The purpose of this study was to assess the risk of Vibrio spp. transmission from crustaceans to humans in two coastal towns of C?te d'Ivoire. Bacteriologic analysis was performed on 322 crustacean samples obtained from six markets in Abidjan and one in Dabou. Suspected Vibrio colonies were identified by morphological, cultural, biochemical, and molecular tests and matrix-assisted laser desorption ionization-time-of-flight mass spectrometry. PCR assays were used to further characterize Vibrio strains. A survey on consumption of crustaceans was conducted among 120 randomly selected households in Abidjan. Overall, Vibrio spp. were isolated from 7.8% of the crustacean samples studied, at levels as high as 6.3 log CFU/g. Of the Vibrio strains identified, 40% were V. alginolyticus, 36% were V. parahaemolyticus, and 24% were nontoxigenic V. cholerae; the latter two species can cause mild to severe forms of seafood-associated gastroenteritis. Among interviewed households, 11.7% reported daily consumption of crustaceans, confirming the high probability of exposure of human population to Vibrio spp., and 7.5% reported symptoms of food poisoning after consumption of crustaceans. The absence of genes encoding major virulence factors in the studied strains, i.e., cholera toxin (ctxA and ctxB) in V. cholerae and thermostable direct hemolysin (tdh) and thermostable direct hemolysin-related hemolysin (trh) in V. parahaemolyticus, does not exclude the possibility of exposure to pathogenic strains. However, human infections are not common because most households (96.7%) boil crustaceans, usually for at least 45 min (85.9% of households) before consumption.  相似文献

Spatial and temporal distribution of singlet oxygen in Lake Superior   总被引:2,自引:0,他引:2  
A multiyear field study was undertaken on Lake Superior to investigate singlet oxygen ((1)O(2)) photoproduction. Specifically, trends within the lake were examined, along with an assessment of whether correlations existed between chromophoric dissolved organic matter (CDOM) characteristics and (1)O(2) production rates and quantum yields. Quantum yield values were determined and used to estimate noontime surface (1)O(2) steady-state concentrations ([(1)O(2)](ss)). Samples were subdivided into three categories based on their absorbance properties (a300): riverine, river-impacted, or open lake sites. Using calculated surface [(1)O(2)](ss), photochemical half-lives under continuous summer sunlight were calculated for cimetidine, a pharmaceutical whose reaction with (1)O(2) has been established, to be on the order of hours, days, and a week for the riverine, river-impacted, and open lake waters, respectively. Of the CDOM properties investigated, it was found that dissolved organic carbon (DOC) and a300 were the best parameters for predicting production rates of [(1)O(2)](ss). For example, given the correlations found, one could predict [(1)O(2)](ss) within a factor of 4 using a300 alone. Changes in the quantum efficiency of (1)O(2) production upon dilution of river water samples with lake water samples demonstrated that the CDOM found in the open lake is not simply diluted riverine organic matter. The open lake pool was characterized by low absorption coefficient, low fluorescence, and low DOC, but more highly efficient (1)O(2) production and predominates the Lake Superior system spatially. This study establishes that parameters that reflect the quantity of CDOM (e.g., a300 and DOC) correlate with (1)O(2) production rates, while parameters that characterize the absorbance spectrum (e.g., spectral slope coefficient and E2:E3) correlate with (1)O(2) production quantum yields.  相似文献

Abstract: Sustained colonic fermentation supplies beneficial fermentative by‐products to the distal colon, which is particularly prone to intestinal ailments. Blunted/delayed initial fermentation may also lead to less bloating. Previously, we reported that starch‐entrapped alginate‐based microspheres act as a slowly fermenting dietary fiber. This material was used in the present study to provide a benchmark to compare to other “slowly fermentable” fibers. Dietary fibers with previous reports of slow fermentation, namely, long‐chain inulin, psyllium, alkali‐soluble corn bran arabinoxylan, and long‐chain β‐glucan, as well as starch‐entrapped microspheres were subjected to in vitro upper gastrointestinal digestion and human fecal fermentation and measured over 48 h for pH, gas, and short‐chain fatty acids (SCFA). The resistant fraction of cooked and cooled potato starch was used as another form of fermentable starch and fructooligosaccharides (FOS) served as a fast fermenting control. Corn bran arabinoxylan and long‐chain β‐glucan initially appeared slower fermenting with comparatively low gas and SCFA production, but later fermented rapidly with little remaining in the final half of the fermentation period. Long‐chain inulin and psyllium had slow and moderate, but incomplete, fermentation. The resistant fraction of cooked and cooled potato starch fermented rapidly and appeared similar to FOS. In conclusion, compared to the benchmark slowly fermentable starch‐entrapped microspheres, a number of the purported slowly fermentable fibers fermented fairly rapidly overall and, of this group, only the starch‐entrapped microspheres appreciably fermented in the second half of the fermentation period. Practical Application: Consumption of dietary fibers, particularly commercial prebiotics, leads to uncomfortable feelings of bloating and flatulence due to their rapid degradation in our large intestine. This article employs claimed potential slowly fermenting fibers and compares their fermentation rates with a benchmark slow fermenting fiber that we fabricated in an in vitro simulation of the human digestive system. Results show a variety of fermentation profiles only some of which have slow and extended rate of fermentation.  相似文献

The SET-RPLA is a commercially available kit for the detection of staphyloccal enterotoxins in foods. Previous reports have shown that non-specific reactions occur on use of the kit with cheeses and thereby restrict its use. In this study a variety of dairy products were tested and non-specific reactions were found to be associated with those products rennetted during manufacture. These reactions can be obviated by addition of 10 mM sodium hexametaphosphate to the diluent provided in the kit, without affecting the ability to detect staphyloccal enterotoxins in dairy products. The sensitivity of the SET-RPLA was demonstrated to be 0.25 ng/ml.  相似文献

Responses to hydrocarbon stress of four tropical plants Panicum maximum, Zea mays, Centrosema sp. and Pueraria sp. grown in crude oil contaminated soils (1%, 5% and 10% w/w) were evaluated in a green house. Plants’ percentage survival, shoot heights, biomass development, and phytotoxicity susceptibility were used as indicators of growth, stress response and hydrocarbon tolerance. Relative to control, shoot heights and biomass of plants reduced with increasing hydrocarbon concentration, but 1% w/w oil-in-soil, stimulated shoot heights (5.9% and 6.4%) and weights (21.9% and 2.3%) in P. maximum and Centrosema sp. respectively. P. maximum tolerated the contaminant stress with biomass yields of 113% and 57% of control respectively in 1% and 10% w/w oil-in-soil. All the plants had 100% survival in 1% w/w, but considerably reduced survival in 10% w/w oil-in-soil. These results show that P. maximum has great potential for phytoremediation of petroleum contaminated soil.  相似文献

A commonly used process for forming low-resistance contacts to thin-film p-type CdTe involves the formation of a Te layer by etching the CdTe film in a concentrated mixture of nitric and phosphoric acids. The authors compare evaporated Te back contacts with ‘control’ back contacts formed by the usual etching process, and demonstrate that evaporating Te onto a CdTe thin film is a viable process for forming a low-resistance contact. The best efficiency achieved for a CdTe solar cell made with an evaporated Te back contact is 12.1%, whereas the efficiency of the device made with the control back contact was 11.9%. The evaporation process offers numerous advantages over acid etching, most notably vacuum compatibility amenable to large-scale production of CdTe solar cell modules.  相似文献

In July 1996, the US Department of Agriculture (USDA), Food Safety and Inspection Service (FSIS), published the Pathogen Reduction; Hazard Analysis and Critical Control Point (HACCP) Systems final rule to improve food safety of meat and poultry products. The final rule established, among other requirements, pathogen reduction performance standards for Salmonella for food animal carcasses and raw ground products. The final rule is to be fully implemented in three stages in successively smaller federally inspected meat and poultry slaughter and processing establishments. Implementation began in January 1998 and was completed in January 2000. Samples of carcasses of four species of food animals (cattle, swine, chickens, turkeys), and raw ground product from each of these species, were collected by FSIS from establishments prior to implementation of the final rule and cultured for Salmonella. This paper reports Salmonella serotype results of samples collected from June 1997 through August 1998. These results represent a baseline for future comparisons.  相似文献
