By exploiting the interferometric antireflection action of a probe sample, consisting of a diamond-like carbon (DLC) film grown on Si, combined with a specific illumination spectrum, we designed and constructed an optical device for the visual remote sensing of radiation (either plasma or atomic oxygen) and for the visual inspection of adsorbed organic contamination as thin as a few molecular layers. The capabilities of this new visual interferometric multi-indicator (VIMI) enable the bare-eye color detection of thickness changes on the order of a few nanometers without the intervention of any instrumental or computer interface.  相似文献

We derive a closed-form expression for the mutual impedance due to eddy-current induction for a pair of cylindrical air-core coils with arbitrary position and orientation above a planar conductor. By extending a recently devised model for individual coils with an arbitrary tilt with respect to the surface, we obtain a remarkably simple result. We validated our model with measurements on a conductive plate. The results should be useful for designing new probe configurations and for evaluating the signals in eddy-current inspections when driver-pickup coil configurations are utilized.  相似文献

The paper presents new detailed emissions inventory for the Greater Area of Thessalonilki, northern Greece, which incorporates a large amount of data on the polluting anthropogenic and biogenic activities in the region, including the location of all individual point, line and area sources along with their daily, weekly and monthly profiles and activity rates. The emission estimate followed both the bottom-up and the top-down approaches. Apart from the base year calculations, scenario estimates are provided for the base year, intended to show the maximum possible benefit of abatement measures and for the future, in order to estimate the evolution of air pollutant emissions in relation to structural changes and land planning. Air pollutant emissions were calculated for an area of 5,120 km2 at two different spatial resolutions (1x1 km2 and 2x2 km2) in order to support air quality simulations in the region. The resulting spatially allocated emission database, which also incorporates elevation and land use data, constitutes a clear improvement in comparison to existing emissions inventories of the area, and the procedure followed can be considered as a 'best practice' guide for developers of urban emissions inventories.  相似文献

In this paper exact analytical expressions for the impedance of a cylindrical air-core coil above a layered metal structure whose conductivity varies continuously with depth are presented. Although the model is general, attention is focused on three conductivity profiles: the linear, the quadratic and the exponential. The derived expressions for the impedance change for each profile could provide a useful tool for the solution of the inverse problem: that of determining the conductivity from variable frequency measurements of the impedance. Furthermore, the obtained final formulas contain elegant mathematical functions and show a substantially higher computational efficiency with respect to existing methods  相似文献

Small molecules inducing protein degradation are important pharmacological tools to interrogate complex biology and are rapidly translating into clinical agents. However, to fully realise the potential of these molecules, selectivity remains a limiting challenge. Herein, we addressed the issue of selectivity in the design of CRL4CRBN recruiting PROteolysis TArgeting Chimeras (PROTACs). Thalidomide derivatives used to generate CRL4CRBN recruiting PROTACs have well described intrinsic monovalent degradation profiles by inducing the recruitment of neo-substrates, such as GSPT1, Ikaros and Aiolos. We leveraged structural insights from known CRL4CRBN neo-substrates to attenuate and indeed remove this monovalent degradation function in well-known CRL4CRBN molecular glues degraders, namely CC-885 and Pomalidomide. We then applied these design principles on a previously published BRD9 PROTAC (dBRD9-A) and generated an analogue with improved selectivity profile. Finally, we implemented a computational modelling pipeline to show that our degron blocking design does not impact PROTAC-induced ternary complex formation. We believe that the tools and principles presented in this work will be valuable to support the development of targeted protein degradation.  相似文献

In human serum immunoglobulin G (IgG), a rare modification of biantennary complex N-glycans lead to a β1,4-galactosylated bisecting GlcNAc branch. We found that the bisecting GlcNAc on a biantennary core-fucosylated N-glycan was enzymatically galactosylated under stringent reaction conditions. Further optimizations led to an efficient enzymatic approach to this particular modification for biantennary substrates. Notably, tri- and tetra-antennary complex N-glycans were not converted by bovine galactosyltransferase. An N-glycan with a galactosylated bisecting GlcNAc was linked to a lanthanide binding tag. The pseudo-contact shifts (PCS) obtained from the corresponding Dy-complex were used to calculate the conformational preferences of the rare N-glycan. Besides two extended conformations only a single folded conformation was found.  相似文献

Object

Cytoplasmic lipid droplets (LDs) are dynamic cellular organelles; their accumulation is associated with several cellular processes, such as cell proliferation, apoptosis and necrosis. 1H Nuclear Magnetic Resonance (NMR) spectroscopy detects resonances from lipids present in cytoplasmic (LDs); an understanding of the relationship between LD characteristics and NMR lipid signals is important.

Materials and methods

In this study, five nervous system cancer cell lines were investigated. Nile red staining was used to measure the diameter of LDs. High-resolution magic angle spinning NMR (HR-MAS) was performed on harvested cell pellets to quantify the patterns of lipid signals.

Results

LDs were present in all five cell lines with different morphology. An average LD diameter of approximately 0.2???m was found in all cell types. Diameter of the largest LDs varied across the cell lines. The intensity of NMR lipid signals varied greatly between cell types, and a good correlation was found between total volume of LDs and the proton NMR lipid signal intensity at 0.9 and 1.3?ppm.

Conclusion

The correlation implied that little NMR signal is detected from LDs of diameters less than approximately 0.34???m, most likely due to restriction of rotational motion of the lipids.  相似文献

By utilizing the second-order vector potential formulation we solve the eddy current problem of a coil inside a conductive tube and in an offset position to it. The study focuses on the derivation of a closed-form expression for the impedance change of the coil, which is used to calculate the signal produced by wobble in eddy current testing of tubes with bobbin coils.  相似文献
