity of the calculation was proven by experiments using model SEI films. The native film formed on a LiCoO2 composite electrode was estimated to be LiF with its thickness of 5 nm. Formation of Co (II) species on top of LiCoO2 was also indicated. Storage of the electrode at 60 °C brought about considerable film growth (30-40 nm) with carbonate compounds formation. SEI film changes during charging of the LiCoO2 electrode were also examined. The main component in the film was deduced to be LiF or a kind of fluorite, with its thickness decreased during charging. The SEI formation mechanisms are also elucidated.  相似文献

Supercritical water gasification (SCWG) has attracted attention as a technology for utilizing wet biomass. We used a fluidized bed of alumina particles to prevent blockage of a SCWG reactor. A glucose solution was heated in the reactor with and without fluidized alumina particles. In the absence of alumina particles, char particles formed homogeneously in the reactor, but the use of a fluidized bed resulted in accumulation of char particles at the reactor’s exit rather than inside the reactor. Therefore, the fluidized bed was effective at preventing blockage of the reactor. However, the alumina particles did not remove deposits from the reactor’s walls. Instead, the fluidized bed caused larger char particles to form, preventing their adhesion to the reactor’s wall.  相似文献

We fabricated CaCO3-coated vesicles as drug carriers that release their cargo under a weakly acidic condition. We designed and synthesized a peptide lipid containing the Val-His-Val-Glu-Val-Ser sequence as the hydrophilic part, and with two palmitoyl groups at the N-terminal as the anchor groups of the lipid bilayer membrane. Vesicles embedded with the peptide lipids were prepared. The CaCO3 coating of the vesicle surface was performed by the mineralization induced by the embedded peptide lipid. The peptide lipid produced the mineral source, CO32−, for CaCO3 mineralization through the hydrolysis of urea. We investigated the structure of the obtained CaCO3-coated vesicles using transmission electron microscopy (TEM). The vesicles retained the spherical shapes, even in vacuo. Furthermore, the vesicles had inner spaces that acted as the drug cargo, as observed by the TEM tomographic analysis. The thickness of the CaCO3 shell was estimated as ca. 20 nm. CaCO3-coated vesicles containing hydrophobic or hydrophilic drugs were prepared, and the drug release properties were examined under various pH conditions. The mineralized CaCO3 shell of the vesicle surface was dissolved under a weakly acidic condition, pH 6.0, such as in the neighborhood of cancer tissues. The degradation of the CaCO3 shell induced an effective release of the drugs. Such behavior suggests potential of the CaCO3-coated vesicles as carriers for cancer therapies.  相似文献

After solvent extraction of Taiheiyo, Miike and Balmer coals using wash oil under nitrogen atmosphere at 370 °C for 30 min, the extraction yield is always within the additivity law. Further studies used Yallourn, Soyakoishi, Taiheiyo, Horonai, Miike, Shin Yubari, Balmer coals and their blends which were hydrogenated in tetralin, wash oil or creosote oil, with or without catalyst, at 400–450 °C under 10 or 3 MPa of initial hydrogen pressure. When hydrogen is available, the additivity law exists for blended coals, but when the hydrogen supply is deficient, the experimental conversion of blended coals is always lower than calculated conversions. This may be due to the faster consumption of the hydrogen by more reactive coals and thus the less reactive coals were unable to react with hydrogen.  相似文献
