Laser-based optical diagnostics, such as planar laser-induced fluorescence and, especially, Raman imaging, often require selective spectral filtering. We advocate the use of an imaging spectrograph with a broad entrance slit as a spectral filter for two-dimensional imaging. A spectrograph in this mode of operation produces output that is a convolution of the spatial and spectral information that is present in the incident light. We describe an analytical deconvolution procedure, based on Bayesian statistics, that retrieves the spatial information while it avoids excessive noise blowup. The method permits direct imaging through a spectrograph, even under broadband illumination. We introduce the formalism and discuss the underlying assumptions. The performance of the procedure is demonstrated on an artificial but pathological example. In a companion paper [Appl. Opt. 43, 5682-5690 (2004)] the method is applied to the practical case of fuel equivalence ratio Raman imaging in a combustible methane-air mixture.  相似文献

This paper discusses code verification of Reynolds-Averaged Navier Stokes (RANS) solvers with the method of manufactured solutions (MMS). Examples of manufactured solutions (MSs) for a two-dimensional, steady, wall-bounded, incompressible, turbulent flow are presented including the specification of the turbulence quantities incorporated in several popular eddy-viscosity turbulence models. A wall-function approach for the MMS is also described. The flexiblity and usefulness of the MS is illustrated with calculations performed in three different exercises: the calculation of the flow field using the manufactured eddy-viscosity; the calculation of the eddy-viscosity using the manufactured velocity field; the calculation of the complete flow field coupling flow and turbulence variables. The results show that the numerical performance of the flow solvers is model dependent and that the solution of the complete problem may exhibit different orders of accuracy than in the exercises with no coupling between the flow and turbulence variables.  相似文献

DNA replication is essential to maintain genome integrity in S phase of the cell division cycle. Accumulation of stalled replication forks is a major source of genetic instability, and likely constitutes a key driver of tumorigenesis. The mechanisms of regulation of replication fork progression have therefore been extensively investigated, in particular with DNA combing, an optical mapping technique that allows the stretching of single molecules and the mapping of active region for DNA synthesis by fluorescence microscopy. DNA linearization in nanochannels has been successfully used to probe genomic information patterns along single chromosomes, and has been proposed to be a competitive alternative to DNA combing. Yet this conjecture remains to be confirmed experimentally. Here, two complementary techniques are established to detect the genomic distribution of tracks of newly synthesized DNA in human cells by optical mapping in nanochannels. Their respective advantages and limitations are compared, and applied them to detect deregulations of the replication program induced by the antitumor drug hydroxyurea. The developments here thus broaden the field of applications accessible to nanofluidic technologies, and can be used in the future as part for molecular diagnostics in the context of high throughput cancer drug screening.  相似文献

This paper shows how the solution of the standard predictive control problem can be recast as a continuous function of the state, the reference signal, the noise and the disturbances, and hence can be approximated arbitrarily closely by a feed-forward neural network. The existence of such a continuous mapping eliminates the need for linear independency of the active constraints, and therefore the resulting analytic constrained predictive controller will combine constraint handling with speed while being applicable to fast and complex control systems with many constraints. The effectiveness of the proposed controller design methodology is shown for a simulation example of an elevator model and for a real-time laboratory inverted pendulum system.  相似文献

Trafficking and metabolism of sphingolipids were examined in undifferentiated (G+) and differentiated (G+ reversed) HT29 human colon adenocarcinoma cell lines. Metabolic experiments employing a fluorescently labeled sphingolipid precursor, 6-[N-(7-nitro-2,1,3-benzoxadiazol-4-yl)amino]hexanoylceramide++ + (C6-NBD-ceramide) revealed that both qualitative and quantitative differences exist in sphingolipid synthesis between the 2 cell lines. One of the C6-NBD-sphingolipids synthesized in G+ cells is not found in the G+ reversed cells. Furthermore, the ratio of the 2 main products, C6-NBD-glucosylceramide and C6-NBD-sphingomyelin, differs: in G+ cells glucosylceramide is by far the main product, whereas G+ reversed cells synthesize C6-NBD-sphingomyelin in slight excess. Once established, these ratios of sphingolipids are quickly restored metabolically when distortion of the ratio is caused by experimental manipulation. This indicates that they represent a true metabolic equilibrium situation of the 2 sphingolipids in these cells, while the distinct ratios are mainly determined by the NBD-lipid pool in the plasma membrane. Preferential synthesis and transfer of glucosylceramide from its site of synthesis to the cell surface do not occur when the plasma membrane pool of glucosylceramide is selectively removed. This suggests that instantaneous replenishment via specific signalling is probably not involved as a mechanism in re-establishing perturbed lipid pools. In conjunction with observations on distinct lipid trafficking pathways of glucosylceramide in G+ and G+ reversed cells, the present metabolic studies emphasize a relation between the expression of this glycolipid and the state of differentiation of HT29 cells.  相似文献

The yeast alcohol acetyl transferase I, Atf1p, is responsible for the major part of volatile acetate ester production in fermenting Saccharomyces cerevisiae cells. Some of these esters, such as ethyl acetate and isoamyl acetate, are important for the fruity flavours of wine, beer and other fermented beverages. In order to reveal the subcellular localization of Atf1p and further unravel the possible physiological role of this protein, ATF1::GFP fusion constructs were overexpressed in brewer's yeast. The transformant strain showed a significant increase in acetate ester formation, similar to that of an ATF1 overexpression strain, indicating that the Atf1p-GFP fusion protein was active. UV fluorescence microscopy revealed that the fusion protein was localized in small, sphere-like organelles. These organelles could be selectively stained by the fluorescent dye Nile red, indicating that they contained high amounts of neutral lipids and/or sterols, a specific characteristic of yeast lipid particles. Purification of lipid particles from wild type and ATF1 deletion cells confirmed that the Atf1p-GFP fusion protein was located in these organelles. Furthermore, a clear alcohol acetyl transferase activity could be measured in the purified lipid particles of both wild type and transformed cells. The localization of Atf1p in lipid particles may indicate that Atf1p has a specific role in the lipid and/or sterol metabolism that takes place in these particles.  相似文献

In this paper, we propose a logic of argumentation for the specification and verification (LA4SV) of requirements on Dung??s abstract argumentation frameworks. We distinguish three kinds of decision problems for argumentation verification, called extension verification, framework verification, and specification verification respectively. For example, given a political requirement like ??if the argument to increase taxes is accepted, then the argument to increase services must be accepted too,?? we can either verify an extension of acceptable arguments, or all extensions of an argumentation framework, or all extensions of all argumentation frameworks satisfying a framework specification. We introduce the logic of argumentation verification to specify such requirements, and we represent the three verification problems of argumentation as model checking and theorem proving properties of the logic. Moreover, we recast the logic of argumentation verification in a modal framework, in order to express multiple extensions, and properties like transitivity and reflexivity of the attack relation. Finally, we introduce a logic of meta-argumentation where abstract argumentation is used to reason about abstract argumentation itself. We define the logic of meta-argumentation using the fibring methodology in such a way to represent attack relations not only among arguments but also among attacks. We show how to use this logic to verify the requirements of argumentation frameworks where higher-order attacks are allowed [A preliminary version of the logic of argumentation compliance was called the logic of abstract argumentation?(2005).]  相似文献

This article describes three generic models and application methods that can be used to analyze temperature development in an electronic product during conceptual design. The models are based on generally known heat transfer and resistor–capacitor network theory and are theoretically and numerically approximated. The result is three easy-to-use tools for conceptual thermal design. Application of the models in design practice has been assessed using a usability experiment and several in-depth interviews with industrial design engineers from the field.  相似文献

A novel process concept for the oxidative coupling of methane followed by the oligomerization to liquids has been developed within the frame of the EU integrated project OCMOL. This technology is based on process intensification principles via cutting‐edge structured microreactor technology. It is also a fully integrated industrial process through the re‐use and the recycling of by‐products, in particular CO2, at every process stage. The focus of this contribution is on the reaction engineering aspects of the core steps, i.e., catalysts, kinetics and reactor design for the methane coupling and reforming.  相似文献
