This study investigated the influence of inoculum levels and manufacturing methods on the survival of Campylobacter (C.) jejuni in raw fermented turkey sausages. Sausages were prepared and inoculated with C. jejuni. After inoculation, these sausages were processed and ripened for 8 days. Samples were taken throughout the ripening process. The presence of C. jejuni was established bacteriologically. Additionally, lactic acid bacteria were enumerated, pH values and water activity were measured to verify the ripening process. To detect changes in genotype and verify the identity of the recovered clones, AFLP analysis was carried out on the re-isolated strains. Whereas no C. jejuni were detectable when inoculating the sausages with the lowest inoculum (0.08-0.44 log(10) cfu/g sausage emulsion), C. jejuni were detectable for 12-24h by enrichment when inoculated with approximately 2 log(10) cfu/g. After inoculation with 4 and 6 log(10) cfu/g respectively, C. jejuni were detectable without enrichment for 12-48 h and by enrichment for 144 h at the most. The greatest decrease of the C. jejuni population occurred during the first 4 h of ripening. Only a very high inoculum level allowed the survival of the organism during a fermentation process and during ripening to pose a potential risk for consumers. Lower initial Campylobacter inoculums will be eliminated during proper ripening of the sausages, if sufficient decrease in water activity and pH-value is ensured.  相似文献

We present transport measurements of single-molecule junctions bridged by a molecule with three benzene rings connected by two double bonds and with thiol end-groups that allow chemical binding to gold electrodes. The I-V curves show switching behavior between two distinct states. By statistical analysis of the switching events, we show that a 300 meV mode mediates the transition between the two states. We propose that breaking and reformation of a S-H bond in the contact zone between molecule and electrode explains the observed bistability.  相似文献

Low-absorption optics made of OH-reduced fused silica are a key technology for future gravitational wave detectors such as the Advanced LIGO. We developed a sensitive method to measure the absorption inside the beam splitter of the GEO 600, taking advantage of the effect of thermal lensing. Using this method, we derived a bulk absorption of less than 0.25 ppm/cm for a piece of Suprasil 311 SV at a wavelength of 1064 nm. To the best of our knowledge, this is the lowest value of light absorption inside fused silica reported so far in the literature.  相似文献

An estimation of the heavy metal and anion mass-balance was made for municipal solid waste incinerator bottom ash deposited at a construction and industrial waste landfill. The mass-balance was found by comparing the content of metals and anions in the landfill leachate to the metal and anion content in the deposited bottom ash. The discharge of heavy metals ranged from 0.001% for Pb to 0.55% for Cr, which is approximately at the same level as in regular municipal solid waste (MSW) landfills. Landfilled organic material and silicates from construction waste might have contributed to the retention of metals. Chloride, and to a lesser extent sulphate, appeared to be readily released from the landfill. It was estimated that a mass corresponding to 80% of the Cl- and 18% of the SO(4)2- in the bottom ash was discharged annually. Low retention, especially of chloride, may lead to a rapid decline in the discharge of this ion in the future when the landfilling of bottom ash is discontinued.  相似文献

Case-based learning is ascribed high potential with respect to the education of pre-service teachers as well as the further education of experienced in-service teachers, as it can provide opportunities for the application of professional knowledge to authentic classroom situations. In addition to the application and acquisition of knowledge, the aspect of knowledge convergence (i.e., increasing similarity of collaborative learners with respect to their knowledge) should be taken into account when homogeneous benefits are desired for all participants of a case-based training. A field study with 29 pre-service teachers and 24 in-service teachers from adult education was conducted to examine effects of different levels of teaching experience on knowledge application, knowledge acquisition, and knowledge convergence in a computer-supported collaborative case-based learning environment. An innovative approach to measuring knowledge convergence was introduced, using the positions of annotations (i.e., flags placed on the timeline of the case video) made by learners during their case analyses as an indicator of their shared focus of attention. This measure was found to positively correlate with the application and acquisition of professional knowledge. Teaching experience affected both processes and outcomes of case-based learning, suggesting different means of instructional support for pre-service and in-service teachers.  相似文献

Over the last two decades, numerous approaches have been developed for automating construction safety assessment, that is, prevention through design and planning (PtD/P), that overcome the limitations of a purely manual safety assessment process. Despite this, such automated approaches are yet to be widely adopted in practice. Barriers include a lack of standardization of construction safety concepts within regulatory frameworks and a lack of confidence in automated approaches on the part of practitioners. This work presents a freely available 4D BIM-based benchmark model (3D Building Information Model and 68 schedules) and construction safety ontology covering the two most frequent and severe construction site accidents: falls from height and struck-by falling object hazards. The benchmark model and ontology enable the construction safety community to test and compare the performance of alternative automated approaches. Moreover, they are utilized in our PtD/P tool SafeConAI, and empirical evaluation results show how the safety situation changes among different construction schedules of the same project.  相似文献
