High performance liquid chromatographic separation of stereoisomeric bile acids as their UV-sensitive esters

High performance liquid chromatographic separation of a series of mono-, di- and trihydroxylated 5β-cholanic acids which differ only in position and configuration of hydroxyl groups at positions C-3, C-7 and/or C-12, is reported. The C-24 free acids were derivatized to four different classes of UV-sensitive esters, i.e.,p-bromophenacyl (BP),m-methoxyphenacyl (MP), 4-nitrophthalimidemethyl (NPM) and 9-anthrylmethyl (AM) esters, and chromatographed on two, variants of C₁₈ reversed-phase columns (Nova-Pak C₁₈ and Zorbax ODS) with methanol-water systems as mobile phase. Separation efficiency and elution order of some isomeric pairs were influenced by both the structure of the C-24 ester groups and the nature of the columns used. Excellent chromatographic properties were found for those derivatives, particularly for the NPM esters.     

The effect of furosemide on evoked otoacoustic emissions in guinea pigs

After recording transiently evoked otoacoustic emissions (TEOAEs) to a click stimulus in guinea pigs by using the IL088 which was developed by Bray and Kemp (1987) for easy recording and analysis of TEOAE, the changes after intravenous administration of furosemide (30 mg/kg or 50 mg/kg) were examined. The wave of the TEOAE could be detected from 20 of 24 ears (83%). After the i.v. injection of furosemide (30 mg/kg), TEOAE powers (total echo power and highest peak power in FFT pictures) decreased quickly and showed minimum values after 5-10 min. Then they increased rapidly and recovered normally within 60 min after injection. However, no ears showed TEOAEs during the 5- to 10-min period following the injection of the 50-mg/kg dose of furosemide. They then recovered slowly as compared with the group treated with the lower dose of furosemide (30 mg/kg). These changes are similar to those of the endocochlear potential (EP) after furosemide injection. These data support the notion that the EP can contribute to the mechanism of TEOAE generation.     

Enzymatic degradation of esters of dichloropropanols: removal of chlorinated glycerides from processed foods

The esters of dichloropropanols from the byproduct of hydrolysates during food processing were degraded by the cell-free extracts of a dichloropropanol-assimilating bacterium of Pseudomonas sp. OS-K-29. 1-Acyloxy-2,3-dichloropropane was hydrolysed into the corresponding carbonic acid and 2,3-dichloropropanol, and then the dichloropropanol was dechlorinated. The esterase activity of the cell-free extracts was effective for the esters of the dichloropropanols and monoacylglycerides, but not for hydrolysing the lipase substrates such as triacylglycerides and Tween 60. A comparative study using a series of octanoyl glycerides and the esters of the dichlopropanols showed that the substrate specificities of the cell-free extracts were similar to the monoacylglyceride lipase from Penicillium camembertii.     

Development of a new evacuation method for emergencies: Control of collective behavior by emergent small groups.

A new evacuation method for emergencies, the Follow Me method, in which each leader takes only one evacuee to an exit without gesturing or loudly directing many evacuees toward the exit, was compared, in two field experiments, with a traditional method, the Follow Directions method, in which each leader indicates the direction with a loud voice and vigorous gestures. Results showed that the Follow Me method was more effective when the leader-to-evacuee ratio was relatively small, such as 1:4, and less effective when the ratio was large, such as 1:8. Effective evacuation by the Follow Me method was shown to be caused by the drawing power of emergent small groups. Generating from around a leader, other evacuees rapidly formed a collective stream toward the exit. Small groups can be used as a lever in moving a larger collectivity in an emergency, as well as in activating a large formal organization. Opioid receptors altered binding nature in guinea-pig brain following the development of morphine dependence

Morphine is well known to produce tolerance and dependence. The mechanisms for these phenomena are not clearly understood and there are a number of conflicting reports that chronic morphine administration decreases, increases, or leaves unchanged the number of opioid binding sites. We examined the potency of MScontin (oral controlled-release preparation of morphine) to induce morphine dependence and also determined the change of mu, delta and kappa opioid receptor types in brain homogenates obtained from morphine-dependent guinea-pigs. 1. Guinea-pigs were implanted subcutaneously with MScontin (300 mg.kg-1) and naloxone was employed to precipitate jumping behavior of withdrawal symptoms at various times. The highest degree of physical dependence was observed on the 2nd day after implantation. Therefore, this period was chosen to investigate opioid receptor binding assay. 2. Two days after implantation, the binding of 3H-DAGO (mu agonist), 3H-DPDPE (delta agonist) and 3H-U69593 (kappa agonist) to brain membranes prepared from morphine dependent and control guinea-pigs was determined. Scatchard plot of the saturation binding data revealed an increase in Bmax values (maximum specific binding) and no change in the Kd values (equilibrium dissociation constants) of 3H-opioid ligand bindings obtained from morphine-dependent animals as compared to controls. These results indicate that brain mu, delta and kappa opioid receptors are up-regulated in morphine dependent guinea-pigs.     

Minor role of fractographic features in basic fatigue crack propagation properties

The fatigue crack propagation rate and the fatigue threshold were investigated for transverse-butt-welded joints of the austenitic stainless steel SUS304. Specimens were of the center-cracked type. In three sets of tests the fatigue crack passed through the weld metal, and in the other two sets, through the base metal. The fatigue crack propagation properties coincided with each other at different stress ratios for the weld metal, and at the higher stress ratios for the base metal. The propagation properties improved at the stress ratio of zero for the base metal. The absence of a stress ratio effect means that the coincided properties are basic ones in which fatigue crack closure does not occur. However, fractographic appearance and surface roughness were quite different between weld metal and base metal. The coincidence of fatigue crack propagation properties in spite of the difference in fractographic features reveals that the fractographic appearance and surface roughness only have a minor effect on basic fatigue crack propagation behaviour.
