The effect of allogeneic cell therapy on tumor growth was studied in a murine model of mammary carcinoma (4T1) as an experimental model of solid tumors in humans. i.v. inoculation of 4T1 (H-2d) cells into syngeneic mice [BALB/c or (BALB/cXC57BL/6)F1] (F1) carrying the H-2d histocompatible antigens results in tumor colonies in the lungs that finally cause the death of all of the mice. Sublethally irradiated F1 mice were inoculated with 4T1 cells to simulate minimal residual disease and with immunocompetent splenocytes derived from naive donors of F1 (syngeneic), BALB/c (syngeneic to the tumor but semiallogeneic to the host), or C57BL/6 (allogeneic to the tumor and semiallogeneic to the host) mice. The survival of F1 tumor-bearing mice that were treated with allogeneic C57BL/6 splenocytes was significantly prolonged (P < 0.02) compared with hosts given F1 or BALB/c-derived splenocytes that are syngeneic to 4T1 tumor cells. Adoptive transfer of lung cells that were isolated from F1 primary mice inoculated with 4T1 cells and syngeneic BALB/c or F1 splenocytes led to local tumor growth and death in secondary recipients. In contrast, only 1 of 22 secondary recipients developed tumors when inoculated with lung cells derived from F1 mice given allogeneic C57BL/6 splenocytes. All of the 21 secondary hosts survived disease-free for a follow-up time of >200 days. These results indicate that immunocompetent cells allogeneic to the mammary carcinoma cells were able to inhibit tumor development in the primary hosts and to prevent tumor growth in the adoptive recipients, which suggests that allogeneic cell therapy may be an efficient antitumor tool to eradicate minimal residual disease in human solid tumors.  相似文献

OBJECTIVE: This study was conducted to evaluate the psychological distress in patients with tinnitus that is often correlated with sleeping disorders, difficulties in concentration, and compromized social relations. METHODS: Eighty-four patients were studied using preliminary clinical and audiologic evaluations, and successive psychological tests. RESULTS: The cluster analysis indicated two essential groups composed of 45 patients (CLST-1) and 38 patients (CLST-2), respectively. The CLST-1 group had higher scores for depression, anxiety, and neuroticism. The IBQ CLST-1 revealed a greater degree hypochondria, conviction of disease dysphoria, and irritability. CONCLUSIONS: Our results indicate that the relationship between the intensity of the tinnitus and the extent of the distress is supported by the larger number of patients with more intense tinnitus in the first cluster. The CLST-2 with its normal psychological test results, apart from marked denial, would support the hypothesis of a somatic expression of the distress.  相似文献

A simple element numbering algorithm is described which yields near-minimal frontwidths for two- and three-dimensional finite element assemblies. Renumbering of E elements requires an immediate-access computer memory size which is proportional to the square root of E in two-dimensional problems, and to the two-thirds power of E in three dimensions. This review gives an account of the coordination chemistry of supramolecular azopolymer complexes. The syntheses and structures of azomonomers and their azopolymer complexes were described. Spectral techniques such as (IR, 1H-NMR, ESR) and thermal analysis were investigated. Supramolecular architectures assembled were exhibited through weak interaction including hydrogen bonding and π–π stacking. The spectral data indicate geometry of azopolymer complexes and the orbital reduction factors. ESR spectral data provided information about their structures on the basis of Hamiltonian parameters and degree of covalency. All the azopolymer complexes are ESR active due to the presence of an unpaired electron. The force constant FU–O(mdyn/Å) and the bond length RU–O (Å) of the U–O bond were calculated from the IR data and related to the electronic properties of the substituents. Wilson's method, the matrix method, Badger's formula, and the Jones and El-Sonbati equations were used to calculate the U–O bond distances from the values of the stretching and interaction force constants. The most probable correlations between U–O force constant to U–O bond distance were satisfactorily discussed in terms of Badger's rule, and the Jones and El-Sonbati equations. The thermal stability was investigated using thermogravimetric analysis. The results showed that the azopolymer complexes are mostly more stable than the homopolymer. The stability of the proton ligand/metal ligand constants in the monomeric and polymeric forms was studied carefully using potentiometery. Based on the thermodynamic functions, the dissociation process is nonspontaneous, endothermic and entropically unfavorable. The metal complexes that were formed exhibited spontaneous, endothermic and entropically favorable behavior.  相似文献
