In river systems, high‐head dams may increase the distance‐decay of fish community similarity by creating nearly impermeable dispersal barriers to certain species from upstream reaches. Substantial evidence suggests that migratory species are impacted by dams, and most previous studies in stream/river networks have focused on small streams and headwaters. Here, we assess whether a high‐head dam (Lock and Dam 19; LD 19) on a large river, the Upper Mississippi River (UMR), substantially alters fish community structure relative to variability expected to occur independent of the dam's effect as a fish dispersal barrier. Using fish catch per unit effort data, we modelled the distance‐decay function for the UMR fish community and then estimated the similarity that would be expected to occur across LD19 and compared it with measured similarity. Measured similarity in the fish community above and below LD19 was close to the expected value based on the distance‐decay function, suggesting LD19 does not create an abrupt transition in the fish community. Although some migratory fish species no longer occur above LD19 (e.g., skipjack herring, Alosa chrysochloris), these species do not occur in high abundance below the dam and so do not drive variation in fish community structure. Instead, much of the variation in species structure is driven by the loss/gain of species across the latitudinal gradient. Lock and Dam 19 does not appear to be a clear transition point in the river's fish community, although it may function as a meaningful barrier for particular species (e.g., invasive species) and warrant future attention from a management perspective. 相似文献

The fixation of cells of petiolar hairs of tomato was monitored by phase contrast and Nomarski interference contrast microscopy. Because the cuticle covering these hairs is relatively impermeable some fixatives enter only the base of the hair so that a ‘fixation front’ can be followed through a single cell. The basal end of such a cell may be immobilized by the fixative while the apical end is still streaming. A variety of fixatives were tested at room temperature and at 273–278 K. All procedures tested failed to stabilize a pleomorphic canalicular system that is a dominant feature of these cells so that no clue to the presence of this system can be seen in sectioned material. It is therefore very desirable that images of fixed cells be compared to the structure of similar cells which are still alive. 相似文献

Green fluorescent protein (GFP) has many applications as a marker in living cells, and has become widely used as a reporter gene in microbial, plant and animal cells. Screening microbial colonies for GFP expression enables various types of assays (e.g. for secretory mutations). However, this is laborious, non-quantitative and potentially hazardous to the operator (due to UV illumination) when performed manually. In order to address this the GloPix robot was developed. The imaging system discriminates between colonies based on the level of fluorescence activity and the picking function automatically transfers cells to microplate wells. In applications in which Doppler processing is not possible, such as side-looking intravascular imaging systems, decorrelation methods can be used to estimate blood speed. Here, a method is presented measuring relative blood speed using an FIR filter bank to estimate temporal decorrelation rates. It can be implemented in a modern commercially available ultrasound imaging system with no additional hardware. Both simulations and experiments using an intraluminal scanner appropriate for coronary artery applications have tested the system. In this study, the FIR filter bank is contrasted with previous methods, and its utility is further demonstrated with real-time color flow images from a pig model. 相似文献

The chicken lysozyme silencer F2 (F2) thyroid hormone response element (TRE) contains an unusual everted palindromic arrangement, has a high affinity for thyroid hormone receptor (TR) homodimers, and is especially sensitive to dominant negative inhibition by, the T3 resistance (RTH) mutant TR beta P453H. We used various TREs and TR mutations to determine the mechanisms for this sensitivity. Changing the F2 orientation from an everted palindrome to a direct repeat with a 4-bp gap (DR+4) (F2-DR) decreased the sensitivity to inhibition at high T3 concentrations, while a loss of this sensitivity occurred with a palindromic arrangement of these same half-sites. F2 contains the dinucleotide TG 5' to each consensus half-site conforming to the optimal TR-binding octamer, YRRGGTCA. A T to A change in position 1 of both F2 half-sites markedly reduced T3-induction, yet only slightly reduced TR homodimer or TR-retinoid X receptor (RXR) heterodimer binding. The TR beta ninth heptad mutation, L428R, prevents TR heterodimerization with RXR and eliminates the inhibitory effect of the P453H mutant TR on the F2-DR, but not the F2 element. Structural features of a TRE that favor strong TR binding of both TR homodimers and TR-RXR heterodimers containing the mutant TR, such as the everted palindromic conformation or the optimal TR-binding consensus octamer, enhance the sensitivity of a TRE to inhibition by the mutant TR. Thus, both half-site orientation and sequence contribute to the sensitivity of a given TRE to dominant negative inhibition by a mutant TR. 相似文献

Hole concentration and mobility profiles have been measured as a function of dose for 450 keV zinc ions implanted into GaAs. For doses above 1014 ions/cm2 implanted at 200°C very broad profiles were obtained suggesting that significant indiffusion has occurred. Evaporated films of tantalum+tantalum oxide, 500 Å thick, were implanted with argon, oxygen and nitrogen ions. The phase structure of the films was determined by transmission electron microscopy and the dose dependence of resistivity and thermal coefficient of resistivity was satisfactorily explained in terms of the precipitation of compounds formed in the film. Argon bombardment had little effect on the structure of a film until sputter etching had reduced the film thickness considerably, when precipitates of TaO2 in the form of rounded islands were created. Oxygen bombardment resulted in the precipitation of the b.c.c. phase of tantalum, and at higher doses precipitates of TaO2 were formed. Nitrogen bombardment resulted in a sudden phase change and the precipitation of Ta4N5 in the form of hexagonal platelets; the conduction process in these films was highly activated, probably as a result of the presence of an amorphous oxide matrix. The dependence of changes in electrical properties on oxygen content of the film is explained by this phase structure. 相似文献

Thin films of triglycine sulfate were prepared by laser evaporation. The properties of these films were evaluated by electrical measurements, x-ray diffraction and IR spectrometry. The deposited material was found to be essentially triglycine sulfate with some glycine present as a decomposition product.
