High concentrations of Escherichia coli in mats of Cladophora in the Great Lakes have raised concern over the continued use of this bacterium as an indicator of microbial water quality. Determining the impacts of these environmentally abundant E. coli, however, necessitates a better understanding of their ecology. In this study, the population structure of 4285 Cladophora-borne E. coli isolates, obtained over multiple three day periods from Lake Michigan Cladophora mats in 2007-2009, was examined by using DNA fingerprint analyses. In contrast to previous studies that have been done using isolates from attached Cladophora obtained over large time scales and distances, the extensive sampling done here on free-floating mats over successive days at multiple sites provided a large dataset that allowed for a detailed examination of changes in population structure over a wide range of spatial and temporal scales. While Cladophora-borne E. coli populations were highly diverse and consisted of many unique isolates, multiple clonal groups were also present and accounted for approximately 33% of all isolates examined. Patterns in population structure were also evident. At the broadest scales, E. coli populations showed some temporal clustering when examined by year, but did not show good spatial distinction among sites. E. coli population structure also showed significant patterns at much finer temporal scales. Populations were distinct on an individual mat basis at a given site, and on individual days within a single mat. Results of these studies indicate that Cladophora-borne E. coli populations consist of a mixture of stable, and possibly naturalized, strains that persist during the life of the mat, and more unique, transient strains that can change over rapid time scales. It is clear that further study of microbial processes at fine spatial and temporal scales is needed, and that caution must be taken when interpolating short term microbial dynamics from results obtained from weekly or monthly samples.  相似文献

The optimization of the manufacturing/assembly tolerances and processes in ITER Experimental Nuclear Fusion Device is one of the key tasks to optimize the fabrication cost, to prevent problems during assembly and to ensure that the critical homogeneity of the magnetic field and the positioning requirements of the plasma facing components can be achieved. This task is further complicated by the strong interplay among the various Tokamak systems, as for instance in the inner region of the machine where the clearances between Central Solenoid, Toroidal Field Coils, Thermal Shield, Vacuum Vessel and In-Vessel components have been minimized for their large influence on the magnetic flux and the overall machine cost.A 3D tolerance simulation analysis of ITER Tokamak machine has been developed based on 3DCS dedicated software. The dimensional variation model is representative of Tokamak functional tolerances and processes, predicting accurate values for the amount of variation on critical areas. In addition, dimensional simulations help to determine the key tolerances that contribute to a particular variation.This paper describes the current status of the Tokamak dimensional variation studies and its management plan, highlighting the status of compliance of allocated tolerances with input requirements. Management of risk issues and corrective actions are also described.  相似文献

While the quality control procedures for multistage has been considerably studied, multistage process capability analysis has not been explored much at all. Fostering on the recent development of the multistage process variation transmission model, a multistage process capability analysis algorithm is developed to prioritize process improvement efforts. People's motivational states--their wishes and preferences--influence their processing of visual stimuli. In 5 studies, participants shown an ambiguous figure (e.g., one that could be seen either as the letter B or the number 13) tended to report seeing the interpretation that assigned them to outcomes they favored. This finding was affirmed by unobtrusive and implicit measures of perception (e.g., eye tracking, lexical decision tasks) and by experimental procedures demonstrating that participants were aware only of the single (usually favored) interpretation they saw at the time they viewed the stimulus. These studies suggest that the impact of motivation on information processing extends down into preconscious processing of stimuli in the visual environment and thus guides what the visual system presents to conscious awareness. We employ first-principles density functional calculations to explore atomic-level interactions and predict the ideal work of adhesion at the SiO2/nickel and ZrO2/SiO2 interfaces. We find that chemical bonding at the interface serves to strengthen significantly interfaces formed with SiO2, which exhibits significant covalent bonding character, relative to those formed using more ionic oxides, such as Al2O3, in place of SiO2. The improved strength of these interfaces due to local bonding interactions may hold materials design implications for practical applications that require optimal adhesion between metal-ceramic layered structures, including thermal barrier coatings.  相似文献

This paper interrogates the socio-spatial environment that is emerging in Singapore with its development of a globalised economy. Using the case studies of two of the city’s dichotomised spaces - the spectacular downtown and gentrified heartland - the paper seeks to examine the resulting spatial reconfigurations, challenges and unintended consequences. While the common assertion is that state-led planning is effective at the macro level, at the micro level against the accelerated change and emergence of the multi-layered package of spaces of globalisation, the planning of the city is no longer limited to physical planning. Like many emergent economies of the global south, the challenge of how to expand emerging sectors, support Singapore’s economic aspirations and meet social objectives remains.  相似文献

Cisplatin is an effective anticancer agent, but also causes permanent hearing loss by damaging hair cells—the sensory receptors essential for hearing. There is an urgent clinical need to protect cochlear hair cells in patients undergoing cisplatin chemotherapy. The zebrafish lateral line organ contains hair cells and has been frequently used in studies to screen for otoprotective compounds. However, these studies have employed a wide range of cisplatin dosages and exposure times. We therefore performed a comprehensive evaluation of cisplatin ototoxicity in the zebrafish lateral line with the goal of producing a standardized, clinically relevant protocol for future studies. To define the dose- and time-response patterns of cisplatin-induced hair-cell death, we treated 6-day-old larvae for 2 h in 50 µM–1 mM cisplatin and allowed them to recover. We observed delayed hair cell death, which peaked at 4–8 h post-exposure. Cisplatin also activated a robust inflammatory response, as determined by macrophage recruitment and phagocytosis of hair cells. However, selective depletion of macrophages did not affect hair cell loss. We also examined the effect of cisplatin treatment on fish behavior and found that cisplatin-induced lateral line injury measurably impaired rheotaxis. Finally, we examined the function of remaining hair cells that appeared resistant to cisplatin treatment. We observed significantly reduced uptake of the cationic dye FM1-43 in these cells relative to untreated controls, indicating that surviving hair cells may be functionally impaired. Cumulatively, these results indicate that relatively brief exposures to cisplatin can produce hair cell damage and delayed hair cell death. Our observations provide guidance on standardizing methods for the use of the zebrafish model in studies of cisplatin ototoxicity.  相似文献

Cold Atmospheric Plasma (CAP) is an emerging technology with great potential for biomedical applications such as sterilizing equipment and antitumor strategies. CAP has also been shown to improve skin wound healing in vivo, but the biological mechanisms involved are not well known. Our study assessed a possible effect of a direct helium jet CAP treatment on keratinocytes, in both the immortalized N/TERT-1 human cell line and primary keratinocytes obtained from human skin samples. The cells were covered with 200 µL of phosphate buffered saline and exposed to the helium plasma jet for 10–120 s. In our experimental conditions, micromolar concentrations of hydrogen peroxide, nitrite and nitrate were produced. We showed that long-time CAP treatments (≥60 s) were cytotoxic, reduced keratinocyte migration, upregulated the expression of heat shock protein 27 (HSP27) and induced oxidative cell stress. In contrast, short-term CAP treatments (<60 s) were not cytotoxic, did not affect keratinocyte proliferation and differentiation, and did not induce any changes in mitochondria, but they did accelerate wound closure in vitro by improving keratinocyte migration. In conclusion, these results suggest that helium-based CAP treatments improve wound healing by stimulating keratinocyte migration. The study confirms that CAP could be a novel therapeutic method to treat recalcitrant wounds.  相似文献
