This work deals with the production and characterization of water-compatible, iron oxide based nanoparticles covered with functional poly(ethylene glycol) (PEG)-biotin surface groups (SPIO-PEG-biotin). Synthesis of the functionalized colloids occurred by incubating the oleate coated particles used as precursor magnetic fluid with anionic liposomes containing 14?mol% of a phospholipid-PEG-biotin conjugate. The latter was prepared by coupling dimyristoylphosphatidylethanolamine (DC(14:0)PE) to activated α-biotinylamido-ω -N-hydroxy-succinimidcarbonyl-PEG (NHS-PEG-biotin). Physical characterization of the oleate and PEG-biotin iron oxide nanocolloids revealed that they appear as colloidal stable clusters with a hydrodynamic diameter of 160?nm and zeta potentials of -?39?mV (oleate coated particles) and -?14?mV (PEG-biotin covered particles), respectively, as measured by light scattering techniques. Superconducting quantum interference device (SQUID) measurements revealed specific saturation magnetizations of 62-73?emu?g(-1) Fe(3)O(4) and no hysteresis was observed at 300?K. MR relaxometry at 3?T revealed very high r(2) relaxivities and moderately high r(1) values. Thus, both nanocolloids can be classified as small, superparamagnetic, negative MR contrast agents. The capacity to functionalize the particles was illustrated by binding streptavidin alkaline phosphatase (SAP). It was found, however, that these complexes become highly aggregated after capturing them on the magnetic filter device during high-gradient magnetophoresis, thereby reducing the accessibility of the SAP.  相似文献

Seven stable austenitic steels (stable with respect to γ → α′ transformation at room temperature) of different alloy compositions (18Cr–12.5Ni, 18Cr–35Ni, 18Cr–8Ni–6Mn–0.25N, 0.6C–23Mn, 1.3C–12Mn, 1C–31Mn–9Al, 18Cr–19Mn–0.8N) were tensile tested in high-pressure hydrogen atmosphere to assess the role of austenite stability on hydrogen environment embrittlement (HEE). The influence of hydrogen on tensile ductility was small in steels that are believed to have a high initial portion of dislocation cross slip (18Cr–12.5Ni, 18Cr–35Ni, 18Cr–8Ni–6Mn–0.25N), while the effects of hydrogen were significantly greater in steels with other primary deformation modes (planar slip in 18Cr–19Mn–0.8N and 1C–31Mn–9Al or mechanical twinning in 0.6C–23Mn and 1.3C–12Mn) despite comparable austenite stability at the given test conditions. It appears that initial deformation mode is one important parameter controlling susceptibility to HEE and that martensitic transformation is not a sufficient explanation for HEE of austenitic steels.  相似文献

The abuse of anabolic substances in animal husbandry is forbidden within the EU and well controlled by detecting substance residues in different matrices. The application of newly designed drugs or substance cocktails represents big problems. Therefore developing sensitive test methods is important. The analysis of physiological changes caused by the use of anabolic agents on the molecular level, for example, by quantifying gene expression response, is a new approach to develop such screening methods. A novel technology for holistic gene expression analysis is RNA sequencing. In this study, the potential of this high-throughput method for the identification of biomarkers was evaluated. The effect of trenbolone acetate plus estradiol on gene expression in liver from Nguni heifers was analyzed with RNA sequencing. The expression of 40 selected candidate genes was verified via RT-qPCR, whereby 20 of these genes were significantly regulated. To extract the intended information from these regulated genes, biostatistical tools for pattern recognition were applied and resulted in a clear separation of the treatment groups. Those candidate genes could be verified in boars and in calves treated with anabolic substances. These results show the potential of RNA sequencing to screen for biomarker candidates to detect the abuse of anabolics. The verification of these biomarkers in boars and calves leads to the assumption that gene expression biomarkers are independent of breed or even species and that biomarkers, identified in farm animals could also act as potential biomarker candidates to detect the abuse of anabolic substances in human sports.  相似文献

Here, design of the first pathogen‐mimicking metal oxide nanoparticles with the ability to enter cancer cells and to selectively target and activate the TLR9 pathway, and with optical and MR imaging capabilities, is reported. The immobilization of ssDNA (CpG ODN 2006) on MnO nanoparticles is performed via the phosphoramidite route using a multifunctional polymer. The multifunctional polymer used for the nanoparticle surface modification not only affords a protective organic biocompatible shell but also provides an efficient and convenient means for loading immunostimulatory oligonucleotides. Since fluorescent molecules are amenable to photodetection, a chromophore (Rhodamine) is introduced into the polymer chain to trace the nanoparticles in Caki‐1 (human kidney cancer) cells. The ssDNA coupled nanoparticles are used to target Toll‐like receptors 9 (TLR9) receptors inside the cells and to activate the classical TLR cascade. The presence of TLR9 is demonstrated independently in the Caki‐1 cell line by western blotting and immunostaining techniques. The magnetic properties of the MnO core make functionalized MnO nanoparticles potential diagnostic agents for magnetic resonance imaging (MRI) thereby enabling multimodal detection by a combination of MR and optical imaging methods. The trimodal nanoparticles allow the imaging of cellular trafficking by different means and simultaneously are an effective drug carrier system.  相似文献

The reductive transformation of p-cyanonitrobenzene (pCNB) was investigated in laboratory batch slurries exhibiting dominant terminal electron accepting processes (TEAPs). Pseudo-first-order rate constants (k(obs)) were measured for the reduction of pCNB in nitrate-reducing, iron-reducing, sulfate-reducing, and methanogenic sediment slurries. Reduction was extremely slow in nitrate-reducing slurries but increased in slurries exhibiting TEAPs with significant concentrations of solution phase Fe(ll). As the reduction of pCNB progressed in the Fe(ll) rich systems, significant but nonstoichiometric decreases in aqueous Fe(ll) concentration were measured. Normalization of k(obs) to initial aqueous Fe(ll) concentrations (k(obs)/[Fe(ll)]t=0) gave values ranging from 0.0040 to 0.0052 d(-1) microM(-1) for nitrate-reducing, iron-reducing, and methanogenic sediment slurries as well as sulfate-reducing sediment slurries in which lactate served as a source of organic carbon. The k(obs)/ [Fe(ll)]t=0 ratios were 1-fold greater for sulfate-reducing batch slurries amended with acetate and iron-reducing slurries equilibrated with a 3% H2 atmosphere indicating that the electron source and system parameters such as pH play a determinant role in the reaction kinetics. Although these data demonstrate that aqueous phase Fe(ll) must be present for significant reduction to occur, a limited role for aqueous phase Fe(ll) as a quantitative indicator of reactivity is suggested.  相似文献

A technology for effecting diagenesis-like transformations in young natural organic matter (NOM) derived from common plant materials is described. Thirteen such materials were processed in liquid-phase water at superheated temperature/pressure conditions. In all cases significant changes in the physical and chemical properties of the raw NOM source materials occurred. Their carbon and nitrogen contents increased, their hydrogen and oxygen contents decreased, their surface areas initially increased dramatically, then sharply decreased, and their bulk densities and equilibrated aqueous phase pH values increased as functions of increasing temperature and pressure. Spectroscopic analyses confirmed marked changes in the molecular structures of the NOMs resulting from superheated water processing; e.g., fractions of alkyl and aromatic carbons increased while oxygen-associated functional organic carbon decreased. These changes in elemental composition and molecular structure indicate that the organic fractions of the raw materials became less polar, increasingly condensed, and more aromatic as a result of the reactions induced by superheated water processing, the changes correlating directly with the temperature/ pressure conditions employed. The results presented confirm that the long-term biogeochemical processes that in nature effect geologically slow advances in the chemical states of organic carbon can be simulated and markedly accelerated by superheated water processing.  相似文献

Chemical speciation modeling is a vital tool for assessing the bioavailability of inorganic species, yet significant uncertainties in thermodynamic parameters and model form limit its potential for decision-making. In this paper we present a novel method for the quantification of thermodynamic parameter uncertainty and ionic strength correction model uncertainty using Bayesian Markov Chain Monte Carlo (MCMC) estimation methods. These methods allow for the inclusion of correlation modeling, which has not been present in previous work. The MCMC simulations are used to model a natural river water to determine the uncertainty in the calculated environmental speciation of ethylenediamenetetraacetate, a chelating agent that has attracted considerable environmental interest. The results indicate that incorporating correlation among related thermodynamic parameters into the uncertainty model is necessary to correctly quantify the overall system uncertainty. This result indicates the superiority of MCMC estimation methods overtraditional Monte Carlo methods when available data are used to estimate parameter uncertainty in systems with closely related model parameters.  相似文献

The aim of the study was to determine the influence of maturation and of cooking processes on water losses and on the vitamin B12 content of meat. Three types of muscle (Longissimus lumborum, Longissimus thoracis and Triceps brachii) were sampled from a total of 16 animals, representative of animals raised for meat production in France. Three durations of maturation were compared: 1, 3 and 14 days. Different cooking processes were applied: Longissimus lumborum was deep-fat fried or roasted, Longissimus thoracis was pan fried or grilled and Triceps brachii was braised. The cooking yield averaged 55-56% for Triceps brachii, 73-77% for Longissimus lumborum and 85-87% for Longissimus thoracis. Vitamin B12 concentration in raw meat was significantly higher in Triceps brachii than in Longissimus lumborum and Longissimus thoracis (20.86, 11.53 and 9.21ng/g wet tissue, in the same respective order). When expressed on a wet weight basis, all concentrations were significantly increased by cooking. When expressed on a lipid-free dry basis, significant losses in vitamin B12 were measured only in the braised Triceps brachii (-25%) and in the deep-fat fried Longissimus lumborum (-5.5%) as a result of long duration and high temperature of cooking, respectively. Maturation did not affect the vitamin B12 content of meat, whether raw or cooked.  相似文献
