This paper presents the effects of cooling methods on residual compressive strength and cracking behavior of concretes containing four different class F fly ash contents of 10%, 20%, 30% and 40% as partial replacement of cement at various elevated temperatures. The residual compressive strength of the aforementioned fly ash concretes is measured after being exposed to 200, 400, 600 and 800 °C temperatures and two different cooling methods, for example, slow cooling and rapid water cooling. Results show that the residual compressive strengths of all fly ash concretes decrease with increase in temperatures irrespective of cooling regimes, which is similar to that of ordinary concrete. Generally, control ordinary concrete and all fly ash concretes exhibited between 10% and 35% more reduction in residual compressive strength because of rapid cooling than slow cooling except few cases. Cracks are observed over concrete specimens after being exposed to temperatures ranging from 400 to 800 °C. Samples that are slowly cooled developed smaller cracks than those rapidly cooled. At 800 °C, all fly ash concretes that are exposed to rapid cooling showed the most severe cracking. X‐ray diffraction analysis shows reduction of Ca(OH)2 peak and formation of new calcium silicate peak in concretes containing 20% and 40% fly ash when subjected to 800 °C in both cooling methods. Thermo gravimetric analysis and differential thermal analysis results show increase in thermal stability of concrete with increase in fly ash contents. The existing Eurocode also predicted the compressive strength of fly ash concretes with reasonable accuracy when subjected to the aforementioned elevated temperatures and cooling methods. Deep annotation of a library of 4-anilinoquin(az)olines led to the identification of 7-iodo-N-(3,4,5-trimethoxyphenyl)quinolin-4-amine 16 as a potent inhibitor (IC50=14 nM) of Protein Kinase Novel 3 (PKN3) with micromolar activity in cells. Compound 16 is a potential tool compound to study the cell biology of PKN3 and its role in pancreatic and prostate cancer and T-cell acute lymphoblastic leukemia. These 4-anilinoquin(az)olines may also be useful tools to uncover the therapeutic potential of PKN3 inhibition in a broad range of diseases.  相似文献

This article addresses effects of weave defects in an angle‐interlock C‐fiber preform on the tensile properties of the resulting fully processed C‐fiber/SiC‐matrix composite. For this purpose, a preform was intentionally sheared in a controlled manner after weaving. The resulting distortions were quantified by analyzing high‐resolution images of the preform surface after the first step of matrix processing, while the tows were still clearly visible. Comparisons are made of tensile test results on specimens cut from this composite panel and from a pristine panel in select loading orientations. Strain maps obtained by digital image correlation are used to identify local strain variations that are attributable to weave defects. The results are discussed in terms of: (i) the shear‐normal coupling that arises in loading orientations of present interest, and (ii) the geometric effects of tow misalignment on tow continuity along the specimen gauge length. The composite is found to perform in a robust manner, in the sense that the tensile properties are not sensitive to the presence of the defects.  相似文献

For quantitative microRNA analyses in formalin-fixed paraffin-embedded (FFPE) tissue, expression levels have to be normalized to endogenous controls. To investigate the most stably-expressed microRNAs in breast cancer and its surrounding tissue, we used tumor samples from primary tumors and from metastatic sites. MiRNA profiling using TaqMan® Array Human MicroRNA Cards, enabling quantification of 754 unique human miRNAs, was performed in FFPE specimens from 58 patients with metastatic breast cancer. Forty-two (72%) samples were collected from primary tumors and 16 (28%) from metastases. In a cross-platform analysis of a validation cohort of 32 FFPE samples from patients with early breast cancer genome-wide microRNA expression analysis using SurePrintG3 miRNA (8 × 60 K)® microarrays from Agilent® was performed. Eleven microRNAs could be detected in all samples analyzed. Based on NormFinder and geNorm stability values and the high correlation (rho ≥ 0.8) with the median of all measured microRNAs, miR-16-5p, miR-29a-3p, miR-126-3p, and miR-222-3p are suitable single gene housekeeper candidates. In the cross-platform validation, 29 human microRNAs were strongly expressed (mean log2-intensity > 10) and 21 of these microRNAs including miR-16-5p and miR-29a-3p were also stably expressed (CV < 5%). Thus, miR-16-5p and miR-29a-3p are both strong housekeeper candidates. Their Normfinder stability values calculated across the primary tumor and metastases subgroup indicate that miR-29a-3p can be considered as the strongest housekeeper in a cohort with mainly samples from primary tumors, whereas miR-16-5p might perform better in a metastatic sample enriched cohort.  相似文献
