Experimental results are presented to show that there are very significant differences in the mean gas void fractions measured in an open tube and a annular gap bubble column, when operated at the same gas superficial velocity, using a porous sparger. Measurements were carried out in a vertical 0.102 m internal diameter column, with a range of concentric inner tubes to form an annular gap, giving diameter ratios from 0.25 to 0.69; gas superficial velocities in the range 0.014–0.200 m/s were investigated. The mean gas void fraction decreases with increasing ratio of the inner to outer diameter of the annular gap column and the transition to heterogeneous flow occurs at lower gas superficial velocities and lower void fractions. Two reasons are proposed and validated by experimental investigations: (1) the presence of the inner tube causes large bubbles to form near the sparger, which destabilise the homogeneous bubbly flow and reduce the mean void fraction; this was confirmed by deliberately injecting large bubbles into a homogeneous dispersion of smaller bubbles, and (2) the shape of the void fraction profiles changes with gap geometry and this affects the distribution parameter in the drift‐flux model. A Gram positive bacterium of the genus Rhodococcus was isolated from a contaminated site in Sydney, Australia, for its ability to tolerate and degrade high concentrations of benzene. To identify fatty acids that may impart this Rhodococcus sp. with tolerance to toxic solvents, a benzene-sensitive strain, labeled M2b, was isolated using EMS mutagenesis. A comparative analysis of fatty acid profiles showed that strain M2b was unable to increase its saturated:unsaturated ratio of fatty acids to the level achieved by the w-t strain when both strains were challenged with benzene. This was due to M2b's increased abundance of myristic acid, and decreased abundance of oleic acid. In addition, by measuring the generalized polarization of the fluorescent membrane probe laurdan using fluorescence spectroscopy, we have shown for the first time the effects of an aromatic hydrocarbon on the membrane fluidity of a Rhodococcus sp. The fluidity of the membranes increased after only 0.5 hr of exposure to benzene, thus suggesting the partitioning of benzene within the lipid bilayer. The response of this Rhodococcus sp. to benzene may suggest a mechanism for how other microorganisms survive when toxic solvents are released within the vicinity of their environment. 相似文献
Inhibition of the biosynthesis of complex N‐glycans in the Golgi apparatus influences progress of tumor growth and metastasis. Golgi α‐mannosidase II (GMII) has become a therapeutic target for drugs with anticancer activities. One critical task for successful application of GMII drugs in medical treatments is to decrease their unwanted co‐inhibition of lysosomal α‐mannosidase (LMan), a weakness of all known potent GMII inhibitors. A series of novel N‐substituted polyhydroxypyrrolidines was synthesized and tested with modeled GH38 α‐mannosidases from Drosophila melanogaster (GMIIb and LManII). The most potent structures inhibited GMIIb (Ki=50–76 μm , as determined by enzyme assays) with a significant selectivity index of IC50(LManII)/IC50(GMIIb) >100. These compounds also showed inhibitory activities in in vitro assays with cancer cell lines (leukemia, IC50=92–200 μm ) and low cytotoxic activities in normal fibroblast cell lines (IC50>200 μm ). In addition, they did not show any significant inhibitory activity toward GH47 Aspergillus saitoiα1,2‐mannosidase. An appropriate stereo configuration of hydroxymethyl and benzyl functional groups on the pyrrolidine ring of the inhibitor may lead to an inhibitor with the required selectivity for the active site of a target α‐mannosidase. 相似文献
Male coconut rhinoceros beetles,Oryctes rhinoceros (L.), produce three sex-specific compounds, ethyl 4-methyloctanoate, ethyl 4-methylheptanoate, and 4-methyloctanoic acid, the first of which is an aggregation pheromone. Synthesis of these compounds involving conjugate addition of organocuprates to ethyl acrylate is reported. In field trapping experiments, (4S)-ethyl 4-methyloctanoate and the racemic mixture were equally attractive and 10 times more effective in attracting beetles than ethyl chrysanthemumate, a previously recommended attractant. Ethyl 4-methylheptanoate was as attractive as ethyl chrysanthemumate and more attractive than 4-methyloctanoic acid, but further studies are required before it can be classed as an aggregation pheromone. Compared to ethyl 4-methyloctanoate alone, combinations of the three male-produced compounds did not increase attraction, whereas addition of freshly rotting oil palm fruit bunches to pheromone-baited traps significantly enhanced attraction. With increasing dose, captures ofO. rhinoceros increased, but doses of 6, 9, and 18 mg/day were competitive with 30 mg/day lures. Newly designed vane traps were more effective in capturing beetles than were barrier or pitfall traps. Results of this study indicate that there is potential for using ethyl 4-methyloctanoate in operational programs to controlO. rhinoceros in oil palm plantations. 相似文献
The seasonal variations in the trace element concentrations and flow rates in the Carnon River system, south west England have been investigated on a monthly basis for a period of 1 yr. Approximately 85% of Fe, Zn and As, and 45% of Cu originate from mine waters. A strong seasonal dependence of the flux of trace elements in the natural and mine waters is observed. There appears to be a direct correlation between trace element concentrations and the proportion of infiltrated surface drainage in mine waters. Rises in these concentrations in the Carnon River due to increased fluxes from mine waters in winter months are offset by higher flow rates in surface discharge.Iron is transported predominantly in the particulate phase in Carnon River waters and virtually all dissolved Fe (< 0.45 μm) precipitates in estuarine waters. Dissolved concentrations of Cu, Zn and As appear to be regulated by sorptive processes particularly with Fe oxyhydroxides in both fresh and saline waters. 相似文献
The additive manufacturing of metals requires optimisation to find the melting conditions that give the desired material properties. A key aspect of the optimisation is minimising the porosity that forms during the melting process. A corresponding analysis of pores of different types (e.g. lack of fusion or keyholes) is therefore desirable. Knowing that pores form under different thermal conditions allows greater insight into the optimisation process. In this work, two pore classification methods were trialled: unsupervised machine learning and defined limits. These methods were applied to 3D pore data from X-ray computed tomography and 2D pore data from micrographs. Data were collected from multiple alloys (Ti-6Al-4V, Inconel 718, Ti-5553 and Haynes 282). Machine learning was found to be the most useful for 3D pore data and defined limits for the 2D pore data; the latter worked by optimising the limits using energy densities.
An experimental search for dark matter particle candidates using cryogenic detectors requires a low radioactive background environment. We discuss the status of a cryogenic dark matter experiment to be performed in the Stanford Underground Facility. The detectors will be cooled in a specially designed cryostat connected to a modified side access Oxford 400 dilution refrigerator. Details of the cryostat design and its operating performance are presented. The effectiveness of the multi-level shield surrounding the cryostat, as well as the background levels we expect to achieve in the pilot experiment are discussed. Translesion replication (TR) past a cyclobutane pyrimidine dimer in Escherichia coli normally requires the UmuD'2C complex, RecA protein, and DNA polymerase III holoenzyme (pol III). However, we find that efficient TR can occur in the absence of the Umu proteins if the 3'-5' exonuclease proofreading activity of the pol III epsilon-subunit also is disabled. TR was measured in isogenic uvrA6 DeltaumuDC strains carrying the dominant negative dnaQ allele, mutD5, or DeltadnaQ spq-2 mutations by transfecting them with single-stranded M13-based vectors containing a specifically located cis-syn T-T dimer. As expected, little TR was observed in the DeltaumuDC dnaQ+ strain. Surprisingly, 26% TR occurred in UV-irradiated DeltaumuDC mutD5 cells, one-half the frequency found in a uvrA6 umuDC+mutD5 strain. lexA3 (Ind-) derivatives of the strains showed that this TR was contingent on two inducible functions, one LexA-dependent, responsible for approximately 70% of the TR, and another LexA-independent, responsible for the remaining approximately 30%. Curiously, the DeltaumuDC DeltadnaQ spq-2 strain exhibited only the LexA-independent level of TR. The cause of this result appears to be the spq-2 allele, a dnaE mutation required for viability in DeltadnaQ strains, since introduction of spq-2 into the DeltaumuDC mutD5 strain also reduces the frequency of TR to the LexA-independent level. The molecular mechanism responsible for the LexA-independent TR is unknown but may be related to the UVM phenomenon [Palejwala, V. A., Wang, G. E., Murphy, H. S. & Humayun, M. Z. (1995) J. Bacteriol. 177, 6041-6048]. LexA-dependent TR does not result from the induction of pol II, since TR in the DeltaumuDC mutD5 strain is unchanged by introduction of a DeltapolB mutation. 相似文献
