Kinetics and selectivity of supercritical carbon dioxide (SC CO2) extraction of Helichrysum italicum flowers were analyzed at pressures in the range of 10-20 MPa and temperatures of 40 °C and 60 °C (density of SC CO2 from 290 to 841 kg/m3) and also at 10 MPa and 40 °C using flowers with different moisture contents (10.5% and 28.4%). Increased moisture content of H. italicum flowers resulted in enchased solubility of solute enabling decrease of SC CO2 consumption necessary for achieving desired extraction yield. The most abundant compounds in the supercritical extracts are sesquiterpenes and waxes while monoterpenes and sesquiterpenes are the main constituents of essential oil obtained by hydrodistillation. The optimal set of working parameters with respect to extraction yield, SC CO2 consumption and chemical composition of extract were defined related to moisture content of raw material and SC CO2 density.  相似文献

Nanocomposite membranes (NCMs) were prepared by using the solution casting technique by incorporating the different amounts like 6, 8, 10 wt.% of Preyssler type heteropolyacid nanoparticles to the sodium alginate. Prepared NCMs were utilized for the sorption and permeation studies for the aqueous isopropanol solutions at different concentrations. Sorption experiment enabled the study of interactions between the components of the mixture and the polymer. Flory-Huggins interaction parameters between the permeating molecules and polymer and solubility parameters were determined to explain the sorption behavior of the NCMs. Permeation properties of the NCMs were tested by applying them for pervaporation dehydration of IPA at different concentrations. Different PV parameters like separation factor, flux, Permeance and selectivity were calculated to explain the permeation behavior of the NCMs. In this study, increased sorption, permeation and selectivity was observed for the NCMs than pristine sodium alginate membrane, indicating that NCMs are efficient for PV dehydration of the IPA. Solution-diffusion model was adopted to study the mass transfer through the membrane by calculating the diffusion coefficients.  相似文献

The association of age, smoking, alcohol, superoxide dismutase (SOD), glutathione peroxidase (GPx), blood lead (BPb) and cadmium (BCd) levels, and serum levels of copper (SCu), zinc (SZn) and selenium (SSe) with atopic status and ventilatory function was examined in the groups of 166 women and 50 men with no occupational exposure to metals or other xenobiotics. Markers of atopy included serum total IgE, skin prick test (SPT) to common inhalatory allergens, non-specific nasal reactivity (NNR) and non-specific bronchial reactivity (NBR). Parameters of ventilatory function included forced vital capacity (FVC) and forced expiratory volume in the first second (FEV(1)). Significantly higher BPb, SZn, IgE and prevalence of positive SPT, and lower SCu and NNR was found in men than in women. Fifteen women taking female sex hormones (HT) had significantly higher SCu than women without HT. Regression models showed significant inverse associations between IgE and SCu (P=0.021) and NNR and SCu (P=0.044) in women. When excluding women with HT, the association of SCu and total IgE became of borderline significance (P=0.051), association between SCu and NNR disappeared, and significant positive association between total IgE and BPb emerged (P=0.046). In men, significant inverse association was found between positive SPT and SSe, and between NBR and SSe. A decrease in FVC% and FEV(1)% was associated with an increase in smoking intensity (P<0.001) and a decrease in SZn (P=0.043 and P=0.053, respectively). These results were observed at the levels of the metals comparable to those in general populations worldwide. The observed differences between men and women may partly be explained by different levels of relevant toxic and essential metals, and their combination. The role of female HT in associations of atopy markers and SCu should be further investigated.  相似文献

Glycosphingolipids (GSLs), composed of a hydrophilic carbohydrate chain and a lipophilic ceramide anchor, play pivotal roles in countless biological processes, including the development of cancer. As part of the investigation of the vertebrate glycome, GSL analysis is undergoing rapid expansion owing to the application of modern mass spectrometry. Here we introduce direct coupling of IR-MALDI-o-TOF mass spectrometry with the TLC overlay binding assay for the structural characterization of GSLs. We matched three complementary methods including (i) TLC separation of GSLs, (ii) their detection with oligosaccharide-specific proteins, and (iii) in situ MS analysis of protein-detected GSLs. The high specificity and sensitivity is demonstrated by use of antibodies, bacterial toxins, and a plant lectin. The procedure works on a nanogram scale, and detection limits of less than 1 ng at its best of immunostained GSLs were obtained. Furthermore, only crude lipid extracts of biological sources are required for TLC-IR-MALDI-MS, omitting any laborious GSL downstream purification procedures. This strategy was successfully applied to the identification of cancer-associated GSLs in human hepatocellular and pancreatic tumors. Thus, the in situ TLC-IR-MALDI-MS of immunolabeled GSLs opens new doors by delivering specific structural information of trace quantities of GSLs with only a limited investment in sample preparation.  相似文献

This study was performed to evaluate the phytochemical composition, in vitro antioxidant capacity, antihyperglycaemic and anti‐inflammatory activities, and simulated gastrointestinal digestion of 7‐day‐old freeze‐dried barley sprouts (BS), one hybrid and one nonhybrid variety: ‘NS565’ (BSNS) and ‘Golozrni’ (BSG), respectively. BSNS expressed significantly higher (P ≤ 0.05) content of total phenols, chlorophyll and carotenoids. Phenolic compounds were the most dominant bioactives in both BSNS and BSG (713.25 and 479.02 mg GAE 100 g?1 DW, respectively). BSNS possessed significantly higher (P ≤ 0.05) antioxidant capacity, evaluated by DPPH and ABTS assays, and reducing power ( = 0.54 mg mL?1; IC50ABTS = 0.79 mg mL?1; RP0.5 = 9.35 mg mL?1). Antihyperglycaemic and anti‐inflammatory activities of BSNS ( = 1.43 mg mL?1;  = 1.86 mg mL?1) were also significantly higher (P ≤ 0.05) than BSG ( = 1.97 mg mL?1;  = 4.40 mg mL?1). In vitro simulation of gastrointestinal digestion showed higher release of phenolic compounds in intestinal fluid than in gastric fluid.  相似文献

The qualitative performance characteristics of a qPCR-based method to detect human adenoviruses in raspberries were determined through a collaborative trial involving 11 European laboratories. The method incorporated a sample process control (murine norovirus) and an internal amplification control. Trial sensitivity or correct identification of 25-g raspberry samples artificially contaminated with between 5?×?102 and 5?×?104?PFU was 98.5%; the accordance and concordance were 97.0%. The positive predictive value was 94.2%. The trial specificity or percentage correct identification of non-artificially contaminated samples was 69.7%; the accordance was 80.0% and the concordance was 61.7%. The negative predictive value was 100%. Application of a method for the detection of human adenoviruses in food samples could be useful for routine monitoring for food safety management. It would help to determine if a route of contamination exists from human source to food supply chain which pathogenic viruses such as norovirus and hepatitis A virus could follow.  相似文献

The aim of this study was to develop optimal NIRS calibration for ash content prediction in legumes by using the thermogravimetric (TGA) and gravimetric (GA) analytical methods. The calibration was performed on the basis of whole and structured sample sets (n = 143 and n = 99, respectively). Samples were scanned using a Rapid Content Analyzer in reflectance mode (400–2500 nm). Different mathematical treatments of the spectra preceded modified partial least squares (MPLS) regression analyses. The performance of the models was assessed by cross validation and external validation (n = 44). Models developed for the whole sample set on the basis of the TGA and GA methods were characterised by standard error of calibration (SEC) ranged from 0.28 to 0.50, standard error of cross validation (SECV) ranged from 0.43 to 0.60, coefficient of determination (R2) ranged from 0.97 to 0.89, explained variance (1 − VR) ranged from 0.94 to 0.85 and residual predictive deviation (RPD) ranged from 4.23 to 2.68, respectively. Models developed for the structured sample set on the basis of the TGA and GA methods were characterised by standard error of calibration (SEC) ranged from 0.32 to 0.42, standard error of cross validation (SECV) ranged from 0.53 to 0.56, coefficient of determination (R2) ranged from 0.97 to 0.94, explained variance (1 − VR) ranged from 0.91 to 0.89 and residual predictive deviation (RPD) ranged from 3.52 to 2.98, respectively. The obtained results showed the potential of NIRS method to accurately predict the ash content of legume grass samples that correspond to ash content determined by the TGA and GA methods.  相似文献
