A high-temperature superconductor (HTS) was used on magnetic resonance imaging (MRI) receiver coils to improve image quality because of its intrinsic low electrical resistivity. Typical HTS coils are surface coils made of HTS thin-film wafers. Their applications are severely limited by the field of view (FOV) of the surface coil configuration, and the improvement in image quality by HTS coil is also reduced as the ratio of sample noise to coil noise increases. Therefore, previous HTS coils are usually used to image small in vitro samples, small animals, or peripheral human anatomies. We used large-size HTS coils (2.5-, 3.5-, and 5.5-in mean diameter) to enhance the FOV and we evaluated their performance through phantom and human MR images. Comparisons were made among HTS surface coils, copper surface coils, and cool copper surface coils in terms of the signal-to-noise ratio (SNR) and sensitivity profile of the images. A theoretical model prediction was also used to compare against the experimental result. We then selected several human body parts, including the wrist, feet, and head, to illustrate the advantage of HTS coil over copper coil when used in human imaging. The results show an SNR gain of 200% for 5.5-in HTS coil versus same size copper coils, while for 2.5- and 3.5-in coils it is 250%. One- and two-dimensional miniaturized electrophoresis of proteins with native fluorescence detection
Miniaturized electrophoresis was successfully coupled with native fluorescence detection for direct analysis of proteins in one- and two-dimensional separations. The detection setup was based on direct observation of the UV-induced fluorescence of proteins using a CCD camera and a Hg (Xe) lamp for sample excitation. Protein mixtures were readily separated by size on a 1-cm segment of the one-dimensional gel in 8 min, and a detection limit of 0.04 ng per band was achieved. The dynamic range of the system was larger than 2 orders of magnitude. Miniaturized slab gel electrophoresis was performed on a special holder designed to couple isoelectric focusing with SDS-PAGE. Two-dimensional separation, including rehydration of IEF strip and fluorescence detection was completed in 2.5 h. Approximately 200 protein spots from Escherichia coli were detected on a 1 cm(2) area. A detection limit of 0.1 microg of total protein was achieved. A simple way to selectively isolate peptides based on their isoelectric points (pI) for MALDI mass spectral analysis is described. An applied voltage was used to electromigrate peptides into a capillary. The capillary was modified with a zwitterionic surfactant, 1,2-dilauroyl-sn-phosphatidylcholine (DLPC), to suppress the electroosmotic flow (EOF) during injection. Hence, either the cationic or the anionic peptides were introduced, depending on the voltage polarity. By controlling the pH, selective loading of peptides was performed to isolate trace components from a mixture. The injected sample plugs were subsequently spotted in nanoliter volumes for MALDI-MS analysis. No significant sample losses resulting from selective sampling were detected. Low attomole-level detection of peptides (adrenocorticotropic hormone fragment 18-39, pI 4.25) was achieved from a mixture containing other peptides (angiotensin I, pI 6.92, and bradykinin, pI 12.00) at 100 000-fold higher concentrations. 相似文献

The effects of intra-arterial infusion of E. coli endotoxin at 1.0 mg. per minute on the gastric total and mucosal blood flows, electrical potential difference, and ionic fluxes across the gastric mucosa were studied in an exteriorized, chambered preparation of canine fundic stomach. Gamma-labelled microsphere technique was used in addition to venous drainage and plasma aminopyrine clearance for the measurement of total and mucosal blood flow, respectively. In spite of normal systemic blood pressure throughout the experiment, E. coli endotoxin infusion caused a significant decrease in total gastric blood flow and in the fractional distribution of flow to the mucosae. There was no significant arteriovenous shunting of microspheres. Significant reduction in potential difference and hydrogen-ion back diffusion also was noted after endotoxin infusion, possibly as a consequence of reduced mucosal blood flow. The results indicate that significant gastric mucosal ischemia can occur and may represent a mechanism in the development of gastric erosions in endotoxemia, even in the absence of systemic hypotension. 相似文献

The sensitivity of a neural network's output to its input perturbation is an important issue with both theoretical and practical values. In this article, we propose an approach to quantify the sensitivity of the most popular and general feedforward network: multilayer perceptron (MLP). The sensitivity measure is defined as the mathematical expectation of output deviation due to expected input deviation with respect to overall input patterns in a continuous interval. Based on the structural characteristics of the MLP, a bottom-up approach is adopted. A single neuron is considered first, and algorithms with approximately derived analytical expressions that are functions of expected input deviation are given for the computation of its sensitivity. Then another algorithm is given to compute the sensitivity of the entire MLP network. Computer simulations are used to verify the derived theoretical formulas. The agreement between theoretical and experimental results is quite good. The sensitivity measure can be used to evaluate the MLP's performance. 相似文献

We studied the aggregation of a rod-shaped bacteria, Bifidobacterium infantis, during capillary electrophoresis (CE). A microscope with an intensified CCD camera was employed to monitor the migration and aggregation of bacteria, which are labeled with fluorescent dye Syto 9 and excited with a 488-nm argon ion laser. A collision-based aggregation mechanism is proposed, in which collisions between microbes result from different mobilities and migration directions in the electric field. Individual microbes are aligned differently with respect to the direction of the electric field and exhibit different drag coefficients. The long-range forces include van der Waals attraction and electrostatic repulsion as qualitatively described by DLVO theory. Collisions in CE produce sufficient energy to overcome electrostatic repulsion, thus improving the efficiency of aggregation. This is supported by the fact that higher electric fields always resulted in faster aggregation. Also, when sodium phosphate buffer was used, increasing the ionic strength resulted in faster aggregation. However, when Tris-boric acid-EDTA (TBE, pH 9.1) buffer was used, the aggregation speed decreased when the ionic strength increased. We attribute this to the change of the surface of the bacteria at high borate and EDTA concentration, such as the loss of polysaccharides or the presence of complexation. This reduces the hydrophobicity of the surface and, thus, the short-range attractive forces. The addition of 0.05% poly(ethylene oxide) (PEO) into high ionic strength TBE buffer increased the aggregation rate. This can be attributed to the bridging effect of PEO between microbes. Further increase in the concentration of polymer reduced the aggregation rate, especially when the electric field was low, due in part to the increase in viscosity. The Motivation Orientation scales of the Inventory of School Motivation (ISM) were validated across Navajo (n = 760) and Anglo (n = 1012) students in the U.S. Confirmatory factor analysis (CFA) supported the 8-factor structure of motivation orientations for the total sample and the Navajo and Anglo subsamples, although Navajo students did not distinguish well between the Effort and Task constructs. However, of 39 survey items, only 30 items were invariant across groups in factor loadings on respective a priori constructs. The findings show that even though the ISM Motivation Orientation scales are applicable to students of different cultural backgrounds, meaningful cross-cultural comparisons should use the 30 items that mean the same to both cultural groups; whereas studies that do not involve cross-cultural comparisons may use the complete version of the scales. 相似文献

This study evaluated the physiological demands and low back neuromuscular fatigue patterns following a daytime work shift of 21 female nurses working in geriatric wards. Subjects' heart rate (HR) at work was monitored for 8 h and surface electromyogram (EMG) of their back muscle was recorded during a 1-minute horizontal trunk holding test before and after work. Results showed that the nurses had heart rates exceeding 90 beats/min in 57% and 110 beats/min in 19% of their working hours. The EMG revealed a significant drop in initial median frequency (MF) (p = 0.04) and increase in negative slope of the MF/time plot (p<0.001) of the back muscle after work. These suggest that the muscles were fatigued after work. The nurses rated patient lifting, transfer and turning as most physically demanding, and these corresponded to the highest heart rates recorded. The physiological and neuromuscular strain on the nurses indicated that extreme care should be exercised at work, particularly towards the end of a work shift when the muscles have become fatigued. 相似文献
