Two monoclonal antibodies which can be used for the unambiguous identification by fluorescence microscopy of Encephalitozoon intestinalis spores in clinical specimens are described. Monoclonal antibody Si91 is specific for the extruded polar filament, and Si13 recognizes the surfaces of E. intestinalis spores. No cross-reaction with spores of Encephalitozoon hellem was observed. Immunogold electron microscopy confirmed the specific reactivities of both antibodies. Combined in an indirect immunofluorescence assay, these antibodies are used to identify spores in feces. Although there was some cross-reaction with fecal bacteria and fungi, the typical morphology of the extruded polar filaments enabled proper identification of the E. intestinalis spores. Parasites could also be demonstrated to be present in urine, nasal swabs, lung brush biopsy specimens, and bronchoalveolar lavage fluid from a patient with disseminated infection with E. intestinalis. The use of these monoclonal antibodies facilitates the detection and species determination of E. intestinalis in clinical specimens.  相似文献

A high accuracy current source has been developed for the generation of very small currents (below 200 pA) based on applying a linear voltage ramp to a capacitor. Using software-controlled compensation for the nonlinearity of the voltage ramp, a stable current can be reached within a few seconds. A voltage ramp within typically 10/spl middot/10/sup -6/ of any desired value between less than 1 mV/s and several hundreds of mV/s can be realized. Using a high quality air-dielectric capacitor, currents below 200 pA can be generated with an uncertainty of 20 /spl mu/A/A (k = 2), with a lower limit of 2.0 aA (k = 2).  相似文献

This paper addresses the problems that lawyers experience retrieving information from legal-text databases. Traditional access mechanisms of text databases require users to know how information is stored. We propose a method for index organisation which shields lawyers from the internal storage structures and which allows them to address the legal databases in their own legal terms. The proposed index is based on a model of legal tasks as opposed to traditional database indexes which represent the contents of the database. We will lay out the architecture of an information system in which this task model is used to determine the information need, to retrieve relevant documents and to give methodical guidance for the legal task itself. To account for the design of a task-based legal information retrieval system, a substantial part of this paper is devoted to analysis and representation of legal tasks.  相似文献

The accurate and precise measurement of process stream temperatures during injection molding can be difficult, since the cyclic operation results in spatial and temporal variations of the stream temperature. This paper examines the application of a spectral infrared (IR) pyrometer to monitor the cooling of a polymer melt within the mold cavity during a typical injection molding cycle. An outline for interpreting the radiation signal collected with the IR pyrometer is presented. The discussion includes theoretical aspects as well as experimental results. The theoretical approach accounts for the polymer transparency (attenuation behavior) at the spectral wavelength of the pyrometer and also for the temperature gradient within the polymer, thereby establishing the concept of a critical depth for a given pyrometer/polymer combination. The final analysis reveals good agreement between the predicted and measured results for the transient cooling conditions of the polymer within the mold cavity. Depending on the degrees of polymer transparency used in the theoretical prediction, the deviation between the measured and predicted transient bulk temperatures after mold filling (during the mold cooling stage) varies from ±2°K to ±9°K.  相似文献

In order to reduce the cost of the manufacturing of Solid Oxide Fuel Cells (SOFC), and to enable metal supported cell fabrication, a new fabrication method called Reactive Spray Deposition Technology (RSDT) for direct deposition of the material onto ceramic or metal support for low temperature SOFC is currently being developed. The present work describes the effect on the performance of a SOFC when a Gd0.2Ce0.8O1.9 (GDC) layer has been introduced as diffusion barrier layer between the yttria stabilized zirconia (YSZ) electrolyte and the La0.6Sr0.4CoO3−δ (LSC) cathode. The dense, thin and fully crystalline GDC films were directly applied by RSDT, without any post-deposition heating or sintering step. The quality of the film and performance of the cell prepared by RSDT was compared to a GDC blocking layer deposited by screen printing (SP) and then sintered. The observed ohmic resistance of the ASC with a GDC layer deposited by RSDT is 0.24 Ω cm2, which is close to the expected theoretical value of 0.17 Ω cm2 for a 5-μm thick 8 mol% yttria YSZ (8YSZ) electrolyte at 873 K.  相似文献

In this paper we present a new robust approach for 3D face registration to an intrinsic coordinate system of the face. The intrinsic coordinate system is defined by the vertical symmetry plane through the nose, the tip of the nose and the slope of the bridge of the nose. In addition, we propose a 3D face classifier based on the fusion of many dependent region classifiers for overlapping face regions. The region classifiers use PCA-LDA for feature extraction and the likelihood ratio as a matching score. Fusion is realised using straightforward majority voting for the identification scenario. For verification, a voting approach is used as well and the decision is defined by comparing the number of votes to a threshold. Using the proposed registration method combined with a classifier consisting of 60 fused region classifiers we obtain a 99.0% identification rate on the all vs first identification test of the FRGC v2 data. A verification rate of 94.6% at FAR=0.1% was obtained for the all vs all verification test on the FRGC v2 data using fusion of 120 region classifiers. The first is the highest reported performance and the second is in the top-5 of best performing systems on these tests. In addition, our approach is much faster than other methods, taking only 2.5 seconds per image for registration and less than 0.1 ms per comparison. Because we apply feature extraction using PCA and LDA, the resulting template size is also very small: 6 kB for 60 region classifiers.  相似文献

This paper addresses the economic feasibility of measures to reduce the material damage of transport companies. Results are presented of a series of interviews among transport companies as well as from a postal questionnaire survey. Next, calculations are presented for three types of companies: a small family company, a large family company and a large formalised company. From the viewpoint of costs and benefits, damage prevention measures appear to be particularly interesting to larger companies. Small companies, being the largest group, tend to have an informal culture in which measures are less effective. Especially those measures for which no large investments are needed, which influence the behaviour of drivers and need not to be contracted out, are perceived as attractive by the transport companies.  相似文献

Early elimination of natural organic matter (NOM) by ion exchange (IEX) in water treatment is expected to improve subsequent water treatment processes and the final drinking water quality. Nine anionic exchange resins were investigated to remove NOM and specific NOM fractions determined by liquid chromatography in combination with organic carbon detection (LC-OCD) and fluorescence excitation-emission matrices (EEM). Breakthrough of NOM was predicted by model calculations using Freundlich isotherms and IEX rate experiments. The time to breakthrough varied from 4 to 38 days. Removal of specific NOM fractions proved to vary considerably for the different types of IEX resins, ranging from 1% to almost 60%. The removal of NOM fractions, specifically humic substances, increased with an increase in water content of the investigated IEX resins and with a decrease in resin size. The best-performing IEX resins consisted of the smallest resins and/or those with the highest water content. The worst-performing IEX resins reflected the highest exchanging capacities and the lowest water contents.  相似文献

Iron oxidation under neutral conditions (pH 6.5-8) may be a homo- or heterogeneous chemically- or a biologically-mediated process. The chemical oxidation is supposed to outpace the biological process under slightly alkaline conditions (pH 7-8). The iron oxidation kinetics and growth of Gallionella spp. - obligatory chemolithotrophic iron oxidizers - were assessed in natural, organic carbon-containing water, in continuous lab-scale reactors and full-scale groundwater trickling filters in the Netherlands. From Gallionella cell numbers determined by qPCR, balances were made for all systems. The homogeneous chemical iron oxidation occurred in accordance with the literature, but was retarded by a low water temperature (13 °C). The contribution of the heterogeneous chemical oxidation was, despite the presence of freshly formed iron oxyhydroxides, much lower than in previous studies in ultrapure water. This could be caused by the adsorption of natural organic matter (NOM) on the iron oxide surfaces. In the oxygen-saturated natural water with a pH ranging from 6.5 to 7.7, Gallionella spp. grew uninhibited and biological iron oxidation was an important, and probably the dominant, process. Gallionella growth was not even inhibited in a full-scale filter after plate aeration. From this we conclude that Gallionella spp. can grow under neutral pH and fully aerated conditions when the chemical iron oxidation is retarded by low water temperature and inhibition of the autocatalytic iron oxidation.  相似文献

A fractionating reactor for equilibrium‐limited reactions is studied theoretically. Reactant A is fed in the center of the countercurrent fractionating system. Product P is effectively transported with the auxiliary phase, while product Q is effectively transported with the main phase, in which the reaction takes place. Model calculations were based on partition and reaction equilibrium at all stages. These show that if the initial reactant concentration and the flow rates are properly selected, the extent of conversion will significantly exceed the corresponding batch conversion. To approach complete conversion in the fractionating reactor, and to recover both products in a pure form, net transport of reactant in either of the countercurrent directions should be prevented. However, irrespective of the number of equilibrium stages, this situation cannot be fully reached when the reactant feed stream is too large (compared with the main and auxiliary streams).
