Photochemical behaviour of sulcotrione, a triketone herbicide, was studied in a variety of aqueous solutions including natural waters (sea and river) under laboratory conditions. Photodegradation experiments were carried out under two irradiation systems (UV-B and simulated solar radiation) in order to evaluate kinetics of active ingredient. The degradation kinetics, more rapid under UV-B radiation than solar simulator, followed a first-order reaction (photolysis half-lives ranged between 3 and 50 h) and appeared strongly dependent on water origin, pH value and molecular structure of the herbicide. Dissolved organic matter showed a retarding effect while low concentrations of nitrate ions had no effect on photolysis rate. Identification of photoproducts indicated that hydrolysis, a pH-dependent process (no degradation at pH >6 but at pH=3, k=0.0344 h(-1)), could be photoassisted. These results were compared to those of mesotrione, another triketone herbicide, which appeared more stable under UV-B irradiation. Toxicological studies on two marine heterotrophic bacteria and one cyanobacterium showed absence of effects up to 100 microgL(-1) for both sulcotrione and its photoproducts.  相似文献

Here, we describe a new fluorescence polarization aptamer assay (FPAA) strategy which is based on the use of the single-stranded DNA binding (SSB) protein from Escherichia coli as a strong FP signal enhancer tool. This approach relied on the unique ability of the SSB protein to bind the nucleic acid aptamer in its free state but not in its target-bound folded one. Such a feature was exploited by using the antiadenosine (Ade)-DNA aptamer (Apt-A) as a model functional nucleic acid. Two fluorophores (fluorescein and Texas Red) were introduced into different sites of Apt-A to design a dozen fluorescent tracers. In the absence of the Ade target, the binding of the labeled aptamers to SSB governed a very high fluorescence anisotropy increase (in the 0.130-0.200 range) as the consequence of (i) the large global diffusion difference between the free and SSB-bound tracers and (ii) the restricted movement of the dye in the SSB-bound state. When the analyte was introduced into the reaction system, the formation of the folded tertiary structure of the Ade-Apt-A complex triggered the release of the labeled nucleic acids from the protein, leading to a strong decrease in the fluorescence anisotropy. The key factors involved in the fluorescence anisotropy change were considered through the development of a competitive displacement model, and the optimal tracer candidate was selected for the Ade assay under buffer and realistic (diluted human serum) conditions. The SSB-assisted principle was found to operate also with another aptamer system, i.e., the antiargininamide DNA aptamer, and a different biosensing configuration, i.e., the sandwich-like design, suggesting the broad usefulness of the present approach. This sensing platform allowed generation of a fluorescence anisotropy signal for aptamer probes which did not operate under the direct format and greatly improved the assay response relative to that of the most previously reported small target FPAA.  相似文献

The effects of fat-soluble vitamins (such as vitamins A and E) and lipid microconstituents (such as carotenoids) on human health are now well established. However, high-performance liquid chromatography (HPLC) methods able to detect these molecules in simultaneous runs are often difficult to set up. We report here a 35-min reversed-phase HPLC method using a single C30 column kept at 35°C with a gradient system of methanol, methyl-tert-butyl ether and water at a flow-rate of 1 mL/min. This method resolves 11 carotenoids, retinol, α- and γ-tocopherol from complex matrixes such as food samples, human plasma and human adipose tissue within 35 min. The method is also able to separate coenzyme Q(10). The intra-day and inter-day coefficients of variation are suitable for routine clinical and scientific applications for the determination of lipid micronutrients from various sample types.  相似文献

We have isolated a mutant (rvs272) of the yeast (Saccharomyces cerevisiae) that displays an altered phenotype in stationary phase. It shows a high proportion of large-budded cells with two non-segregated nuclei staying in the mother cell. This phenotype is also expressed in various conditions when cells are synchronized, energy depleted or treated with the antimitotic drug benomyl. The RVS272 gene has been identified as the NUM1 gene already described. This gene presents a 192 bp tandemly repeated motif and we show that the number of repeats can vary from 1 to about 24 among different strains, without apparently affecting the function of the encoded protein. We suggest that this protein could be involved in polymerization catalysis and/or stabilization of microtubules.  相似文献

Pharmacokinetic/pharmacodynamic data are often analysed using nonlinear mixed-effect models, and model evaluation should be an important part of the analysis. Recently, normalised prediction distribution errors (npde) have been proposed as a model evaluation tool. In this paper, we describe an add-on package for the open source statistical package R, designed to compute npde. npde take into account the full predictive distribution of each individual observation and handle multiple observations within subjects. Under the null hypothesis that the model under scrutiny describes the validation dataset, npde should follow the standard normal distribution. Simulations need to be performed before hand, using for example the software used for model estimation. We illustrate the use of the package with two simulated datasets, one under the true model and one with different parameter values, to show how npde can be used to evaluate models. Model estimation and data simulation were performed using NONMEM version 5.1.  相似文献

Information and Communication Technology‐enabled Development (ICT4D) discourse relies upon the idea that ICTs can foster development in particular by encouraging wider participation in development initiatives. In this paper, we question how the blogging practices of development professionals shape such ICT4D discourse. Through a combination of interviews and analyses of blog contents, we examine two major purposes of blogging: reflecting upon development practices and engaging with a self‐selected audience. Our analyses reveal that these two purposes were interwoven in ways that contributed to making bloggers' ICT4D discourse innovative but oriented towards a small community of peers rather than a larger audience. Through blogging, development professionals refined their expertise on ICT4D. As they did so, they also generated a personal speaker's corner that primarily attracted like‐minded peers rather than promoting larger participation in ICT4D discourse. This research contributes to the emerging literature on social media practices by showing how blogging practices enable the formation of what a discourse is about, and by highlighting differences between perceived and actual levels of interactions between bloggers and their audience. The paper also adds to the ICT and development literatures by revealing that blogging practices can deepen ICT4D discourse, but that they do not necessarily enhance participation in development. Such insight is crucial for development professionals to develop realistic expectations of blogging for ICT4D.  相似文献
