To assess the fruit-specific determinants of pericarp browning, litchi pericarp was characterized in terms of appearance, the polyphenol pattern as specified by HPLC-DAD-MS n without and after thiolysis, and the activities of polyphenol oxidase (PPO) and peroxidase (POD) by exploring “Kwang Jao,” “O-Hia,” “Kim Cheng,” and “Chacapat” fruit on the respective harvest day, “Hong Huey” fruit also throughout 52 days of cold storage (5 °C, 95% relative humidity). At harvest, PPO activity was maximum for “Kim Cheng” pericarp (126 μkat/hg), whereas POD activity was striking for that of “O-Hia” (512 μkat/hg, including membrane-bound isoforms). Flavan-3-ol and proanthocyanidin patterns were consistent for all cultivars. However, cultivars with sharp-pointed and round–obtuse protuberances differed in pericarp anthocyanin and flavonol glycosylation patterns. The molar ratio of cyanidin 3-O-rutinoside to its glucoside was ≤6:1 for “Hong Huey” and “Kwang Jao,” but ≥43:1 for “Kim Cheng” and “Chacapat” pericarp. Long-term storage gave evidence of two key processes involved in pericarp browning: (1) PPO-mediated oxidation of abundant (?)-epicatechin (1.4–2.0 g/hg), resulting in dark brown pigments, and (2) microcrack-induced formation of light brown surface scurf, supposably with involvement of POD. Accordingly, an improved scheme for litchi pericarp browning was proposed. As regards recommendable postharvest concepts for each cultivar, “Chacapat” suited most for long-distance transports due to its overall low susceptibility to pericarp browning. Properties of “O-Hia” litchi, being prone to surface scurf formation, suggested preferred distribution via domestic markets. High contents of flavonols (e.g., quercetin glycosides, 166 mg/hg) and A-type-linked procyanidins (e.g., procyanidin A2, 1,092 mg/hg) qualified pericarp of “Hong Huey” litchi as raw material for polyphenol extracts exerting antioxidant properties.  相似文献

A meta-analysis of 117 studies evaluated the effects of behavior modeling training (BMT) on 6 training outcomes, across characteristics of training design. BMT effects were largest for learning outcomes, smaller for job behavior, and smaller still for results outcomes. Although BMT effects on declarative knowledge decayed over time, training effects on skills and job behavior remained stable or even increased. Skill development was greatest when learning points were used and presented as rule codes and when training time was longest. Transfer was greatest when mixed (negative and positive) models were presented, when practice included trainee-generated scenarios, when trainees were instructed to set goals, when trainees' superiors were also trained, and when rewards and sanctions were instituted in trainees' work environments. Three experiments investigated the aging of implicit spatial and spatiotemporal context learning in 2 tasks. In contextual cuing, people learn to use repeated spatial configurations to facilitate search for a target, whereas in higher order serial learning, they learn to use subtle sequence regularities to respond more quickly and accurately to a series of events. Results reveal a dissociation; overall contextual cuing is spared in healthy aging, whereas higher order sequence learning is impaired in the same individuals. This finding suggests that these 2 forms of implicit learning rely on different neural substrates that age differently; the results are also consistent with recent evidence that fronto-striatal circuits are particularly susceptible to decline in health aging. This study examined relationships between medical complications resulting in hospital stays and alcohol and illicit substance use in 71 persons with recent spinal cord injury (SCI). At 5 intervals after injury, medical records were reviewed for pressure ulcers and urinary tract infections (UTIs). Abstainers with histories of drinking problems before SCI were at greater risk for UTIs from 7 to 12 months after injury and for longer hospital stays. Former drinkers may not have implemented the self-care skills that were a focus during inpatient rehabilitation. Preinjury illicit substance abuse was related to an increased risk of pressure ulcers 30 months after SCL Clinical implications are clear: Psychologists should inquire about substance use patterns, monitor psychological well-being, and explore the ways in which self-care habits are related to substance use. The identification of T cell epitopes presented by alternative HLA-B and -C alleles may provide a means to counteract the tumor escape mechanism based on the selection of tumor cells no longer susceptible to HLA-A-restricted T cell recognition. Several T cell clones and lines were obtained from T lymphocytes purified from melanoma-infiltrated or noninfiltrated lymph nodes of a patient who remained disease free 8 yr after surgery. Selected T cells recognized the autologous melanoma as evaluated by direct cytolysis and production of cytokines. These effectors were directed against the tyrosinase-related protein-2 (TRP-2) and gp100 melanoma epitopes restricted by HLA-Cw8. The nonamer and decamer peptides containing the sequence ANDPIFVVL (residues 387-395) of TRP-2 and the octamer, nonamer, and decamer peptides containing the sequence SNDGPTLI (residues 71-78) of gp100 reconstituted the epitope for TRP-2- and gp100-specific T cell lines and clones, respectively. However, only the nonameric form of TRP-2 and the nonameric and octameric forms of gp100 were able to induce peptide-specific T cells recognizing the autologous tumor in an HLA-class I-restricted fashion from PBMC of the melanoma patient studied. Together these data indicate that HLA-Cw8 can restrict the recognition of gp100 and TRP-2 epitopes by CTL, and that such peptides could stimulate a patient's PBL, suggesting that these Ags could have contributed to a systemic immunity against melanoma.  相似文献

Mercury (Hg) profiles were compared to profiles of climate indicators including microfossil remains and algal-derived or S2 carbon (C) in dated sediment cores from 14 lakes spanning latitudinal and longitudinal gradients across the Canadian high and subarctic. Hg fluxes increased postindustrialization (post-~1850) in 11 of these lakes (postindustrialization Hg fluxes (ΔHgF(F)) = 2-24 μg m(-2) y(-1)). Correction of HgF(F) for catchment contributions demonstrated that Hg deposition originating from catchment-independent factors, such as atmospheric deposition, increased since industrialization in all 14 lakes. Several of these lakes also showed postindustrial shifts in algal assemblages consistent with climate-induced changes. Eleven lakes showed post-1850s increases in S2F(F), suggesting that lake primary productivity has recently increased in the majority of our sites (ΔS2F(F) = 0.1-4 g m(-2) y(-1)). Other studies have interpreted significant relationships between Hg:S2 concentrations in Arctic sediment as support for the algal scavenging hypothesis, which postulates that Hg fluxes to Arctic sediments are largely driven by S2. However, in six of our lakes we observed no Hg:S2 relationship, and in one lake a significant negative Hg:S2 relationship was observed due to increased Hg and decreased S2 C deposition during the postindustrialization period. In six of the seven lakes where a significant positive Hg:S2 relationship was observed, algal assemblages either did not change through time or the timing of the shifts did not correspond to changes in Hg deposition. Our results demonstrate that, although Arctic lakes are experiencing a myriad of changes, including increased Hg and S2 deposition or changing algal assemblages, increased lake primary productivity does not appear to be driving changes in Hg fluxes to sediments.  相似文献

A highly selective method was established to study the dynamic of biogenic amine formation during the brewing process. The presence of histamine, tyramine, cadaverine, putrescine, phenylethylamine, spermidine, and spermine was determined in raw materials (barley, hop, yeast), intermediate products (sweet wort, hopped wort, green and matured beer), and beers. Dansylated amines were separated on a Zorbax-XDB C18 column using a binary mobile phase composed of acetonitrile and water. Chromatographic conditions were optimized using an experimental design, and validation was performed following International Conference on Harmonization (ICH) recommendations. Calibration data fitted a linear regression model with R 2?>?0.99. Repeatability (n?=?6) and intermediate precision (n?=?3) in matrix showed relative standard deviation (RSD) values lower than 7.62 and 8.51 %, respectively. Recoveries, at three different concentrations, ranged from 73.97 to 114.94 %. The proposed method was applied to determine the dynamic of biogenic amine content during brewing process of three microbreweries, allowing the detection of the principal sources or stages of biogenic amine (BA) contribution/formation.  相似文献

Bismuth nano hexagons were synthesized using potentiostatic electrodeposition and studied for their performance towards electrochemical hydrogen storage and evolution. Regular hexagons with edge length ≈ 500 nm and thickness ≈ 80 nm were observed in scanning electron microscopy (SEM). X-ray diffraction (XRD) pattern indicates the presence of poly-crystalline bismuth and bismuth oxide in rhombohedral and cubic phases respectively. X-ray photoelectron spectroscopy (XPS) studies further confirm the presence of two phases of bismuth i.e. elemental bismuth and bismuth (III) oxide. Results indicate that these nano hexagons show good hydrogen ion (H+) storage and successive hydrogen gas (H2) evolution characteristics.  相似文献

Mutations or polymorphisms in the gene of the enzyme methylenetetrahydrofolate (MTHFR) are associated with hyperhomocysteinemia and possibly with an elevated risk for vascular diseases. A study was conducted on 83 individuals with type 2 diabetes in order to determine the allelic and genotypic frequencies of the G1793A mutation and to assess the effect of folic acid supplementation on plasma homocysteine concentrations. The patients were attended by the Diabetes and Hypertension Program--Balneario Camboriu/SC and received daily supplements containing 1 mg of folic acid for 3 months. DNA was previously extracted from leukocytes and the G1793A mutation was detected by PCR-RFLP. Blood samples were collected during the basal period and after supplementation for the determination of homocysteine by HPLC, and of folic acid and vitamin B(12) by RIA. The allele frequency for the G1793A mutation was 3.01% and no homozygous individuals with mutant alleles were detected. Hyperhomocysteinemia was diagnosed in 27.71% of the patients, folic acid deficiency in 15.66%, and vitamin B(12) deficiency in 7.23%. Plasma homocysteine concentrations were inversely correlated with folic acid (r = -0.27, p = 0.01) and vitamin B(12) (r = -0.21; p = 0.05) concentrations. The individuals with a heterozygous genotype for the G1793A mutation showed borderlines or deficient values in folic acid and vitamin B(12) concentrations compared to individuals with a normal genotype. Hyperhomocysteinemia and the vitamin deficiencies presented by type 2 diabetic individuals, included with a heterozygous genotype for the G1793A mutation in the MTHFR gene, reached normal values by daily folic acid supplementation.  相似文献
