Electric field methods are powerful tools for cell characterization and manipulation. Important biotechnological applications, e.g., electrofusion of cells or electroinjection of macromolecules into living cells, rely on membrane breakdown. Despite the widespread use of these techniques, the process of membrane breakdown is poorly understood: a better understanding can be expected to lead to higher efficiencies. The present study used planar lipid bilayers in order to limit the number of experimental parameters. Membrane rupture was induced by careful application of short electric field pulses: measurement of the subsequent increase in membrane conductivity with time permitted the underlying mechanism to be characterized. The initial process of pore formation starts a few μs after the onset of the pulse, and the ensuing breakdown of the entire lipid membrane occurs within about a millisecond (pore widening velocity of a few cm/s). The kinetics of the pore-opening process could be influenced qualitatively by adding surfactants and lipid-attached macromolecules, respectively. Changing the effective mass per unit area of membrane, which could be brought about by using known percentages of lipids with covalently bound, hydrophilic polymers, allowed a model for the pore widening process to be quantitatively tested  相似文献

A questionnaire survey was carried out to examine the attitudes and practices of Australian and New Zealand intensivists with regard to brain death and organ donation. A return rate of 82.5% was achieved. Fifty-eight per cent had written evidence of their own wishes to donate organs and 94% would agree to donation from a dependent. At least one intensivist is involved in certifying brain death on 95% of occasions. Intensivists are involved in the request for organ donation over 90% of the time although one-third do not believe that it is their role to request organ donation. Although two-thirds believe that the family should always be approached for organ donation, another 52 out of 254 indicated that it was their (the intensivist's) role to decide if families should be asked for organ donation. Possible reasons for not requesting are language or other communication problems, perceptions of cultural differences and degrees of family distress. Twenty per cent of respondents do not provide haemodynamic support before brain death confirmation. Australian and New Zealand intensivists overwhelmingly support the concept of brain death, current methods of confirmation of brain death, organ donation and transplantation. Possible reasons behind loss of potential donors include decisions not to resuscitate both before and after brain death is confirmed. Perceptions of family grief and cultural differences clearly inhibit requests for organ donation. A very few units have an effective policy on approaching families about organ donation. Intensivists have almost exclusive control over requests for organ donation and thus bear a full professional responsibility for this element of hospital practice.  相似文献

To determine the clinical usefulness of Acridine Orange (AO) staining of spermatozoa as a screening test for the evaluation of semen quality during basic infertility investigation, semen smears from 103 randomly chosen males of subfertile couples were examined. The median duration of infertility was 4.5 years (range 1-15) and the median age was 33 years (range 21-43). The outcome of AO staining ranged from 5 to 81%, with a median of 24%, green fluorescent spermatozoa. Results were not significantly related to the parameters of semen analysis (sperm count, motility, standard morphology, viability, pH and volume, as well as fructose concentration and number of found cells) or to local sperm antibody testing and semen cultures. Fluorescence after AO staining was also not related to sperm functional capacity (evaluated using sperm-mucus interaction tests in vitro and in vivo), or the medical history of the patient. No significant differences in the AO test outcome were seen in patients with explained and unexplained infertility, or with regard to subsequent fertility [with a median value of 21% (range 5-46) green fluorescence in the fertile group, compared with a median value of 28% (range 9-81) green fluorescence in the other men]. The results of this prospective study indicate that under the usual conditions of conception, the AO test is not clinically useful as a screening procedure to determine semen quality during basic infertility investigation.  相似文献

The authors describe and discuss the new technique, multistep adaptive flux interpolation (MAFI), and its application to image data for coding. When applied to an image, MAFI produces an output which is also in an image form, but which has a more uniform feature density and a greatly reduced size. MAFI warps the input image by removing those rows and columns which contain a majority of redundant pixels. The side information required for reconstruction is minimal, and the image can be further compressed using conventional coders, making the compression ratio even higher. Because of its warped nature, the MAFI output's statistics are also more consistent with the properties assumed by block-based discrete cosine transform (DCT) methods  相似文献

A unified theoretical method for the calculation of the radio capacity of multiple-access schemes such as FDMA (frequency-division multiple access), TDMA (time-division multiple access), CDMA (code-division multiple access) and SSMA (spread-spectrum multiple access) in noncellular and cellular mobile radio systems is presented for AWGN (additive white Gaussian noise) channels. The theoretical equivalence of all the considered multiple-access schemes is found. In a fading multipath environment, which is typical for mobile radio applications, there are significant differences between these multiple-access schemes. These differences are discussed in an illustrative manner revealing several advantages of CDMA and SSMA over FDMA and TDMA. Raman spectroscopic studies on photoreactive retinal proteins are comprehensively described, including the basic physics of Raman scattering and illustrative examples of the types of information on the structure and function of the retinal chromophore and its environment which can be obtained from the vibrational Raman spectra. In addition, practical advice and recipes are given which should enable the reader to plan and eventually perform a Raman experiment in a photolabile retinal protein. A dominant role is played by the resonance Raman (RR) experiment with visible laser excitation which selectively probes the retinal chromophore. Much discussion is devoted to bacteriorhodopsin (bR) and its photocycle as a paradigm for a light-induced reaction of a retinal protein. Various time-resolved techniques are described to study the temporal evolution of the bR chromophore by probing RR spectra of intermediate states. Vibrational Raman spectra are interpreted in terms of structure and structural changes of the chromophore. RR spectroscopic studies on halorhodopsin, sensory rhodopsin, and visual pigments are reported, as well as on modified proteins in which retinal analogues are incorporated, and on site-specific mutants. Results of ultraviolet RR experiments which selectively probe the aromatic side chains in the protein backbone are reported. In addition, a promising new technique of near-infrared Raman excitation is discussed. Finally, application of coherent anti-Stokes Raman spectroscopy (CARS) to retinal proteins is reported.  相似文献

Dynamic Programming (DP) applies to many signal and image processing applications including boundary following, the Viterbi algorithm, dynamic time warping, etc. This paper presents an array processor implementation of generic dynamic programming. Our architecture is a SIMD array attached to a host computer. The processing element of the architecture is based on an ASIC design opting for maximum speed-up. By adopting a torus interconnection network, a dual buffer structure, and a multilevel pipeline, the performance of the DP chip is expected to reach the order of several GOPS. The authors demonstrate pure self-starting mode-locked operation in a fiber Fabry-Perot cavity. The laser produces 1.6 ps transform-limited soliton pulses by means of nonlinear polarization evolution in conjunction with an intracavity fiber polarizer. This laser system can be used for the generation and study of soliton pulses in the picosecond regime. With a careful choice of the fiber dispersion and fiber length, the system should also be able to operate in the femtosecond regime. The advantage of the system over existing self-starting fiber soliton lasers is that an optical isolator (an expensive component) is not required. The replacement of the two mirrors with fiber reflection gratings would make this configuration a truly all-fiber device  相似文献
