Extracellular ATP (eATP) has long been established in animals as an important signalling molecule but this is less understood in plants. The identification of Arabidopsis thaliana DORN1 (Does Not Respond to Nucleotides) as the first plant eATP receptor has shown that it is fundamental to the elevation of cytosolic free Ca2+ ([Ca2+]cyt) as a possible second messenger. eATP causes other downstream responses such as increase in reactive oxygen species (ROS) and nitric oxide, plus changes in gene expression. The plasma membrane Ca2+ influx channels involved in eATP-induced [Ca2+]cyt increase remain unknown at the genetic level. Arabidopsis thaliana Annexin 1 has been found to mediate ROS-activated Ca2+ influx in root epidermis, consistent with its operating as a transport pathway. In this study, the loss of function Annexin 1 mutant was found to have impaired [Ca2+]cyt elevation in roots in response to eATP or eADP. Additionally, this annexin was implicated in modulating eATP-induced intracellular ROS accumulation in roots as well as expression of eATP-responsive genes.  相似文献

The spectral range of sunlight absorbed by a luminescent solar concentrator (LSC) is increased by using multiple dyes. Absorption, fluorescence, and fluorescence excitation spectra, and relative light output are reported for LSCs made with one, two, or three BODIPY dyes in a thin polymer layer on glass. Losses caused by multiple emission and reabsorption events are minimized by optimizing resonance excitation energy transfer between dyes. Increases in the outputs from the multiple-dye LSCs are directly proportional to increases in the number of photons absorbed. The output of the three-dye LSC is 45–170% higher than those of the single-dye LSCs.  相似文献

Dams, increasingly common in riverine systems worldwide, are particularly prevalent on the Columbia River (CR) in the United States. Hydroelectric projects, including both storage and run‐of‐river (i.e., minimal storage) structures, on the mainstem CR highly manage water flow, often by releasing water over (rather than through) dams as “spill.” To test the effects of run‐of‐river dam spill on microplankton abundance and composition, we sampled above and below two dams in the lower CR before and during spill conditions in spring 2016 and during and after spill conditions in late summer 2007. We tested the effects of location (i.e., above vs. below dams), spill condition (i.e., before, during, and after spill), and their interaction on microplankton abundance. Generally, diatoms were most abundant during springtime, whereas cyanobacteria were most abundant in late summer. Most taxa were not significantly different in abundance above and below dams, regardless of spill status; although cyanobacteria abundance was marginally higher below dams in summer 2007 (p = .04). Abundances of all taxa were significantly different between pre‐spill and spill periods in spring 2016, whereas only diatom and flagellate abundances were significantly different between spill and post‐spill periods in summer 2007. We conclude that spill conditions may influence microplankton abundance, but are not likely to affect microplankton communities on either side of run‐of‐river dams on the CR. This is important information for dam managers concerned about ecosystem impacts of spill.  相似文献

River channel substrate size and mobility are important to Atlantic salmon spawning and rearing success. We compare morphology and bed sediment between two North American Atlantic coastal streams (Narraguagus River, Maine, USA and Jacquet River, New Brunswick, Canada). The watersheds have similar drainage areas and mean annual precipitation, but differing relief structure, channel longitudinal profiles and numbers of returning salmon. The lower‐relief Narraguagus River is segmented into steeper (gradient >0.002) and flatter reaches (gradient <0.0005). Flat reaches, including mainstem lakes, act as sediment sinks, preventing the continuity of downstream sediment transport. Based on field measurements, the Narraguagus River has a larger high‐flow width to depth ratio than the Jacquet River, but this difference is principally the result of outliers from low‐gradient channel reaches. Measurements of substrate grain size reveal finer river‐bed sediments on the Narraguagus River, however, Shields parameter calculations indicate that bed sediment should be mobile during high flows in both streams. We use the Shields equation to predict grain size based on channel slope, width and drainage area measured from digital elevation models (DEM) and aerial photographs. Predictions of median grain size agree with field measurements within a factor of 2 for 62–70% of the survey stations. We suggest ways that model misfits may provide opportunities to prioritize reach‐based restoration efforts. Based on expected grain size, we estimate 62% spawning and 68% rearing habitat along the length of the Narraguagus River, and 28 and 95% respectively on the Jacquet. Harmful algal blooms are occurring in large river ecosystems and at the mouth of large rivers with increasing frequency. In lentic systems, the chemical and physical conditions that promote harmful algal blooms are somewhat predictable but tracking prevalence and conditions that promote harmful algal blooms in lotic systems is much more difficult. We captured two of the most extreme discharge years within the last 20 years occurring in the Upper Mississippi River, allowing a natural experiment that evaluated how major shifts in discharge drive environmental variation and associated shifts in phytoplankton. Statistical models describing significant environmental covariates for phytoplankton assemblages and specific taxa were developed and used to identify management‐relevant numeric breakpoints at which environmental variables may promote the growth of specific phytoplankton and/or cyanobacteria. Our analyses supported that potentially toxin‐producing cyanobacteria dominate under high phosphorus concentration, low nitrogen concentration, low nitrogen‐to‐phosphorus ratio, low turbulence, low flushing, adequate light and warm temperatures. Cyanobacteria dominated in 2009 when low discharge and low flushing likely led to optimal growth environments for Dolichospermum, Aphanizomenon and Microcystis. Rarely will a single factor lead to the dominance, but multiple positive factors working in concert can lead to cyanobacteria proliferation in large rivers. Certain isolated backwaters with high phosphorus, low nitrogen, warm water temperatures and low potential for flushing could benefit from increased connection to channel inputs to reduce cyanobacterial dominance. Numerous examples of this type of habitat currently exist in the Upper Mississippi River and could benefit from reconnection to channel habitats.  相似文献

Lake trout (Salvelinus namaycush) reared in hatcheries are exposed to an environment and feeding regime that is different from wild lake trout, and are stocked at substantially larger sizes with higher lipid reserves. In addition to differences in diet and growth, this early experience may alter habitat use compared to the wild cohort. We used seasonal data on the depth and temperature distribution of wild and stocked juvenile lake trout to test for differences in habitat use and inform sampling strategies to evaluate annual recruitment. Bottom trawling was conducted from 2015 to 2019 in the central basin of Lake Champlain every two to four weeks during the ice-free season. Differences in distribution of wild and stocked lake trout were most pronounced during thermal stratification, when wild juveniles were more abundant than stocked juveniles at shallower depths and warmer temperatures and stocked juveniles were more abundant at deeper depths and colder temperatures. Temperature preferences may be a consequence of different early rearing environments; wild lake trout are acclimated to lake temperatures and forage, whereas stocked fish entered the lake with high lipid content and little foraging experience. Unbiased assessment of the proportion of wild lake trout and growth and survival of the entire juvenile lake trout population using bottom trawl sampling should either take place in the pre- and post-stratification seasons when wild and stocked fish are at the same depths, or include the full range of depths and temperatures that wild and stocked fish occupy during the stratified period.  相似文献

Formaldehyde, Bronopol, tylosin and pimaricin were tested as silage additives in simulated silage based on laboratory media and microbial inocula. Fermentation was restricted by 33 mM formaldehyde, 1.2 mM Bronopol and 0.2 mM tylosin. Pimaricin had little effect on fermentation but a level of 0.3 mM prevented mould growth when the silages were exposed to air. The antimicrobial effects of formaldehyde and Bronopol, the antibacterial effect of tylosin and the antimycotic effect of pimaricin support the results obtained in earlier work with cultures of silage microorganisms. The concentrations required to prevent growth of lactic acid bacteria were, however, about four times higher than in pure culture tests. The levels of formaldehyde and Bronopol required to prevent fermentation in the simulated silages are, however, less than in needed in the test-tube silos containing forage.  相似文献

Understanding variation in fish populations is valuable from both a management and an ecological perspective. Great Lakes sea lampreys are controlled primarily by treating tributaries with lampricides that target the larval stage. Great Lakes streams were divided into four categories based on their regularity of parasitic lamprey production inferred from the historic regularity of chemical treatments. This categorization was intended to direct future assessment efforts, but may also reflect differences in early demographics. We analyzed assessment data collected from 1959 to 2005 using mixed-effects models and variance components analyses to test for differences in recruitment and growth to age 1 among stream categories. Recruitment was twice as large in regularly treated streams as in irregularly treated streams, indicating that age-1 year-class strength is correlated with consistent chemical treatments. We found no differences in length at age 1 among stream categories; however, Lake Superior streams with irregular treatment histories exhibit more variation in length at age 1 than streams that are treated regularly. The majority of variation in length at age 1 was due to within-year variation, which was fairly consistent across stream types within each lake. Our results indicate that early life history differs among subsets of the Great Lakes sea lamprey population, and management practices should be modified to account for these differences. Mixed-effects models and variance components analyses are useful tools for analyzing large historical datasets for patterns of demographic variation within and among populations, whether the ultimate goal is pest control, harvesting, or conservation.  相似文献

The Safety Walks Group is an initiative that evolved from the Stay on Your Feet Program. The strategies used in this program target both behavioural and environmental change and are based on the five areas for action under the Ottawa Charter (WHO, 1986) and Jakarta Declaration (WHO, 1997). The Safety Walks Group addresses the issue of public hazards via the use of a standard checklist covering pedestrian areas, business houses and accommodation. The project provided a forum for seniors to be proactive, working with the authorities to address the issue of public hazards and make the environment safer.  相似文献
