The bacterial community of fermented horse milk (koumiss) from Mongolia was studied using three methods: cultivation, direct identification by 16S rRNA clone library and denaturing gradient gel electrophoresis (DGGE). Ninety-eight strains were isolated by traditional cultivation and 61 of those were randomly selected for further identification by 16S rRNA gene sequencing. The strains were dominated by lactic acid bacteria (LAB; six different lactobacilli), Acinetobacter, Bacillus and Psychrobacter. Construction of the clone library analysis revealed that 16S sequences of 220 clones, genus Lactobacillus was dominant, but Streptococcus thermophilus, Acetobacter pasteurianus and uncultured clones were also detected. Ten unique bands were sequenced from the DGGE and revealed: Lactococcus lactis, Lactococcus lactis subsp. lactis, Clostridium acidurici, Acinetobacter johnsonii, Dickeya sp., Enterobacter sp., Pseudomonas sp., Raoultella sp., and Ruminococcus sp.. In vitro growth inhibition of three human pathogens, Escherichia coli, Enterobacter sakazakii and Staphylococcus aureus by 14 culturable bacteria displayed that only three of the isolates tested inhibit growth of E. sakazakii while most of the other bacteria delayed growth of the target bacteria.  相似文献

The feasibility of UV photoionization for single unipolar charging of nanoparticles at flow rates up to 100 l· min ?1 is demonstrated. The charging level of the aerosol particles can be varied by adjusting the intensity of the UV radiation. The suitability of a UV photocharger followed by a DMA to deliver monodisperse nanoparticles at high aerosol flow rates has been assessed experimentally in comparison to a radioactive bipolar charger ( 85 Kr, 10 mCi). Monodisperse aerosols with particle sizes in the range of 5 to 25 nm and number concentrations between 10 4 and 10 5 cm ?3 have been obtained at flow rates up to 100 l· min ?1 with the two aerosol chargers. In terms of output particle concentration, the UV photoionizer performs better than the radioactive ionizer with increasing aerosol flow rate. Aerosol charging in the UV photoionizer is described by means of a photoelectric charging model that relies on an empirical parameter and of a diffusion charging model based on the Fuchs theory. The UV photocharger behaved as a quasi-unipolar charger for polydisperse aerosols with particles sizes less than 30 nm and number concentrations ~10 7 cm ?3 . Much reduced diffusion charging was observed in the experiments, with respect to the calculations, likely due to ion losses onto the walls caused by unsteady electric fields in the irradiation region.  相似文献

The absolute grain boundary mobility of 388 nickel grain boundaries was calculated using a synthetic driving force molecular dynamics method; complete results appear in the Supplementary materials. Over 25% of the boundaries, including most of the non-Σ3 highest mobility boundaries, moved by a coupled shear mechanism. The range of non-shearing boundary mobilities is from 40 to 400 m/s GPa, except for Σ3 incoherent twins which have mobilities of 200–2000 m/s GPa. Some boundaries, including all the 〈1 1 1〉 twist boundaries, are immobile within the resolution of the simulation. Boundary mobility is not correlated with scalar parameters such as disorientation angle, Σ value, excess volume or boundary energy. Boundaries less than 15° from each other in five-dimensional crystallographic space tend to have similar mobilities. Some boundaries move via a non-activated motion mechanism, which greatly increases low-temperature mobility. Thermal roughening of grain boundaries is widely observed, with estimated roughening temperatures substantially among boundaries.  相似文献

Iterative application of two simulation tools to enable the determination of the required airing to preclude mould growth in corners. A newly developed whole building model is able to determine the effect of different ventilation schemes on indoor climate. Feeding the resulting surface temperatures and humidities to a biohygrothermal model allows to assess the risk of mould growth. For given ambient conditions it is thus possible to determine the necessary minimum ventilation rate by iteratively applying the two models. As an example, different building constructions are investigated, showing the effect of different natural ventilation schemes (continuous ventilation versus pulse ventilation by opening the window) on the minimum mean air change rate required for preventing mould growth.  相似文献

Enterprise Architecture (EA) is an approach used to provide decision support based on organization-wide models. The creation of such models is, however, cumbersome as multiple aspects of an organization need to be considered, making manual efforts time-consuming, and error prone. Thus, the EA approach would be significantly more promising if the data used when creating the models could be collected automatically—a topic not yet properly addressed by either academia or industry. This paper proposes network scanning for automatic data collection and uses an existing software tool for generating EA models (ArchiMate is employed as an example) based on the IT infrastructure of enterprises. While some manual effort is required to make the models fully useful to many practical scenarios (e.g., to detail the actual services provided by IT components), empirical results show that the methodology is accurate and (in its default state) require little effort to carry out.  相似文献

Glioblastoma (GBM) is an obligatory lethal brain tumor with a median survival, even with the best standard of care therapy, of less than 20 months. In light of this fact, the evaluation of new GBM treatment approaches such as oncolytic virotherapy (OVT) is urgently needed. Based on our preliminary preclinical data, the YB-1 dependent oncolytic adenovirus (OAV) XVir-N-31 represents a promising therapeutic agent to treat, in particular, therapy resistant GBM. Preclinical studies have shown that XVir-N-31 prolonged the survival of GBM bearing mice. Now using an immunohumanized mouse model, we examined the immunostimulatory effects of XVir-N-31 in comparison to the wildtype adenovirus (Ad-WT). Additionally, we combined OVT with the inhibition of immune checkpoint proteins by using XVir-N-31 in combination with nivolumab, or by using a derivate of XVir-N-31 that expresses a PD-L1 neutralizing antibody. Although in vitro cell killing was higher for Ad-WT, XVir-N-31 induced a much stronger immunogenic cell death that was further elevated by blocking PD-1 or PD-L1. In vivo, an intratumoral injection of XVir-N-31 increased tumor infiltrating lymphocytes (TILs) and NK cells significantly more than Ad-WT not only in the virus-injected tumors, but also in the untreated tumors growing in the contralateral hemisphere. This suggests that for an effective treatment of GBM, immune activating properties by OAVs seem to be of greater importance than their oncolytic capacity. Furthermore, the addition of immune checkpoint inhibition (ICI) to OVT further induced lymphocyte infiltration. Consequently, a significant reduction in contralateral non-virus-injected tumors was only visible if OVT was combined with ICI. This strongly indicates that for an effective eradication of GBM cells that cannot be directly targeted by an intratumoral OV injection, additional ICI therapy is required.  相似文献
