Monitoring of excitation activity of nerve cells is very useful for not only brain research but also assessment of the effects of various chemicals, including drugs and toxins. We previously reported a novel enzyme-luminescence method for real-time monitoring of l-glutamate release from C6 glioma cells with high levels of sensitivity ( approximately 10 nM) and temporal resolution (<1 s) using a luminescence plate reader. In the present study, we tested the applicability of this novel system for assessment of effects of drugs in vitro. Several drugs (e.g., veratridine and 4-aminopyridine) were administered to C6 glioma cells for inducing glutamate release. Moreover, antagonists of voltage-dependent Ca (2+) channels (e.g., nifedipine, flunarizine, and NiCl 2) and Na (+) channels (e.g., carbamazepine and lidocaine) were applied separately for evaluating the effects of these chemicals on glutamate release from the cells. The combined effect of carbamazepine and lidocaine was also investigated by using our method, and the combined effect was found to be more potent than that of single drug administration. These results indicated that the glutamate release from C6 cells was modulated by these drugs in a way similar to that found by using several conventional analytical techniques. We therefore conclude that the developed monitoring system for real-time detection of dynamic l-glutamate release from cells could be very useful for application to assessment of drugs acting on the nervous system.  相似文献

Over 30 years ago, Fahnestock and Rich reported intriguing data showing the capability of the ribosome to polymerize phenyllactic acid. Although the polymerization was initiated and terminated randomly on polyuridic acids, the given data convincingly suggested that the generated polymer was composed of an approximately 7:3 mixture of phenyllactic acid and phenylalanine. Despite the fact that Fahnestock's conclusion was very likely correct, there have been no reports to follow up the ribosome-catalyzed polymerization of alpha-hydroxy acids until very recently. At the end of 2007, we reported messenger RNA (mRNA)-directed polyester synthesis by using the new emerging method of genetic-code reprogramming in which alpha-hydroxy acids with various kinds of side-chains are assigned to arbitrarily chosen codons. In this work, we have achieved the ribosomal synthesis of polyesters with the sequence composition and length in a fully controlled manner according to the sequence of mRNA. This Concept article describes the background of the method development and its application to the synthesis of polyesters.  相似文献

Osteolysis caused by wear particles from polyethylene in the artificial hip joints is a serious issue. We have used photo-induced radical graft polymerization to graft 2-methacryloyloxyethyl phosphorylcholine (MPC) polymer onto the surface of cross-linked polyethylene (CLPE-g-MPC) in order to reduce friction and wear at the bearing surface of the joint. The physical and mechanical properties of CLPE and CLPE-g-MPC were not significantly different, expect that the friction coefficient of untreated CLPE cups was 0.0075, compared with 0.0009 for CLPE-g-MPC cup, an 88% reduction. After 3.0 × 106 cycles in the hip joint simulator test, we could not observe any wear of CLPE-g-MPC cups. We concluded that the advantage of photo-induced radical graft polymerization technique was that the grafted MPC polymer gave a high lubricity only on the surface and has no effect on the bulk properties of the CLPE substrate.  相似文献

A novel N-linked oligosaccharide (N-glycan) with "beta1-4 bisecting branch (galactose beta1-4 bisecting N-acetylglucosamine)" was found in human serum IgG. Its structure was efficiently analyzed by using beta-galactosidase digestion, a MSn spectral library database, and negative-ion MS2 spectral matching. For confirmation, the novel N-glycan was synthesized by using an expected standard N-glycan (acceptor), UDP-galactose (donor), and beta1-4 galactosyltransferase. This work also demonstrates that the MSn spectral library database, in particular, negative-ion MS2 spectral matching, can efficiently reduce the number of specific, sequential exoglycosidase digestions required and is useful for rapid structural analysis of unknown glycans not in the database.  相似文献

We prepared polycrystalline pellets of (U,Y)O2, containing YO1.5 up to 11 mol.%. We performed indentation tests on the pellets, and evaluated the Young’s modulus and hardness. We measured the heat capacity and the thermal diffusivity, and evaluated the thermal conductivity. We succeeded in evaluating the effect of Y content on the thermophysical properties of (U,Y)O2. We revealed that the Young’s modulus, hardness, and thermal conductivity of (U,Y)O2 decreased with increasing the Y content.  相似文献

For stroke-order free online multi-stroke character recognition, stroke-to-stroke correspondence search between an input pattern and a reference pattern plays an important role to deal with the stroke-order variation. Although various methods of stroke correspondence have been proposed, no comparative study for clarifying the relative superiority of those methods has been done before. In this paper, we firstly review the approaches for solving the stroke-order variation problem. Then, five representative methods of stroke correspondence proposed by different groups, including cube search (CS), bipartite weighted matching (BWM), individual correspondence decision (ICD), stable marriage (SM), and deviation-expansion model (DE), are experimentally compared, mainly in regard of recognition accuracy and efficiency. The experimental results on an online Kanji character dataset, showed that 99.17%, 99.17%, 96.37%, 98.54%, and 96.59% were attained by CS, BWM, ICD, SM, and DE, respectively as their recognition rates. Extensive discussions are made on their relative superiorities and practicalities.  相似文献

In order to prepare phosphoric acid resins (RGPs) with large cation exchange capacities, effects of porosity and cross-linking of the precursory poly(glycidyl methacrylate-co-divinylbenzene) beads on their functionalization with phosphoric acid were studied. Two series of precursory copolymers were prepared: one was prepared by changing the amount of divinylbenzene (1–25 mol %) but by fixing that of isobutyl acetate (porogen) at 140 vol % per monomer mixture; the other by changing the amount of the porogen (40–160 vol %) but by fixing that of the cross-linker at 10 mol %. It was clarified that porosity of the precursors plays an important role in the functionalization. Highly porous precursors were functionalized with high efficiency; for example, even the precursors containing 10 mol % of divinylbenzene resulted in RGPs having cation exchange capacities as large as 6–7 meq/g, so long as BET specific surface areas of the precursors were greater than ca. 30 m2/g. The selectivity study has revealed that RGP exhibits the characteristic metal ion selectivity. Lithium ion was adsorbed in preference to sodium and potassium ions; and so-called hard Lewis acid cations, such as uranyl, ferric, and aluminum ions, are adsorbed even from strongly acidic media (1 < pH < 2).
