Increasing conjugated linoleic acid (CLA) content of milk fat from lactating dairy cattle has become a research interest due to the possible health benefits afforded humans consuming CLA. Dietary supplementation of CLA to lactating dairy cows is one potential method by which CLA content of milk and dairy products may be enhanced. Feeding CLA in calcium salt form could potentially deliver CLA to the lower digestive tract through prevention of biohydrogenation by rumen microbes. Milk fat depression (MFD) occurs when cows receive CLA-60, a commercially available CLA source containing numerous CLA isomers, abomasally. Our objectives were to determine the quantity of CLA as calcium salts required to elicit maximal MFD and to evaluate the effects of CLA supplementation on fatty acid composition of milk fat. Five Holstein cows at approximately 93 DIM were utilized in a 5 x 5 balanced Latin square crossover design. Periods were 14-d in length with a 5-d treatment phase and 9-d rest phase. Treatments were 5-d supplementation of 0, 12.5, 25, 50, and 100 g of CLA-60 in calcium salt form. Milk samples were collected on d 5 of CLA supplementation and analyzed for composition and fatty acid profile. Regression analysis of milk fat data suggested that MFD was not maximized over the dose levels investigated, despite delivery of 34.5 g of trans-10, cis-12 CLA in the 100-g dose of CLA. Supplementation with 50 and 100 g of CLA per day resulted in a reduction of milk fat percent of 29 and 34%, respectively. Trend analysis indicated a linear decrease in the milk fat content of caprylic, capric, and lauric acids as the dose of CLA increased. Milk fat content of cis-9, trans-11, and trans-10, cis-12 CLA increased at an increasing rate as dose increased.  相似文献

Pichia sorbitophila grows rapidly in the presence of very high NaCl concentrations. Under these conditions, even when the K(+) concentration is low, P. sorbitophila cells can maintain low Na(+) and high K(+) contents. This remarkable capacity of P. sorbitophila fails when the external pH is not acidic. This indicates that Na(+) efflux is mediated by an electroneutral Na(+)/H(+) antiporter. We have cloned and sequenced two genes designated as PsNHA1 and PsNHA2, which probably encode two antiporters of this type. The genes present high similarity with the corresponding genes from other yeasts. The heterologous expression of PsNHA1 or PsNHA2 in a Saccharomyces cerevisiae mutant lacking the Na(+) efflux systems and sensitive to high concentrations of Na(+) and K(+) rescued the tolerance and the ability to extrude both cations. The genus Fusarium comprises a diverse group of fungi including several species that produce mycotoxins in food commodities. In this study, a multiplex polymerase chain reaction (PCR) assay was developed for the group-specific detection of fumonisin-producing and trichothecene-producing species of Fusarium. Primers for genus-level recognition of Fusarium spp. were designed from the internal transcribed spacer regions (ITS1 and ITS2) of rDNA. Primers for group-specific detection were designed from the TRI6 gene involved in trichothecene biosynthesis and the FUM5 gene involved in fumonisin biosynthesis. Primer specificity was determined by testing for cross-reactivity against purified genomic DNA from 43 fungal species representing 14 genera, including 9 Aspergillus spp., 9 Fusarium spp., and 10 Penicillium spp. With purified genomic DNA as a template, genus-specific recognition was observed at 10 pg per reaction; group-specific recognition occurred at 100 pg of template per reaction for the trichothecene producer Fusarium graminearum and at 1 ng of template per reaction for the fumonisin producer Fusarium verticillioides. For the application of the PCR assay, a protocol was developed to isolate fungal DNA from cornmeal. The detection of F. graminearum and its differentiation from F. verticillioides were accomplished prior to visible fungal growth at <10(5) CFU/g of cornmeal. This level of detection is comparable to those of other methods such as enzyme-linked immunosorbent assay, and the assay described here can be used in the food industry's effort to monitor quality and safety.  相似文献

Biotrickling filters for air pollution control are expected to encounter fluctuating conditions or periods without pollutant supply. In the present study, we investigated the effect of pollutant starvation in bench-scale biotrickling filters treating toluene. The experimental protocol consisted of starving biotrickling filters under various conditions: with or without airflow, with or without liquid recycle, and with or without an alternate carbon source (glucose) supply. The duration of the period without toluene was varied from 2 to 9 days, during time which the biotrickling filters were monitored for biomass content, endogenous and toluene-induced oxygen uptake rates during starvation, and toluene overall elimination capacity after restart. During starvation, all reactors lost their ability to degrade toluene within 5 days, regardless of the mode of starvation. The biomass content significantly decreased during starvation, in particular in those reactors where the recycle liquid was maintained, but this decrease was not critical for future re-acclimation. Glucose addition to starved biotrickling filters had several detrimental effects. It resulted in a faster decrease of the biomass content and slowed the reacclimation phase. Overall, the results show that the reacclimation of toluene-degrading biotrickling filters after periods of nonuse is short (10-24 h to re-establish full performance), and they suggest that, in the case of toluene-degrading biotrickling filters, re-acclimation time is largely governed by the induction of key pollutant-degrading enzymes.  相似文献

Mercury contamination in the Gold-Cyanide Process (GCP) is a serious health and environmental problem. Following the heap leaching of gold and silver ores with NaCN solutions, portions of the mercury-cyano complexes often adhere to the activated carbon (AC) used to extract the gold. During the electrowinning and retorting steps, mercury can be (and often is) emitted to the air as a vapor. This poses a severe health hazard to plant workers and the local environment. Additional concerns relate to the safety of workers when handling the mercury-laden AC. Currently, mercury treatment from the heap leach solution is nonexistent. This is due to the fact that chelating ligands which can effectively work under the adverse pH conditions (as present in the heap leachate solutions) do not exist. In an effort to economically and effectively treat the leachate solution prior to passing over the AC, a dipotassium salt of 1,3-benzenediamidoethanethiol (BDET2-) has been developed to irreversibly bind and precipitate the mercury. The ligand has proven to be highly effective by selectively reducing mercury levels from average initial concentrations of 34.5 ppm (parts per million) to 0.014 ppm within 10 min and to 0.008 ppm within 15 min. X-ray powder diffraction (XRD), proton nuclear magnetic resonance (1H NMR), Raman, and infrared (IR) spectroscopy demonstrate the formation of a mercury-ligand compound, which remains insoluble over pH ranges of 0.0-14.0. Leachate samples from an active gold mine in Peru have been analyzed using cold vapor atomic fluorescence (CVAF) and inductively coupled plasma optical emission spectroscopy (ICP-OES) for metal concentrations before and after treatment with the BDET2- ligand.  相似文献

A current industry goal is to produce a 75 to 80% fat-reduced Cheddar cheese that is tasty and appealing to consumers. Despite previous studies on reduced-fat cheese, information is critically lacking in understanding the flavor and flavor chemistry of reduced-fat and nonfat Cheddar cheeses and how it differs from its full-fat counterpart. The objective of this study was to document and compare flavor development in cheeses with different fat contents so as to quantitatively characterize how flavor and flavor development in Cheddar cheese are altered with fat reduction. Cheddar cheeses with 50% reduced-fat cheese (RFC) and low-fat cheese containing 6% fat (LFC) along with 2 full-fat cheeses (FFC) were manufactured in duplicate. Cheeses were ripened at 8°C and samples were taken following 2 wk and 3, 6, and 9 mo for sensory and instrumental volatile analyses. A trained sensory panel (n = 10 panelists) documented flavor attributes of cheeses. Volatile compounds were extracted by solid-phase microextraction or solvent-assisted flavor evaporation followed by separation and identification using gas chromatography-mass spectrometry and gas chromatography-olfactometry. Selected compounds were quantified using external standard curves. Sensory properties of cheeses were distinct initially but more differences were documented as cheeses aged. By 9 mo, LFC and RFC displayed distinct burnt/rosy flavors that were not present in FFC. Sulfur flavor was also lower in LFC compared with other cheeses. Forty aroma-active compounds were characterized in the cheeses by headspace or solvent extraction followed by gas chromatography-olfactometry. Compounds were largely not distinct between the cheeses at each time point, but concentration differences were evident. Higher concentrations of furanones (furaneol, homofuraneol, sotolon), phenylethanal, 1-octen-3-one, and free fatty acids, and lower concentrations of lactones were present in LFC compared with FFC after 9 mo of ripening. These results confirm that flavor differences documented between full-fat and reduced-fat cheeses are not due solely to differences in matrix and flavor release but also to distinct differences in ripening biochemistry, which leads to an imbalance of many flavor-contributing compounds.  相似文献

Our objectives were to determine if dietary cation-anion difference (DCAD) and source of anions influence periparturient feed intake and milk production of dairy cattle during the transition period. Diets differed in DCAD (cationic or anionic) and anionic supplement. The 4 diets used prepartum were (1) control [DCAD +20 mEq/100 g of dry matter (DM)], (2) Bio-Chlor (DCAD −12 mEq/100 g of DM; Church & Dwight Co. Inc., Princeton, NJ), (3) Fermenten (DCAD −10 mEq/100 g of DM; Church & Dwight Co. Inc.), and (4) salts (DCAD −10 mEq/100 g of DM). Urine pH was lower for cows that consumed an anionic diet prepartum compared with control. Prepartum diet had no effect on prepartum dry matter intake (DMI) of multiparous or primiparous cows. Postpartum DMI and milk yield for multiparous cows fed anionic diets prepartum were greater compared with those fed the control diet. Postpartum DMI and milk yield of primiparous cows were similar for prepartum diets. Feeding prepartum anionic diets did not affect plasma Ca at or near calving. However, cows fed anionic diets began their decline in plasma Ca later than control cows. Postpartum β-hydroxybutyrate and nonesterified fatty acids were lower for primiparous cows fed prepartum anionic diets compared with those fed the control diet. Prepartum and postpartum plasma glucose concentrations were not affected by prepartum diet for all cows. Liver triglyceride differed for parity by day. Parities were similar at 21 d prepartum, but at 0 d and 21 d postpartum, levels were greater for multiparous cows. Results indicate that decreasing the DCAD of the diet during the prepartum period can increase postpartum DMI and milk production of multiparous cows without negatively affecting performance of primiparous cows.  相似文献

Bovine lameness results in pain and suffering in cattle and economic loss for producers. A system for automatically detecting lame cows was developed recently that measures vertical force components attributable to individual limbs. These measurements can be used to calculate a number of limb movement variables. The objective of this investigation was to explore whether gait scores, lesion scores, or combined gait and lesion scores were more effectively captured by a set of 5 limb movement variables. A set of 700 hind limb examinations was used to create gait-based, lesion-based, and combined (gait- and lesion-based) models. Logistic regression models were constructed using 1, 2, or 3 d of measurements. Resulting models were tested on cows not used in modeling. The accuracy of lesion-score models was superior to that of gait-score models; lesion-based models generated greater values of areas under the receiving operating characteristic curves (range 0.75 to 0.84) and lower mean-squared errors (0.13 to 0.16) compared with corresponding values for the gait-based models (0.63 to 0.73 and 0.26 to 0.31 for receiving operating characteristic and mean-squared errors, respectively). These results indicate that further model development and investigation could generate automated and objective methods of lameness detection in dairy cattle.  相似文献

Biogeochemically modified pore waters from subterranean estuaries, defined as the mixing zone between freshwater and saltwater in a coastal aquifer, are transported to coastal waters through submarine groundwater discharge (SGD). SGD has been shown to impact coastal and perhaps global trace metal budgets. The focus of this study was to investigate the biogeochemical processes that control arsenic cycling in subterranean estuaries. Total dissolved As, as well as a suite of other trace metals and nutrients, were measured in a series of wells and sediment cores at the head of Waquoit Bay, MA. Dissolved As ranged from below detection to 9.5 microg/kg, and was associated with plumes of dissolved Fe, Mn, and P in the groundwater. Sedimentary As, ranging from 360 to 7500 microg/kg, was highly correlated with sedimentary Fe, Mn, and P. In addition, amorphous Fe (hydr)oxides were more efficient scavengers of dissolved As than the more crystalline forms of solid-phase Fe. Given that dissolved As in the surface bay water was lower than within the subterranean estuary, our results indicate that the distribution and type of Fe and Mn (hydr)oxides in coastal aquifers exert a major influence on the biogeochemical cycling of As in subterranean estuaries and, ultimately, the fate of groundwater-derived As in marine systems influenced by SGD.  相似文献
