Δ9-tetrahydrocannabinol (THC) and merthiolate have been utilized as lysophospholipid acyltransferase inhibitors in metabolic studies. However, their effects on acyltransferases other than lysophosphatidylcholine:acyl-CoA acyltransferase (LPCAT) are not known. We have therefore investigated the effectiveness of THC and merthiolate in inhibiting the acylation of lysophosphatidylcholine, lysophosphatidylethanolamine, lysophosphatidylserine, lysophosphatidylinositol (LPI) and lysophosphatidic acid (LPA) in guinea pig liver microsomes using oleoyl-CoA and arachidonoyl-CoA as acyl donors. THC inhibited LPCAT and lysophosphatidylethanolamine: acyl-CoA acyltransferase (LPEAT) by 40–50%, but had no effect or only slightly increased the activities of the other acyltransferases when assayed with oleoyl-CoA as the acyl donor. The results obtained with arachidonoyl-CoA were similar to those with oleoyl-CoA, with the exception of a 40% inhibition of lysophosphatidylserine:acyl-CoA acyltransferase (LPSAT) at concentrations of 50 μM or higher. At similar concentrations, merthiolate was more effective than THC in inhibiting the acyltransferases examined. Selective effects on the acyltransferases were observed at low concentrations of merthiolate (20 μM or less). Thus, LPCAT was most susceptible, followed by LPI acyltransferases, LPSAT, LPEAT and lysophosphatidic acid:acyl-CoA acyltransferases (LPAAT). The presence of LPA did not affect the inhibition of LPCAT by merthiolate. Thus the resilience of LPAAT to merthiolate inhibition was not due to chelation of the compound by the acidic lysolipid. Thiol reagents includingN-ethylmaleiamide, 5,5′-dithio-bis-nitrobenzoic acid, iodoacetate, β-mercaptoethanol and dithiothreitol had little or no effect on the acyltransferases relative to equimolar concentrations of merthiolate. The above results indicate that merthiolate is a much more effective inhibitor of lysophospholipid:acyl-CoA acyltransferases than is THC, and that the selectivity exhibited by merthiolate may be due to direct and specific interaction with the acyltransferases.  相似文献

However, some studies have demonstrated age-related sparing on tasks where individuals have substantial experience, often attained over many decades. Here, the authors examined whether decades of experience in a fast-paced demanding profession, air traffic control (ATC), would enable older controllers to perform at high levels of proficiency. The authors also investigated whether older controllers would show diminished age-related decrements on domain-relevant cognitive abilities. Both young and old controllers and noncontrollers performed a battery of cognitive and ATC tasks. Results indicate that although high levels of experience can reduce the magnitude of age-related decline on the component processes that underlie complex task performance, this sparing is limited in scope. More important, however, the authors observed experience-based sparing on simulated ATC tasks, with the sparing being most evident on the more complex air traffic control tasks. These results suggest that given substantial experience, older adults may be quite capable of performing at high levels of proficiency on fast-paced demanding real-world tasks. Heat transfer to constant-property, fully developed, laminar flows in circular-segment ducts with uniform wall temperature (T) has been analyzed. Besides representing a compact surface, the segment duct geometry models the flow cross section of a circular tube with a straight-tape insert. Two variations in the T thermal boundary condition are considered: constant axial and circumferential wall temperature, and constant temperature on the curved surface but an adiabatic flat wall. These two conditions model the extremes of the fin effects of a straight-tape insert, i.e., 100% and zero fin efficiencies, respectively. Numerical solutions, obtained by using finite difference techniques, are presented for both the velocity and temperature fields. The isothermal friction factors are in excellent agreement with analytical solutions reported in the literature. The Nusselt number results for the two thermal boundary conditions are presented for different segment shapes, 0° ≤, 6 ≤, 90°, and they represent the lower limits of the heat transfer enhancement due to twisted-tape inserts.  相似文献

This publication is a case study of the seasonal variability of mine water drainage from the Saint Louis Tunnel (SLT) at the inactive Rico-Argentine mine site located in southwestern Colorado. It is an introductory paper for the two passive water treatment system technology evaluations contained in this issue. Mine water chemistry changes from baseflow to a snowmelt runoff event (SMRE) where snowmelt runoff follows preferential migration pathways to flush acidic weathering products from the upper mine workings to the SLT. Baseflow mine drainage is characterized as circumneutral, with Zn, Cd, Mn, and Ni concentrations primarily in the dissolved form. Dissolved Zn, Mn, Fe, and potentially Cd illustrate equilibrium with carbonate minerals. Total concentrations of Fe, Cu, Pb, and As are primarily in the suspended form and suggest sorption to Fe oxides. Mine water chemistry during the SMRE reflects mixing of circumneutral baseflow waters with more acidic waters flushing the upper mine workings. Geothermal activity provides for a consistently warm mine water discharge from the SLT. The two seasons that provide the most challenge to passive water treatment of SLT mine drainage are the SMRE period and the low flow stage of the Dolores River. Mine water flow and chemistry during SMRE are highly correlated with Dolores River flow and this site conceptual model was and will be used to assist in pilot project evaluation, water treatment system design, monitoring system design, a seasonal compliance approach, and water management.

Psychoanalysis has been in a constant uninterrupted debate about its identity as a discipline and as a social institution. This article considers the place of science in psychoanalysis, on the one hand, and the hermeneutic nature of our discipline, on the other. The aim is to articulate a typology of psychoanalytic knowledge that characterizes psychoanalysis as a form of therapy, an intellectual movement, and a theoretical system. This typology considers psychoanalysis as a thought collective that influences its members by exchanging and maintaining ideas. To be a well-rounded psychoanalytic thinker or practitioner one must be able to move easily among three realms of knowledge--the humanities, the social sciences, and the natural sciences. Successful mobility requires appropriate decision-making. Seniors with reduced executive functioning—such as senior fallers—may be prone to poor mobility judgments, especially under dual-task conditions. We classified participants as “At-Risk” and “Not-At-Risk” for falls using a validated physiological falls-risk assessment. Dual-task performance was assessed in a virtual reality environment where participants crossed a simulated street by walking on a manual treadmill while listening to music or conversing on a phone. Those “At-Risk” experienced more collisions with oncoming cars and had longer crossing times in the Phone condition compared to controls. IDH (isocitrate dehydrogenase) mutation, hypoxia, and neo-angiogenesis, three hallmarks of diffuse gliomas, modulate the expression of small non-coding RNAs (miRNA). In this paper, we tested whether pro-angiogenic and/or pro-hypoxic miRNAs could be used to monitor patients with glioma. The miRNAs were extracted from tumoral surgical specimens embedded in the paraffin of 97 patients with diffuse gliomas and, for 7 patients, from a blood sample too. The expression of 10 pro-angiogenic and/or pro-hypoxic miRNAs was assayed by qRT-PCR and normalized to the miRNA expression of non-tumoral brain tissues. We confirmed in vitro that IDH in hypoxia (1% O2, 24 h) alters pro-angiogenic and/or pro-hypoxic miRNA expression in HBT-14 (U-87 MG) cells. Then, we reported that the expression of these miRNAs is (i) strongly affected in patients with glioma compared to that in a non-tumoral brain; (ii) correlated with the histology/grade of glioma according to the 2016 WHO classification; and (iii) predicts the overall and/or progression-free survival of patients with glioma in univariate but not in a multivariate analysis after adjusting for sex, age at diagnosis, and WHO classification. Finally, the expression of miRNAs was found to be the same between the plasma and glial tumor of the same patient. This study highlights a panel of seven pro-angiogenic and/or pro-hypoxic miRNAs as a potential tool for monitoring patients with glioma.  相似文献

Single crystals of CaO-stabilized ZrO2 containing between 15.3 and 18.9 mol% CaO were heat-treated for 5000 h at 1200°C to study the stability of the ordered defect-fluorite phase, CaZr4O91). Subsequent TEM analysis of the equiaxed φ1 domains in samples richer than approximately 18 mol% in CaO showed a random distribution of φ1 variants, with no preferred interfacial habit planes. A critical review of the literature, combined with the new data, supports the Stubican-Hellmann-Hannon version of the phase diagram in the region 15 to 26 mol% CaO and 1000° to 1400°C and strongly suggests that φ1 is a stable phase in the ZrO2-CaO systems.  相似文献
