The impact of extreme low flows on the water quality of the Lower Murray River and Lower Lakes (Alexandrina and Albert) in South Australia was assessed by comparing water quality from five sites during an extreme low flow period (March 2007?CNovember 2009) and a preceding reference period (March 2003?CNovember 2005). Significant increases in salinity, total nitrogen, total phosphorus, chlorophyll a and turbidity were observed in the Lower Lakes during the low flow period. Consequently, water quality guidelines for the protection of aquatic ecosystems were greatly exceeded. Principal Component Analysis, empirical and mass balance model calculations suggested these changes could be attributed primarily to the lack of flushing resulting in concentration of dissolved and suspended material in the lakes, and increased sediment resuspension as the lakes became shallower. The river sites also showed significant but more minor salinity increases during the extreme low flow period, but nutrient and turbidity concentrations decreased. The most plausible reasons for these changes were decreased catchment inputs and increased influence of saline groundwater inputs. The results highlight the vulnerability of arid and semi-arid lake systems to reduced flow conditions as a result of climatic changes and/or water management decisions.  相似文献

During development and tissue repair, progenitor cells are guided by both biochemical and biophysical cues of their microenvironment, including topographical signals. The topographical cues have been shown to play an important role in controlling the fate of cells. Systematic investigation of topographical structures with different geometries and sizes under the identical experimental conditions on the same chip will enhance the understanding of the role of shape and size in cell–topography interactions. A simple customizable multi‐architecture chip (MARC) array is therefore developed to incorporate, on a single chip, distinct topographies of various architectural complexities, including both isotropic and anisotropic features, in nano‐ to micrometer dimensions, with different aspect ratios and hierarchical structures. Polydimethylsiloxane (PDMS) replicas of MARC are used to investigate the influence of different geometries and sizes in neural differentiation of primary murine neural progenitor cells (mNPCs). Anisotropic gratings (2 μm gratings, 250 nm gratings) and isotropic 1 μm pillars significantly promote differentiation of mNPCs into neurons, as indicated by expression of β‐III‐tubulin (59%, 58%, and 58%, respectively, compared to 30% on the control). In contrast, glial differentiation is enhanced on isotropic 2 μm holes and 1 μm pillars. These results illustrate that anisotropic topographies enhance neuronal differentiation while isotropic topographies enhance glial differentiation on the same chip under the same conditions. MARC enables simultaneous cost‐effective investigation of multiple topographies, allowing efficient optimization of topographical and biochemical cues to modulate cell differentiation.  相似文献

Parallel conductance (electric current flow through surrounding tissue) is an important determinant of accurate measurements of arterial lumen diameter, using the conductance method. The present study is focused on the role of non-uniform geometrical/electrical configurations of surrounding tissue, which are a primary source of electric current leakage. Computational models were constructed to simulate the conductance catheter measurement with two different excitation electrodes spacings (i.e. 12 and 20 mm for coronary and peripheral sizing, respectively) for different vessel–tissue configurations: (i) blood vessel fully embedded in muscle tissue, (ii) blood vessel superficially embedded in muscle tissue, and (iii) blood vessel superficially embedded in muscle tissue with fat covering half of the arterial vessel (anterior portion). The simulations suggest that the parallel conductance and accuracy of measurement is dependent on the inhomogeneous/anisotropic configuration of surrounding tissue, including the asymmetric dimension and anisotropy in electrical conductivity of surrounding tissue. Specifically, the measurement was shown to be accurate as long as the vessel was superficial, regardless of the considerable total surrounding tissue dimension for coronary or peripheral arteries. Moreover, it was shown that the unfavourable impact of parallel conductance on the accuracy of conductance catheter measurement is decreased by the combination of a lower transverse electrical conductivity of surrounding muscle tissue, a smaller electrode spacing and a larger lumen diameter. The present findings confirm that the conductance catheter technique provides an accurate platform for sizing of clinically relevant (i.e. superficial and diseased) arteries.  相似文献

Bone degradation by osteoclasts depends on the formation of a sealing zone, composed of an interlinked network of podosomes, which delimits the degradation lacuna into which osteoclasts secrete acid and proteolytic enzymes. For resorption to occur, the sealing zone must be coherent and stable for extended periods of time. Using titanium roughness gradients ranging from 1 to 4.5 µm Ra as substrates for osteoclast adhesion, we show that microtopographic obstacles of a length scale well beyond the range of the ‘footprint’ of an individual podosome can slow down sealing-zone expansion. A clear inverse correlation was found between ring stability, structural integrity and sealing-zone translocation rate. Direct live-cell microscopy indicated that the expansion of the sealing zone is locally arrested by steep, three-dimensional ‘ridge-like barriers’, running parallel to its perimeter. It was, however, also evident that the sealing zone can bypass such obstacles, if pulled by neighbouring regions, extending through flanking, obstacle-free areas. We propose that sealing-zone dynamics, while being locally regulated by surface roughness, are globally integrated via the associated actin cytoskeleton. The effect of substrate roughness on osteoclast behaviour is significant in relation to osteoclast function under physiological and pathological conditions, and may constitute an important consideration in the design of advanced bone replacements.  相似文献

We report on the preparation of the first material for therapeutic delivery of CO. A peptide amphiphile was synthesized with a covalently attached ruthenium tricarbonyl. Self-assembled nanofiber gels containing this peptide spontaneously released CO with prolonged release kinetics compared to soluble CO donors. Oxidatively stressed cardiomyocytes had improved viability when treated with this peptide, demonstrating its potential as a biodegradable gel for localized therapeutic CO delivery.  相似文献

A recent study on the environmental impact of automatic fire sprinklers is documented in a two part series. The present paper examines the relationship of automatic fire sprinkler technology to environmental sustainability. Large-scale fire tests were conducted using identically constructed and furnished living rooms. In one test, fire extinguishment was achieved solely by fire service intervention, and in the other, a single residential automatic fire sprinkler was used to control the fire until final extinguishment was achieved by the fire service. Comparisons of the total greenhouse gas production, quantity of water required to extinguish the fire, quality of water runoff, potential impact of wastewater runoff on groundwater and surface water, and mass of materials requiring disposal between the two tests were made. The results show that in addition to providing life safety and limiting property damage, the use of automatic fire sprinklers is a key factor in achieving sustainability.  相似文献

A fuel cell using an enzymatic biocathode operating in a gas phase mode is reported. The electrode was prepared using a three-dimensional conductive electrode matrix. An enzyme solution containing laccase and a mediator was distributed into a hydrophilic matrix of carbon felt fibers creating a porous gas-flowing electrode. A Pt-based gas diffusion electrode served as the anode. A maximum power density of 9.4 W m−2 (2.9 kW m−3) was obtained with 15 U of enzyme cm−2, with hydrogen as the fuel. Power density was found to be a function of the enzyme loading, air flow rate, volume of the liquid phase and the humidity of the air stream. The ability to use methanol and ethanol as vapors in gas phase was also shown. The introduction of three-dimensionality into the electrode architecture and operation of the fuel cell in a gas phase mode to supply the fuel and the oxidant demonstrates an avenue for improving the power density of EFCs.  相似文献

In association with the Department of Energy–funded Position of Offshore Wind Energy Resources (POWER) project, we present results from compositing a 3‐year dataset of 80‐m (above ground level) wind forecasts from the 3‐km High‐Resolution Rapid Refresh (HRRR) model over offshore regions for the contiguous United States. The HRRR numerical weather prediction system runs once an hour and features hourly data assimilation, providing a key advantage over previous model‐based offshore wind datasets. On the basis of 1‐hour forecasts from the HRRR model, we highlight the different climatological regimes of the nearshore environment, characterizing the mean 80‐m wind speed as well as the frequency of exceeding 4, 12, and 25 m s?1 for east and west coast, Gulf of Mexico, and Great Lake locations. Preliminary verification against buoy measurements demonstrates good agreement with observations. This dataset can inform the placement of targeted measurement systems in support of improving resource assessments and wind forecasts to advance offshore wind energy goals both in New England and other coastal regions of the United States.  相似文献
