We recently found that, in human osteoblasts, Homer1 complexes to Calcium-sensing receptor (CaSR) and mediates AKT initiation via mechanistic target of rapamycin complex (mTOR) complex 2 (mTORC2) leading to beneficial effects in osteoblasts including β-catenin stabilization and mTOR complex 1 (mTORC1) activation. Herein we further investigated the relationship between Homer1 and CaSR and demonstrate a link between the protein levels of CaSR and Homer1 in human osteoblasts in primary culture. Thus, when siRNA was used to suppress the CaSR, we observed upregulated Homer1 levels, and when siRNA was used to suppress Homer1 we observed downregulated CaSR protein levels using immunofluorescence staining of cultured osteoblasts as well as Western blot analyses of cell protein extracts. This finding was confirmed in vivo as the bone cells from osteoblast specific CaSR−/− mice showed increased Homer1 expression compared to wild-type (wt). CaSR and Homer1 protein were both expressed in osteocytes embedded in the long bones of wt mice, and immunofluorescent studies of these cells revealed that Homer1 protein sub-cellular localization was markedly altered in the osteocytes of CaSR−/− mice compared to wt. The study identifies additional roles for Homer1 in the control of the protein level and subcellular localization of CaSR in cells of the osteoblast lineage, in addition to its established role of mTORC2 activation downstream of the receptor. 相似文献

Plants recognize biotic challengers and respond with the appropriate defense by utilizing phytohormone signaling and crosstalk. Despite this, microbes and insects have evolved mechanisms that compromise the plant surveillance system and specific defenses, thus ensuring successful colonization. In nature, plants do not experience insect herbivores and microbes in isolation, but in combination. Over time, relationships have developed between insects and microbes, varying on a continuum from no-relationship to obligate relationships that are required for both organisms to survive. While many reviews have examined plant-insect and plant-microbe interactions and the mechanisms of plant defense, few have considered the interface where microbes and insects may overlap, and synergies may develop. In this review, we critically evaluate the requirements for insect-associated microbes to develop synergistic relationships with their hosts, and we mechanistically discuss how some of these insect-associated microbes can target or modify host plant defenses. Finally, by using bioinformatics and the recent literature, we review evidence for synergies in insect-microbe relationships at the interface of plant-insect defenses. Insect-associated microbes can influence host-plant detection and/or signaling through phytohormone synthesis, conserved microbial patterns, and effectors, however, microbes associated with insects must be maintained in the environment and located in opportunistic positions. 相似文献

This paper presents a practical approach to the robust speed control of electrical drives. The approach is based on sliding mode control (SMC), but it is shown how sampling time, system noise, actuation limit, and antiwindup integrator reduce the control to an online maximum-gain selection constrained by system noise. The paper proposes a trajectory-reference compensator and fuzzy gain interpolator to fully exploit the system actuation while avoiding chattering and remaining within specified noise limits, only the SMC switching line needs to be selected; the remaining control design is systematic and, thus, should be of interest to drive engineers in industry. We extend the SAFT‐VR+DE equation of state to describe 19 aqueous electrolyte solutions with both a fully dissociated and a partially dissociated model. The approach is found to predict thermodynamic properties such as the osmotic coefficient, water activity coefficient, and solution density, across different salt concentrations at room temperature and pressure in good agreement with experiment using only one or two fitted parameters. At higher temperatures and pressures, without any additional fitting, the theory is found to be in qualitative agreement with experimental mean ionic activities and osmotic coefficients. The behavior of the dielectric constant as a function of salt concentration is also reported for the first time using a statistical associating fluid theory (SAFT)‐based equation of state. A Monte Carlo sky coverage model for laser guide star adaptive optics systems was proposed by Clare and Ellerbroek [J. Opt. Soc. Am. A 23, 418 (2006)]. We refine the model to include (i) natural guide star (NGS) statistics using published star count models, (ii) noise on the NGS measurements, (iii) the effect of telescope wind shake, (iv) a model for how the Strehl and hence NGS wavefront sensor measurement noise varies across the field, (v) the focus error due to imperfectly tracking the range to the sodium layer, (vi) the mechanical bandwidths of the tip-tilt (TT) stage and deformable mirror actuators, and (vii) temporal filtering of the NGS measurements to balance errors due to noise and servo lag. From this model, we are able to generate a TT error budget for the Thirty Meter Telescope facility narrow-field infrared adaptive optics system (NFIRAOS) and perform several design trade studies. With the current NFIRAOS design, the median TT error at the galactic pole with median seeing is calculated to be 65 nm or 1.8 mas rms. 相似文献

Organic electronics technology, in which at least the semiconducting component of the integrated circuit is an organic material, offers the potential for fabrication of electronic products by low-cost printing technologies, such as ink jet, gravure offset lithography and flexography. The products will typically be of lower performance than those using the present state of the art single crystal or polysilicon transistors, but comparable to amorphous silicon. A range of prototypes are under development, including rollable electrophoretic displays, active matrix liquid crystal (LC) displays, flexible organic light emitting diode displays, low frequency radio frequency identification tag and other low performance electronics. Organic semiconductors that offer both electrical performance and stability with respect to storage and operation under ambient conditions are required. This work describes the development of reactive mesogen semiconductors, which form large crosslinked LC domains on polymerization within mesophases. These crosslinked domains offer mechanical stability and are inert to solvent exposure in further processing steps. Reactive mesogens containing conjugated aromatic cores, designed to facilitate charge transport and provide good oxidative stability, were prepared and their liquid crystalline properties evaluated. The organization and alignment of the mesogens, both before and after crosslinking, were probed by grazing incidence wide-angle X-ray scattering of thin films.
