The objective of this study was to evaluate the antifungal and antioxidant activities of curcumin, ascorbic acid and the mixture of these two compounds. For the antifungal assay, the minimum inhibitory concentrations (MIC) were determined using Candida strains (ATCC and clinical isolates). Curcumin alone inhibited growth of Candida albicans yeast cells, whereas ascorbic acid did not present effects. However, when the mixture of ascorbic acid and curcumin was assayed to determine the association of the two compounds, the curcumin MIC decreased 5- to 10-fold. In the antioxidant assays, the sum of the alone activities of curcumin and ascorbic acid were lower than the activity of the two-compound mixture. This study highlights the importance of the association between two common antioxidants in foods, to improve the antifungal and antioxidant activities of curcumin (in vitro), and can be applied to Candida spp. infection and diseases associated with oxidative stress.  相似文献

eWALL framework, under development in the European eWALL project (project no. 610658: “eWALL for Active Long Living”) is meant for care of population with age related impairments, mild dementia and COPD. Expanded eWALL (e2WALL) is planned to be an integrated quality framework for long-term care services which will cover very different types of care: all healthcare (both preventive and rehabilitative), societal services, care for cognitive diseases, services delivered at home as well as support for caregivers as an integral part of the quality improvement process. Thus, we present e2WALL home system that would encompass installation (setup) of e2WALL in homes (flats, houses and aging homes) and their management and maintenance across Europe, starting from the partners involved in eWALL (Austria, Bulgaria, Croatia, Denmark, Former Yugoslav Republic of Macedonia, Germany, Greece, Italy, Montenegro, The Netherlands and Romania) and expanding the partnership to the most of Europe (e.g. United Kingdom, Poland, Czech Republic…). This would be a direct application of the developed e2WALL platform and should be done through a network of collaborating small-to-medium sized enterprises (SMEs), each of whom would lead the process of setting up e2WALL system and continuing to provide support services to homes in their respective country. In this way, the growth of high tech and innovative SMEs will be ensured at pan European level, which will enable growth over restricted national market and stimulate European investments in SMEs and startups. Actually, the bigger platform will enable e2WALL European Ecosystem (e4WALL), which can then expand into other non-European markets (e.g. Asia, USA) and means sustainability beyond the life of the project. e2WALL will lead to connected tech start-up hubs to the larger European business ecosystem and will contribute to increased access of country-specific and customized new product to the pan European market, which will lead to increase of employed qualified individuals. Another important focus would be creating an energy efficient environment for a new social network where registered users would provide on-demand services.  相似文献

Recurrent protein folding motifs include various types of helical bundles formed by α-helices that supercoil around each other. While specific patterns of amino acid residues (heptad repeats) characterize the highly versatile folding motif of four-α-helical bundles, the significance of the polypeptide chain directionality is not sufficiently understood, although it determines sequence patterns, helical dipoles, and other parameters for the folding and oligomerization processes of bundles. To investigate directionality aspects in sequence-structure relationships, we reversed the amino acid sequences of two well-characterized, highly regular four-α-helical bundle proteins and studied the folding, oligomerization, and structural properties of the retro-proteins, using Circular Dichroism Spectroscopy (CD), Size Exclusion Chromatography combined with Multi-Angle Laser Light Scattering (SEC-MALS), and Small Angle X-ray Scattering (SAXS). The comparison of the parent proteins with their retro-counterparts reveals that while the α-helical character of the parents is affected to varying degrees by sequence reversal, the folding states, oligomerization propensities, structural stabilities, and shapes of the new molecules strongly depend on the characteristics of the heptad repeat patterns. The highest similarities between parent and retro-proteins are associated with the presence of uninterrupted heptad patterns in helical bundles sequences.  相似文献

The methyllysine reader protein Spindlin1 has been implicated in the tumorigenesis of several types of cancer and may be an attractive novel therapeutic target. Small‐molecule inhibitors of Spindlin1 should be valuable as chemical probes as well as potential new therapeutics. We applied an iterative virtual screening campaign, encompassing structure‐ and ligand‐based approaches, to identify potential Spindlin1 inhibitors from databases of commercially available compounds. Our in silico studies coupled with in vitro testing were successful in identifying novel Spindlin1 inhibitors. Several 4‐aminoquinazoline and quinazolinethione derivatives were among the active hit compounds, which indicated that these scaffolds represent promising lead structures for the development of Spindlin1 inhibitors. Subsequent lead optimization studies were hence carried out, and numerous derivatives of both lead scaffolds were synthesized. This resulted in the discovery of novel inhibitors of Spindlin1 and helped explore the structure–activity relationships of these inhibitor series.  相似文献

This article is aimed to discuss the chemical aspects of detonation spraying of powder materials. In this method of coating deposition, ceramic, metallic or composite powders are injected into the barrel of a detonation gun filled with an explosive gaseous mixture. When the latter is ignited, the powders are heated and accelerated toward the substrate. Subjected to high temperatures, the powders are prone to chemical reactions, the reaction products possibly becoming the major phase constituents of the coatings. What types of reactions are possible? Can these reactions be carried out in a controlled manner? We answer these questions considering the interactions of the sprayed powders with the gaseous environment of the barrel as well as those between the phases of a composite feedstock powder. In Computer-Controlled Detonation Spraying (CCDS), the explosive charge and stoichiometry of the fuel-oxygen mixtures are precisely measured and can be flexibly changed. Our studies demonstrate that with the introduction of a highly flexible process of CCDS, detonation spraying has entered a new development stage, at which it can be considered as a powerful method of composition and microstructure tailoring of thermally sprayed coatings. During CCDS of TiO2-containing powders, chemical reduction of titanium dioxide can be carried out to different levels to form either oxygen-deficient TiO2−x or Ti3O5 suboxide. CCDS of Ti3Al can produce titanium oxide coatings when oxidation by the detonation products dominates or titanium nitride-titanium aluminide coatings when oxidation is hindered but the interaction of the powders with nitrogen—a carrier gas component—is favored. During detonation spraying of Ti3SiC2–Cu composites, the Ti3SiC2 phase is preserved only in cold conditions; otherwise, Si de-intercalates from the Ti3SiC2 phase and dissolves in Cu resulting in the formation of the TiCx–Cu(Si) composite coatings.

Since 2003 India has been actively promoting the cultivation of Jatropha on unproductive and degraded lands (wastelands) for the production of biodiesel suitable as transportation fuel. In this paper the life cycle energy balance, global warming potential, acidification potential, eutrophication potential and land use impact on ecosystem quality is evaluated for a small scale, low-input Jatropha biodiesel system established on wasteland in rural India. In addition to the life cycle assessment of the case at hand, the environmental performance of the same system expanded with a biogas installation digesting seed cake was quantified. The environmental impacts were compared to the life cycle impacts of a fossil fuel reference system delivering the same amount of products and functions as the Jatropha biodiesel system under research. The results show that the production and use of Jatropha biodiesel triggers an 82% decrease in non-renewable energy requirement (Net Energy Ratio, NER = 1.85) and a 55% reduction in global warming potential (GWP) compared to the reference fossil-fuel based system. However, there is an increase in acidification (49%) and eutrophication (430%) from the Jatropha system relative to the reference case. Although adding biogas production to the system boosts the energy efficiency of the system (NER = 3.40), the GWP reduction would not increase (51%) due to additional CH4 emissions. For the land use impact, Jatropha improved the structural ecosystem quality when planted on wasteland, but reduced the functional ecosystem quality. Fertilizer application (mainly N) is an important contributor to most negative impact categories. Optimizing fertilization, agronomic practices and genetics are the major system improvement options.  相似文献
