The diversity of the free-living nitrogen-fixing cyanobacterial community in the floodplain sediments along the Solimões and Amazon Rivers and some of their tributaries (Japurá, Negro and Madeira) was investigated. Five cyanobacterial genera were morphologically identified, four of which (Nostoc, Calothrix, Cylindrospermum and Fischerella) have not previously been isolated from the Brazilian Amazon floodplain. Nostoc strains were the most commonly found heterocyst-forming cyanobacteria. Five strains (N. muscorum CENA18 and CENA61, N. piscinale CENA21, Cylindrospermum sp. CENA33 and Fischerella sp. CENA19) were selected for growth measurement, ability to fix N2 and phylogenetic analysis, based on their widespread distribution and morphological distinction. Molecular analyses employing 16S rRNA sequences indicated that some of the isolates may represent novel cyanobacterial species. Dinitrogen fixed by these strains was measured indirectly as acetylene reduction activity and ranged from 11.5 to 22.2 nmol C2H4 μg Chl a−1 h−1. These results provide evidence of widespread and importance of nitrogen-fixing cyanobacteria as a source of N inputs in the Amazonian ecosystem.  相似文献

Organochlorine and organophosphorous pesticide concentrations have been monitored in the Paraná River, Argentina, at a point 600 km upstream from its mouth, since March 1981. Chromatographic analyses performed up to December 1981 indicate that parathion, lindane and α-BHC (benzene hexachloride) are the most ubiquitous species with mean concentrations of 22, 9, and 9 ng 1?1, respectively. The dissolved-in-water pesticides do not show a clear association with discharge — only lindane, which is adsorbed on the sediments, exhibits a significant positive correlation with discharge. Other chemical compounds (heptachlor, aldrin, methylparathion) have been occasionally determined in the examined samples. Total discharge-weighted mean concentrations for dissolved (175 ng l?1) and adsorbed (453 ng g?1) compounds allow the calculation of a mass transport rate of 10.85 × 107 g y?1. a figure valid for the period of study.  相似文献

In this paper, we propose and analyze by means of simulations the use of surrogate data algorithms for blind detection of nonlinearities in multiple-echo ultrasonic signals.We assume a blind scheme so that no information about the input (emitted ultrasonic pulse) can be used. The metrics and equations that model some nonlinear situations are carefully reviewed. Also, closed form equations of the third-order metrics from a simplified second-order Volterra kernel are derived. Computer simulations show that the surrogate data technique is a potentially powerful tool for blind detection of nonlinearities in multiple-echo ultrasonic signals if adequate metrics are chosen. They also reveal interesting trade-offs among parameters that model ultrasonic systems and detection percentages.  相似文献

This study aimed at investigating in vitro osteogenesis on three fluorcanasite glass-ceramic compositions with different solubilities (K3, K5, and K8). Osteoblastic cells were obtained from human alveolar bone fragments and cultured under standard osteogenic condition until subconfluence. First passage cells were cultured on K3, K5, and K8 and on Bioglass((R)) 45S5 (45S5-control). Cell adhesion was evaluated at 24 h. For proliferation and viability, cells were cultured for 1, 4, and 10 days. Total protein content and alkaline phosphatase (ALP) activity were measured at 7, 14, and 21 days. Cultures were stained with Alizarin red at 21 days, for detection of mineralized matrix. Data were compared by ANOVA followed by Duncan's test. Cell adhesion, cell proliferation, viability, total protein content, and ALP activity were not affected by fluorcanasite glass-ceramic composition and solubility. Bone-like formation was similar on all fluorcanasite glass-ceramics and was reduced compared to 45S5. The changes in the chemical composition and consequently solubility of the fluorcanasite glass-ceramics tested here did not significantly alter the in vitro osteogenesis. Further modifications of the chemical composition of the fluorcanasite glass-ceramic would be required to improve bone response, making this biomaterial a good candidate to be employed as a bone substitute.  相似文献

X-ray fluorescence microscopy (microXRF) is applied for the first time to study macrophages exposed to unpurified and purified single-walled (SW) and multiwalled (MW) carbon nanotubes (CNT). Investigating chemical elemental distributions allows one to (i) image nanotube localization within a cell and (ii) detect chemical modification of the cell after CNT internalization. An excess of calcium is detected for cells exposed to unpurified SWCNT and MWCNT and related toxicological assays are discussed.  相似文献

Synthesis and tribological evaluation of three tetraalkylammonium thiomolybdate (R4N)2MoS4 (R = methyl, propyl, or ammonia) aqueous-based lubricant additives on a ball-on-disk tribometer was carried out for a steel–aluminum contact. Tests were performed at the same conditions of load, entrainment speed, sliding distance, temperature, and concentration of MoS2 to compare the activity (lubrication effect) of the thiomolybdates prepared. A friction reduction is observed for the three salts compared to pure water; however, significant differences in friction coefficient are observed depending on the alkyl group. SEM/EDAX and Raman analysis of the wear tracks reveal the in-contact formation of a MoS2-lubricating film, rich in molybdenum and sulfur.  相似文献

The alarming growth of the antibiotic-resistant superbugs methicillin-resistant Staphylococcus aureus (MRSA) and vancomycin-resistant Enterococcus (VRE) is driving the development of new technologies to investigate antibiotics and their modes of action. We report the label-free detection of vancomycin binding to bacterial cell wall precursor analogues (mucopeptides) on cantilever arrays, with 10 nM sensitivity and at clinically relevant concentrations in blood serum. Differential measurements have quantified binding constants for vancomycin-sensitive and vancomycin-resistant mucopeptide analogues. Moreover, by systematically modifying the mucopeptide density we gain new insights into the origin of surface stress. We propose that stress is a product of a local chemical binding factor and a geometrical factor describing the mechanical connectivity of regions activated by local binding in terms of a percolation process. Our findings place BioMEMS devices in a new class of percolative systems. The percolation concept will underpin the design of devices and coatings to significantly lower the drug detection limit and may also have an impact on our understanding of antibiotic drug action in bacteria.  相似文献

Flow Structure at Different Stages in a Meander-Bend with Bendway Weirs   总被引:3,自引:0,他引:3  
Streambank erosion is an important management issue, particularly for meandering rivers. Recently, bendway weirs have become popular control measures for bank erosion along small meandering streams in the agricultural Midwest. Although these structures have successfully mitigated bank erosion in some cases, there is evidence that the weirs do not always perform as anticipated. Scientific understanding of how bendway weirs influence flow dynamics, streambank erosion, and aquatic habitat is limited. Current design criteria are based primarily on expert judgment rather than a formalized technical design procedure. At field-scale studies, the present paper represents a first step toward an integrated geomorphological and engineering evaluation of the performance of bendway weirs in rivers. To accomplish this initial phase, three-dimensional (3D) velocity data were collected on Sugar Creek at Brookside Farm, Ill., and 3D numerical simulations for low-flow conditions were performed to validate the computational fluid dynamic model. Overall results show good agreement between measured and simulated data for streamwise velocities and turbulence kinetic energy. The model is less accurate at predicting the velocity and turbulence kinetic energy in the shear layer immediately downstream from the weir tips. Based on the validation for low-flow condition, 3D simulations were carried out for medium and high flows where the bendway weirs are completely submerged. These simulations indicate that 3D patterns of flow, especially flow near the outer bank, change dramatically with changes in flow stage. Flow patterns at high-flow condition indicate that bank retreat over the tops of weirs is associated with locally high-shear stresses, thus producing a “shelf” along the base of the outer bank as observed in the field.  相似文献

This paper presents the Cantabrian portable infiltrometer (CP infiltrometer), a specially designed device based on rainfall simulation for the assessment of the infiltration capacity of all types of urban pavements. Several pervious and impervious surfaces were tested with the LCS permeameter, an existing infiltration test based on the use of a column of water, and the CP Infiltrometer, simulating rain intensities with return periods of 10, 50, and 500 years and 5?min duration. The discussion of the results indicates that the CP infiltrometer could be used successfully to identify different levels of infiltration capacity and to assess the correct performance of pervious surfaces on which design, construction, and maintenance decisions are based.  相似文献

The aim of this study was to evaluate the effectiveness of treating depression with coping-oriented couples therapy (COCT) as compared with cognitive-behavioral therapy (CBT; A. T. Beck, C. Ward, & M. Mendelson, 1961) and interpersonal psychotherapy (IPT; M. M. Weissman, J. C. Markowitz, & G. L. Klerman, 2000). Sixty couples, including 1 clinically depressed partner, completed pre- and posttest questionnaires as well as follow-up assessments at 6-month intervals over the subsequent 1.5 years. Effects of the 3 treatments on depressive symptomatology assessed by the Beck Depression Inventory (A. T. Beck, A. J. Rush, B. L. Shaw, & G. Emery, 1979) and Hamilton Rating Scale for Depression (M. Hamilton, 1960); recovery rates; and relapse rates were examined. Additionally, changes in relationship quality were evaluated. Results suggest that the COCT is as effective in improving depressive symptomatology as are the well-established, evidenced-based CBT and IPT approaches. The COCT did not demonstrate a significantly better outcome with regard to self-reported relationship satisfaction or dyadic coping; however, it did produce significant improvements in partners' expressed emotion, changes that were not seen in other treatment conditions.
