Metabolic syndrome is associated with an increased risk of colorectal cancer. This study investigated the impact of hypertension, a component of metabolic syndrome, on azoxymethane (AOM)-induced colorectal carcinogenesis using SHRSP/Izm (SHRSP) non-diabetic/hypertensive rats and SHRSP.Z-Leprfa/IzmDmcr (SHRSP-ZF) diabetic/hypertensive rats. Male 6-week-old SHRSP, SHRSP-ZF, and control non-diabetic/normotensive Wister Kyoto/Izm (WKY) rats were given 2 weekly intraperitoneal injections of AOM (20 mg/kg body weight). Two weeks after the last injection of AOM, the SHRSP and SHRSP-ZF rats became hypertensive compared to the control WKY rats. Serum levels of angiotensin-II, the active product of the renin-angiotensin system, were elevated in both SHRSP and SHRSP-ZF rats, but only the SHRSP-ZF rats developed insulin resistance, dyslipidemia, and hyperleptinemia and exhibited an increase in adipose tissue. The development of AOM-induced colonic preneoplastic lesions and aberrant crypts foci, was significantly accelerated in both SHRSP and SHRSP-ZF hypertensive rats, compared to WKY normotensive rats. Furthermore, induction of oxidative stress and exacerbation of inflammation were observed in the colonic mucosa and systemically in SHRSP and SHRSP-ZF rats. Our findings suggest that hypertension plays a role in the early stage of colorectal carcinogenesis by inducing oxidative stress and chronic inflammation, which might be associated with activation of the renin-angiotensin system.  相似文献

An optimum nanostructure and pore size of catalyst supports is very important in achieving high catalytic performances. In this instance, we evaluated the effects of various carbon nanostructures on the catalytic performances of carbon‐supported platinum (Pt/C) electrocatalysts experimentally and numerically. The Pt/C catalysts were prepared using a hybrid method involving the preparation of dense, hollow, and porous nanostructured carbon particle via aerosol spray pyrolysis followed by microwave‐assisted Pt deposition. Electrochemical characterization of the catalysts showed that the porous Pt/C catalyst gave the best performance; its electrochemical surface area was much higher, more than twice than those of hollow or dense Pt/C. The effects of pore size on electrocatalysis were also studied. The results showed the importance of a balance between mesopores and macropores for effective catalysis with a high charge transfer rate. The mechanism controlling the fatigue life of a precipitation-hardened Al–Mg–Si alloy (6061-T6) at a high-cycle fatigue (HCF) regime of over 107 cycles was investigated in detail. It was found that over 90% of the total fatigue life was occupied by the growth process of a microstructurally small crack at relatively low stress amplitude. The small crack was often found to be arrested and halted for a long period (more than 106 cycles) before it began to grow again, which resulted in a significantly slow growth process. The small crack was then analyzed not only by the conventional fractography but also by the cross-sectional observation of the crack tip region using a focused ion beam and transmission electron microscopy. These observations, supplemented also by a grain orientation analysis using electron backscattered diffraction, explicitly revealed the following points: (i) the small crack growth observed on the specimen surface is primarily related to facet-type cracking that occurs exclusively at the specimen surface; (ii) the growth direction of the small crack has strong anisotropy (i.e. surface-induced growth); (iii) the facet-type cracking is related to the formation of persistent fine slip bands that accompany no structural change of the matrix. On the basis of these results, the micromechanism of small crack growth and its relation to the concept of fatigue limit at the HCF regime is discussed in detail.  相似文献

Pressure drop characteristics of flow in a periodically grooved channel are investigated experimentally. It is well known that a self-sustained oscillatory flow occurs from a steady-state flow at a certain critical Reynolds number in such grooved channels. The oscillatory flow enhances fluid mixing and leads to an increase in pressure drop. We measure the pressure drop with a pressure transducer. It is found that the pressure drop increases near the critical Reynolds numbers where the two- and three-dimensional oscillatory flows occur. In addition, the three-dimensional flow is confirmed by flow visualization.  相似文献

Potato starch is known to have a higher concentration of phosphate than other starches. The presence of phosphate groups in amylopectin results in resistance to digestion by amylase. Therefore, there is a possibility that potato starch is slowly digested, inducing a physiological effect similar to that of resistant starch and indigestible oligosaccharides. The amount of phosphate group in starch differs with potato cultivar. In the present study, we investigated the effects of gelatinized potato starch containing a high level of phosphorus on lipid metabolism in rats. For this purpose, we determined lipid levels in the serum and liver in rats fed two kinds of gelatinized potato starches with different phosphorus contents. Four groups of male Sprague-Dawley rats were fed a 60% sucrose diet (control) or one of three diets containing cornstarch (CS), Benimaru (BM) potato starch or Hokkaikogane (HK) potato starch. Fat pad weight was slightly decreased in the HK diet group compared with that in the other groups. Free fatty acids in serum were significantly lowered by dietary HK starch compared with control, and serum triglyceride in rats fed the HK diet was also decreased. In the BM and HK diet groups, triglyceride levels in the liver were decreased compared with that in the control and CS groups. As for hepatic total cholesterol level, there were no significant differences among three starch diet groups. Fecal bile acid excretion was greater in the two potato starch groups than in the control group. On the other hand, there were no significant differences in cecal short-chain fatty acid content or pH. Thus, we conclude that dietary gelatinized potato starch reduces free fatty acid and triglyceride in serum and hepatic triglyceride, but does not affect cecal fermentation.  相似文献

Novel azobenzene-based photo-responsive amorphous molecular materials, 4-[bis(9,9-dimethylfluoren-2-yl)amino]-4′-cyanoazobenzene and 4-[bis(9,9-dimethylfluoren-2-yl)amino]-4′-nitroazobenzene, have been synthesized and the formation of surface relief grating on their amorphous films has been investigated. It was found that a relatively large surface relief grating could be inscribed on both amorphous films upon interference exposure to the writing laser beams. The modulation depth of the surface relief grating inscribed on the amorphous film of the cyano-substituted material was found to be larger than that inscribed on the film of the nitro-substituted one and seemed to be comparable to that inscribed on the amorphous film of the parent material, 4-[bis(9,9-dimethylfluoren-2-yl)amino]azobenzene. These results were discussed from the viewpoint of their trans–cis photoisomerizations as amorphous films and glass-transition temperatures.  相似文献

The hyperbranched polymers (HBP-SA-Acs) with both a sulfonic acid group as a functional group and an acryloyl group as a cross-linker at terminals in different ratios of sulfonic acid group/acryloyl group (SO3H/Ac) were successfully synthesized as a new thermally stable proton-conducting electrolyte. The cross-linked hyperbranched polymer electrolyte membranes (CL-HBP-SAs) were prepared by thermal polymerizations of the HBP-SA-Acs using benzoyl peroxide, and their ionic conductivities under dry condition and thermal properties were investigated. The ionic conductivities of the CL-HBP-SAs were found to be in the range of 2.2 × 10−4 to 3.3 × 10−6 S/cm, depending upon the SO3H unit contents, at 150 °C under dry condition, and showed the Vogel-Tamman-Fulcher (VTF) type temperature dependence, indicating that proton transfer is cooperated by local polymer chain motion. All CL-HBP-SAs were thermally stable up to 260 °C, and they had suitable thermal stability as electrolyte membranes for the high-temperature fuel cells under dry condition. Fuel cell measurement using a single membrane electrode assembly cell with a cross-linked electrolyte membrane was successfully performed under non-humidified condition. It was demonstrated that applying the concept of dry polymer system to proton conduction is one possible approach toward high-temperature fuel cells.  相似文献

Ruthenium-oxide nanosheet (RuO2ns) crystallites with thickness less than 1 nm were prepared via chemical exfoliation of a layered potassium ruthenate and deposited onto carbon supported platinum (Pt/C) as a potential co-catalyst for fuel cell anode catalysts. The electrocatalytic activity towards carbon monoxide and methanol oxidation was studied at various temperatures for different RuO2ns loadings. An increase in electrocatalytic activity was evidenced at temperatures above 40 °C, while little enhancement in activity was observed at room temperature. The RuO2ns modified Pt/C catalyst with composition of RuO2:Pt = 0.5:1 (molar ratio) exhibited the highest methanol oxidation activity. CO-stripping voltammetry revealed that RuO2ns promotes oxidation of adsorbed CO on Pt. In addition to the enhanced initial activity, the RuO2ns modified Pt/C catalyst exhibited improved stability compared to pristine Pt/C against consecutive potential cycling tests.  相似文献

Stroke is a major cause of death worldwide, leading to serious disability. Post-ischemic injury, especially in the cerebral ischemia-prone hippocampus, is a serious problem, as it contributes to vascular dementia. Many studies have shown that in the hippocampus, ischemia/reperfusion induces neuronal death through oxidative stress and neuronal zinc (Zn2+) dyshomeostasis. Glutathione (GSH) plays an important role in protecting neurons against oxidative stress as a major intracellular antioxidant. In addition, the thiol group of GSH can function as a principal Zn2+ chelator for the maintenance of Zn2+ homeostasis in neurons. These lines of evidence suggest that neuronal GSH levels could be a key factor in post-stroke neuronal survival. In neurons, excitatory amino acid carrier 1 (EAAC1) is involved in the influx of cysteine, and intracellular cysteine is the rate-limiting substrate for the synthesis of GSH. Recently, several studies have indicated that cysteine uptake through EAAC1 suppresses ischemia-induced neuronal death via the promotion of hippocampal GSH synthesis in ischemic animal models. In this article, we aimed to review and describe the role of GSH in hippocampal neuroprotection after ischemia/reperfusion, focusing on EAAC1.  相似文献
